Article Synopsis
  • The study investigates how abnormal DNA methylation patterns may influence the severity of adolescent idiopathic scoliosis (AIS), focusing on twins to control for genetic factors.
  • Researchers analyzed blood samples from 7 pairs of monozygotic twins, identifying several differentially methylated genes linked to curve severity, with a significant majority being hypermethylated in individuals with more severe scoliosis.
  • Functional analysis revealed that these differentially methylated genes are involved in critical processes like skeletal development and signaling pathways, providing insights into the biological mechanisms behind AIS progression.

AS160 is a lipid-responsive regulator of cardiac Ca homeostasis by controlling lysophosphatidylinositol metabolism and signaling.

Nat Commun

November 2024

State Key Laboratory of Pharmaceutical Biotechnology, Department of Cardiology, Nanjing Drum Tower Hospital, The Affiliated Hospital of Nanjing University Medical School, Model Animal Research Center, School of Medicine, Nanjing University, Nanjing, China.

The obese heart undergoes metabolic remodeling and exhibits impaired calcium (Ca) homeostasis, which are two critical assaults leading to cardiac dysfunction. The molecular mechanisms underlying these alterations in obese heart are not well understood. Here, we show that the Rab-GTPase activating protein AS160 is a lipid-responsive regulator of Ca homeostasis through governing lysophosphatidylinositol metabolism and signaling.

Niels Lassen's seminal 1959 cerebral autoregulation plot, a cornerstone in understanding the relationship between mean arterial pressure (MAP) and cerebral blood flow (CBF), was based on preexisting literature. However, this work has faced criticism for selective data presentation, leading to inaccurate interpretation. This review revisits and validates Lassen's original plot using contemporary data published since 2000.

FoxO1 Deficiency in Monocytic Myeloid-Derived Suppressor Cells Exacerbates B Cell Dysfunction in Systemic Lupus Erythematosus.

Arthritis Rheumatol

November 2024

Nanjing University, The State Key Laboratory of Pharmaceutical Biotechnology and Jiangsu Key Laboratory of Molecular Medicine, Nanjing, People's Republic of China.

Objective: Myeloid-derived suppressor cells (MDSCs) contribute to the pathogenesis of systemic lupus erythematosus (SLE), in part due to promoting the survival of plasma cells. FoxO1 expression in monocytic MDSCs (M-MDSCs) exhibits a negative correlation with the SLE Disease Activity Index score. This study aimed to investigate the hypothesis that M-MDSC-specific FoxO1 deficiency enhances aberrant B cell function in aggressive SLE.

The relevant mechanism of tumor-associated macrophages (TAMs) in the treatment of colorectal cancer patients with immune checkpoint inhibitors (ICIs) is discussed, and the application prospects of TAMs in reversing the treatment tolerance of ICIs are discussed to provide a reference for related studies. As a class of drugs widely used in clinical tumor immunotherapy, ICIs can act on regulatory molecules on cells that play an inhibitory role-immune checkpoints-and kill tumors in the form of an immune response by activating a variety of immune cells in the immune system. The sensitivity of patients with different types of colorectal cancer to ICI treatment varies greatly.
