Cancer cell membrane-camouflaged curcumin nanoparticles trigger ferroptosis for accurate gastric cancer therapy.

Eur J Pharm Biopharm

November 2024

Department of Traditional Chinese Medicine, Shengjing Hospital of China Medical University, Shenyang 110004, PR China. Electronic address:

Curcumin (CUR) is a hydrophobic polyphenol with considerable antitumor efficiency, but its clinical application is limited because of its poor solubility and low stability in aqueous solution and lack of targeting in vivo. Herein, we fabricated a tumor-targeting drug delivery system by loading CUR and cloaking homologous cancer cell membrane (CM) onto mesoporous silica NPs (MSN-CUR@CM). Characterization analysis showed that MSN-CUR@CM with a size of approximately 70 nm showed high water solubility and biocompatibility.

Dronedarone hydrochloride (DH) induces pancreatic cancer cell death by triggering mtDNA-mediated pyroptosis.

Cell Death Dis

October 2024

Department of Gastroenterology, the First Affiliated Hospital (Southwest Hospital), Third Military Medical University (Army Medical University), Chongqing, 400038, China.

Pancreatic cancer is one of the leading causes of cancer-associated mortality, with a poor treatment approach. Previous study has shown that inducing pyroptosis in pancreatic ductal adenocarcinoma (PDAC) slows the growth of PDACs, implying that pyroptosis inducers are potentially effective for PDAC therapy. Here, we found that Dronedarone hydrochloride (DH), an antiarrhythmic drug, induces pyroptosis in pancreatic cancer cells and inhibits PDAC development in mice.

Background: Stomach adenocarcinoma (STAD) is a major contributor to cancer-related mortality worldwide. Alterations in amino acid metabolism, which is integral to protein synthesis, have been observed across various tumor types. However, the prognostic significance of amino acid metabolism-related genes in STAD remains underexplored.

Article Synopsis
  • Older patients who are frail before surgery may have a higher chance of experiencing confusion after brain surgery (called postoperative delirium or POD).
  • In a study of 300 patients who had this surgery, about 28% were frail and around 23% experienced POD afterwards.
  • Researchers found that being frail before surgery, along with a few other health factors, increased the risk of developing POD, which typically leads to a longer hospital stay and higher medical costs.

The estrogen response in fibroblasts promotes ovarian metastases of gastric cancer.

Nat Commun

September 2024

Department of Gastric Surgery, Zhejiang Cancer Hospital, Institute of Basic Medicine and Cancer (IBMC), Chinese Academy of Sciences, Hangzhou, China.

Article Synopsis
  • Younger premenopausal women show a higher likelihood of developing ovarian metastases from gastric cancer compared to other cancers, but the exact biological processes are not fully understood.
  • Researchers used single-cell RNA sequencing on tumor samples from gastric cancer patients with ovarian metastases, discovering that ovarian fibroblasts express high levels of estrogen receptors and midkine, which facilitates communication with tumor cells through a specific signaling pathway.
  • The study suggests that estrogen influences ovarian fibroblasts to secrete midkine, which promotes gastric cancer cell migration and invasion, potentially leading to new treatment approaches for ovarian metastases in gastric cancer.

September 2024

Department of Plastic Surgery, State Key Laboratory of Trauma and Chemical Poisoning, Southwest Hospital, Army Medical University (Third Military Medical University), Chongqing, 400038, China.

Background: During wound healing, fibroblast to myofibroblast transition is required for wound contraction and remodeling. While hypoxia is an important biophysical factor in wound microenvironment, the exact regulatory mechanism underlying hypoxia and fibroblast-to-myofibroblast transition remains unclear. We previously found that tetraspanin CD9 plays an important role in oxygen sensing and wound healing.

Essential role of Dhx16-mediated ribosome assembly in maintenance of hematopoietic stem cells.

Leukemia

December 2024

Department of Radiological Medicine, School of Basic Medical Sciences, Chongqing Medical University, Chongqing, 400016, China.

Article Synopsis
  • Hematopoietic stem cells (HSCs) are crucial for producing mature blood cells, and their regulation relies on various factors, including alternative splicing, which increases protein diversity.
  • The study investigates the ATP-dependent RNA helicase DHX16, finding that its absence in mice leads to severe depletion of HSCs, bone marrow failure, and high mortality rates, along with disruptions in cell cycle and protein synthesis.
  • The research identifies that loss of DHX16 affects ribosome assembly and activates the p53 pathway due to altered Emg1 mRNA, suggesting that targeting Emg1 could provide therapeutic options for ribosomopathies in hematopoietic diseases.

Article Synopsis
  • Intestinal stem cells (ISCs) help keep our intestines healthy and can heal after injuries like burns, and glutamine (Gln) is important for their growth and recovery.
  • Researchers studied mice with burn injuries to see how Gln affects ISCs, specifically checking how a protein called TIGAR plays a role in this process.
  • The results showed that Gln helped boost TIGAR levels, which in turn helped ISCs grow and protect themselves from damage, showing that Gln is really important for intestinal health after injuries.
