Objective: Hypertension significantly contributes to morbidity and mortality. Nuclear receptor subfamily 4 group a member 1 (Nur77) participates in regulating oxidative stress, but the mechanism in hypertension remains unclear. This study aimed to explore the function of Nur77 in oxidative stress induced by Angiotensin II (Ang II) in vascular smooth muscle cells (VSMCs) in hypertension.

Sclerostin as a new target of diabetes-induced osteoporosis.

Front Endocrinol (Lausanne)

December 2024

Department of Endocrinology and Metabolism, Want Want Hospital, Changsha, Hunan, China.

Sclerostin, a protein synthesized by bone cells, is a product of the gene. Sclerostin is a potent soluble inhibitor of the WNT signaling pathway, and is known to inhibit bone formation by inhibiting osteocyte differentiation and function. Currently, sclerostin has been the subject of numerous animal experiments and clinical investigations.

Article Synopsis
  • A study evaluated a new long-lasting pain relief medication called QP001, compared to a placebo, to determine its effectiveness and safety for patients after abdominal surgery.
  • Conducted across 23 centers, 255 patients were administered either QP001 or placebo, and the results showed QP001 significantly reduced pain intensity for up to 24 hours post-surgery.
  • The trial indicated that QP001 also reduced the need for morphine and increased patient satisfaction, with no significant difference in adverse events compared to placebo.

Background: Modern pharmacological studies have demonstrated that although triptolide (TP) is effective against hepatocellular carcinoma, it has poor water solubility and more toxic side effects. In this study, we used triptolide (TP), a bioactive constituent in Hook F, as a model drug to develop a novel nano-liposome drug delivery system for the treatment of liver tumours.

Methods: We constructed a functionally-modified triptolide liposome (FA+TPP-TP-Lips) using the film-dispersion method and investigated its physicochemical properties, mitochondrial targeting of hepatic tumour cells, in vitro and in vivo anti-hepatic tumour activity and its mechanism.

Beijing Key 1aboratory of Work Safety Intelligent Monitoring, School of Electronic Engineering, Beijing University of Posts and Telecommunications, Beijing 100876, China.

The pH and O are the two important parameters to evaluate environmental conditions, healthcare, and industrial processes. Herein, we developed a ratiometric fluorescent capillary-based dual-functional sensor for simultaneous monitoring of pH and O fluctuations. The fluorescent sensor consists of a pH sensing unit and an O sensing unit on both ends of the capillary inner wall.

  • Recaticimab is a new monoclonal antibody that significantly lowers LDL cholesterol levels with less frequent dosing compared to current treatments, potentially every 12 weeks.
  • The REMAIN-2 trial examined the efficacy and safety of recaticimab as an add-on to statin therapy for patients with nonfamilial hypercholesterolemia over 48 weeks.
  • Results showed that patients receiving recaticimab experienced a significant reduction in LDL cholesterol levels compared to those on a placebo, particularly notable at the 24-week mark.

Article Synopsis
  • Polymyxins, such as colistin and polymyxin B, are crucial for treating Gram-negative bacterial infections, but their use is limited due to concerns about neurotoxicity.
  • The study aimed to compare the neurotoxic effects of colistin and polymyxin B by conducting both in vitro (cell culture) and in vivo (rat model) experiments, focusing on cell viability, apoptosis, oxidative stress, and brain injury.
  • Findings revealed that colistin caused less neurotoxicity than polymyxin B, showing better cell viability and lower levels of oxidative stress, which may support more rational clinical use of these antibiotics.

Article Synopsis
  • This study investigated the effectiveness and safety of a new pain medication, QP001, for managing postoperative pain in patients after orthopaedic surgery in China.
  • In a clinical trial with 198 participants, those taking QP001 consumed significantly less morphine for pain relief compared to those receiving a placebo, both at 24 hours and 48 hours post-surgery.
  • QP001 not only reduced the need for opioids but also had fewer adverse drug reactions, indicating it could be a promising alternative for pain management after surgery.

  • Monoclonal antibodies like recaticimab target PCSK9 to effectively lower LDL cholesterol levels, offering an alternative to more frequent treatments due to its long-acting nature.
  • The study evaluated recaticimab's effectiveness and safety in patients with nonfamilial hypercholesterolemia and low-to-moderate atherosclerotic cardiovascular disease risk, comparing various dosing strategies.
  • Results showed significant LDL-C reductions (up to 52.8%) compared to placebo, with safety profiles similar to that of placebo, suggesting recaticimab may provide a flexible treatment option for patients.

The role of forkhead box M1-methionine adenosyltransferase 2 A/2B axis in liver inflammation and fibrosis.

Nat Commun

September 2024

Karsh Division of Gastroenterology and Hepatology, Department of Medicine, Cedars-Sinai Medical Center, LA, Los Angeles, CA, 90048, USA.

Methionine adenosyltransferase 2 A (MAT2A) and MAT2B are essential for hepatic stellate cells (HSCs) activation. Forkhead box M1 (FOXM1) transgenic mice develop liver inflammation and fibrosis. Here we examine if they crosstalk in male mice.

Background: Observational researches have suggested a connection between iron deficiency anemia (IDA) and an increased likelihood of ischemic stroke (IS), yet establishing causality is challenging owing to the inherent limitations of such studies, including their vulnerability to confounding factors and the potential for reverse causation. This study employs a bidirectional two-sample Mendelian randomization (MR) approach to assess the causal linkage between IDA and IS and its subtypes.

Methods: Identifiable single nucleotide polymorphisms (SNPs) with significant links to either IDA or IS and its subtypes were employed as instrumental variables (IVs).

Background: Observational research has highlighted a potential relationship between rheumatoid arthritis (RA) and neurodegenerative diseases (NDs). However, the confirmation of a causal connection is impeded by the inherent limitations of such studies, including vulnerability to confounding factors and the possibility of reverse causality. This study employs a two-sample Mendelian randomization (MR) approach to assess the causal impact of RA on three NDs, including Alzheimer's disease (AD), Parkinson's disease (PD), and amyotrophic lateral sclerosis (ALS).

EBioMedicine

September 2024

MOE Key Laboratory of Pediatric Rare Diseases, Hengyang Medical School, University of South China, Hengyang, China; Center for Medical Genetics and Hunan Key Laboratory of Medical Genetics, School of Life Sciences, Central South University, Changsha, China. Electronic address:

Background: Medications prescribed for chronic diseases can lead to short-term neuropsychiatric symptoms, but their long-term effects on brain structures and psychiatric conditions remain unclear.

Methods: We comprehensively analyzed the FDA Adverse Event Reporting System database and conducted drug target Mendelian Randomization (MR) studies on six categories of common drugs, 477 brain imaging-derived phenotypes (IDPs) and eight psychiatric disorders. Genetic instruments were extracted from expression quantitative trait loci (eQTLs) in blood, brain, and other target tissues, protein quantitative trait loci (pQTLs) in blood, and genome-wide association studies (GWAS) of hemoglobin and cholesterol.

Background: Breast cancer is a prevalent disease that has a dismal prognosis for patients and a bad outlook for treatments. Ubiquitination is a reversible biological process that regulates protein production and degradation, as well as plays a vital role in protein transport, localization, and biological activity.

Methods: We obtained the breast cancer patient sample data and used a machine learning technique to create a novel index called Deubiquitinating enzyme related index (DUBRI) by gathering genes associated to deubiquitinating enzymes.
