Background: Glucocorticoid (GC) overuse is the main cause of osteonecrosis of the femoral head (ONFH). The dysfunction of bone marrow mesenchymal stem cells (BMSCs) plays an important role in ONFH pathogenesis. Physiological concentrations of GCs can induce the osteogenic differentiation of BMSCs; however, intervention with high concentrations of GC may lead to changes in aging and autophagy in certain cell types.

Alzheimer's disease (AD) is a neurodegeneration driven by beta-amyloid (Aβ) deposits in the brain involving autophagy dysfunction. Ginsenoside Rg1, a pharmacologically active compound found in ginseng, has possible therapeutic effects for AD. This study discovered that FGR proto-oncogene (FGR) was a therapeutic target of Rg1 in AD and it was possibly involved in autophagy.

Background: Hidradenitis suppurativa (HS) is a chronic, inflammatory and common skin disease. Observation studies have reported the association between HS and cancers, however no studies reported whether a causal relationship exists between HS and cancers. This study aimed to explore the causal relationship between HS and differential subtypes of cancers by conducting a bidirectional Mendelian randomization (MR) analysis.

DNA double-strand breaks (DSBs) are the most severe form of DNA damage, primarily repaired by the non-homologous end joining (NHEJ) pathway. A critical step in this process is DNA synapsis, where the two broken ends are brought together to facilitate timely repair. Deficiencies in NHEJ synapsis can lead to improper DNA end configurations, potentially resulting in chromosomal translocations.

Hydrogen sulfide (HS) is an essential endogenous gasotransmitter that can regulate a wide range of physiological processes. However, overproduction of HS is toxic to humans and causes liver injury, cardiovascular diseases, central nervous system-related disease, diabetes, even cancer. Hence, designing efficient imaging probes for real-time monitoring of the alterations in endogenous HS is a viable tactic for accurate diagnosis of these diseases.

Metabolic dysregulation triggered by nutrient influx at reperfusion onset induces reactive oxygen species (ROS) burst and cellular injury, contributing to the detrimental effects observed in ischemia/reperfusion (I/R) injury. Thus, implementing controlled reperfusion emerges as a superior cardioprotective strategy to alleviate reperfusion injury. Kinesin KIF5B transports GLUT4- and CD36-containing vesicles to the plasma membrane, facilizing the import of glucose and fatty acids into cells, suggesting a role in controlled reperfusion.

Sepsis shock is caused by a systemic infection characterized by circulatory disorders and metabolic abnormalities. Microorganisms or their toxins enter the bloodstream, releasing inflammatory mediators and triggering systemic inflammatory reactions, leading to multiple organ dysfunction and even failure. To explore new treatment methods, we studied the improvement effect of sesamoside on the inflammatory response in septic shock.

Recently the importance of B cells has been highlighted for therapy of several autoimmune diseases including rheumatoid arthritis (RA). Still, the functional role of B cells and antibodies in the disease process are unclear. Using animal models, antibodies specifically binding cartilage are pathogenic, but it has also recently been shown that both B cells and antibodies could be protective.

Triggering receptor expressed on myeloid cells-2 (TREM2) mainly expressed on microglia in the brain, and its mutations can increase the risk of Alzheimer's disease (AD). Upregulation or activation of TREM2 has been found to ameliorate several pathological features of AD, such as the reduction of amyloid beta (Aβ) plaques and tau hyperphosphorylation. However, the effects of TREM2 on neurogenesis are little understood.

Inefficient and low-precision delivery of exogenous nucleic acids (ENA) severely limits gene therapy on ischemic stroke (IS). Two problems need to be urgently addressed to improve the efficacy of gene therapy; first, the blood brain barrier (BBB) should be open to promote the accumulation of ENA or genetic material carriers in the ischemic brain parenchyma, and second, the efficient delivery of ENA into the ischemic cells. Previous studies applied ultrasonic cavitation either for opening BBB or for inducing sonoporation to deliver genetic materials into cells.

Background: Mortality from familial hypercholesterolemia (FH) remains high due to late diagnosis, and the rate of timely diagnosis remains low (< 10% globally and < 1% in China). Early screening and treatment could significantly reduce mortality risk, especially among young adults. This study aims to evaluate the cost-effectiveness of universal genetic screening of young adults aged 18-40 years compared to universal cholesterol screening or current passive screening strategies (opportunistic cholesterol screening and genetic cascade testing) for FH in China.

Introduction: XPO1 plays a crucial role in the nuclear export machinery, making it an attractive target for inhibiting nuclear-cytoplasmic transport in melanoma, where its overexpression is linked to unfavorable prognosis. However, XPO1 monotherapy has not demonstrated sufficient efficacy to be considered a first-line treatment option for melanoma.
Objectives: This research aimed to delve into the resistance mechanism of XPO1-targeting therapy in melanoma and fabricate a proteinoid microsphere which could target XPO1 and β-catenin to maximize the effect of XPO1 inhibitors.

Arthroscopic tenodesis of the long head of the biceps tendon (LHBT) is a common procedure in a series of bicipital tendinopathies, including tendinitis, SLAP lesions, and tendon instability. Locating and exposing the LHBT during arthroscopy present a technical challenge in tenodesis. Blind exploration of the subacromial space may result in inadvertent tendon transection and iatrogenic damage to surrounding structures, leading to increased patient trauma, bleeding, and operation time.
