Article Synopsis
  • Tumor-specific CD8 T cells in lymph nodes associated with tumors (TdLNs) act as a source of exhausted T cell types within the tumor microenvironment, including progenitor exhausted T cells and new memory T cell subsets.
  • These two subsets play a key role in enhancing the effects of PD-1/PD-L1 immune checkpoint blockade (ICB) by working together over time and space.
  • While ICB treatment boosts the growth and differentiation of these T cell subsets, it does not prevent them from eventually becoming terminally exhausted, which may contribute to the frequent relapses seen in patients after initial treatment.

Peking University People's Hospital, Peking University Institute of Hematology, National Clinical Research Center for Hematologic Disease, Beijing Key Laboratory of Hematopoietic Stem Cell Transplantation, Beijing, China.

Article Synopsis
  • Chronic myelomonocytic leukemia (CMML) is a type of blood cancer where allogeneic hematopoietic stem cell transplantation (allo-HSCT) is currently the only curative treatment, although not suitable for all patients, and relapse is a common issue.
  • A nationwide study analyzed data from 238 CMML patients who underwent allo-HSCT across 27 medical centers and 307 patients from a research database to establish a risk scoring system to predict early relapse based on certain prognostic factors.
  • Four key factors were identified that significantly increased the risk of relapse: bone marrow blasts over 10%, age greater than 60 years, low hemoglobin levels, and specific gene mutations, leading to a scoring system that categor

Background: Cell death plays an essential role in carcinogenesis, but its function in the recurrence and postoperative prognosis of head and neck cancer (HNC), which ranks as the 7 most common malignancy globally, remains unclear.

Methods: Data from five main subtypes of HNC related single-cell RNA sequencing (scRNA-seq) were recruited to establish a single-cell atlas, and the distribution of cell death models (CDMs) across different tissues as well as cell subtypes were analyzed. Bulk RNA-seq from the Cancer Genome Atlas Program (TCGA) dataset was subjected to a machine learning-based integrative procedure for constructing a consensus cell death-related signature risk score (CDRscore) model and validated by external data.

Communication of gut hormones with the central nervous system is important to regulate systemic glucose homeostasis, but the precise underlying mechanism involved remain little understood. Nesfatin-1, encoded by nucleobindin-2 (NUCB2), a potent anorexigenic peptide hormone, was found to be released from the gastrointestinal tract, but its specific function in this context remains unclear. Herein, we found that gut nesfatin-1 can sense nutrients such as glucose and lipids and subsequently decreases hepatic glucose production.
