We report here a simple and effective method to assess the CAG repeat size of HD gene for gene diagnosis of Huntington disease. Genomic DNA sequences in polymorphic CAG repeat HD gene was amplified by PCR with TaKaRa LA Taq DNA polymerase and GC buffer. To distinguish normal alleles from HD alleles, DNA fragments of affected alleles were recovered as templates for secondary PCR.

Fifty-one children with aplastic anemia (AA) from 1993 to 2004 in the authors' institution were treated by 3 therapies: 11 patients in group 1 received the SSL-6 protocol; 16 patients in group 2 had CsA alone, where the dose of CsA began from 9-12 mg/kg in the initial 2 weeks and tapered off to 5 mg/kg later; 24 patients in group 3 were treated combining rabbit ATG (Pasteur, Merieux) 2.5 mg/kg for 5 days with CsA, which was the same dose as in group 2. The response was 27, 50, and 79%, respectively.

Comprehensive treatment of arteriovenous malformations in the oral and maxillofacial region.

J Oral Maxillofac Surg

October 2005

Department of Oral and Maxillofacial Surgery, the Second Affiliated Hospital of Sun Yat-sen University, 107 Yanjiang Road, Guangzhou 510-120, China.

Purpose: Arteriovenous malformations (AVMs) in the oral and maxillofacial region are rare but potentially life-threatening vascular lesions. We report our experience in treating these lesions in the oral and maxillofacial region.

Patients And Methods: Superselective intra-arterial embolization (SIAE), sclerotherapy, bone wax packing of bone cavity and curettage, radiotherapy, and surgical resection were used alone or in combination in 28 patients with AVMs in the oral and maxillofacial region.

Objective: The previous studies indicated that mesenchymal stem cells (MSCs) either from umbilical cord blood (UCB) or from bone marrow (BM) had the same biological characteristics and the function of secreting hematopoietic growth factors (HGFs). The present study aimed to understand the effects of human UCB MSCs on the expansion of CD(34)(+) cells from UCB.

Methods: 1.

Objective: To isolate, culture and purify mouse bone marrow mesenchymal stem cells (MSCs) and observe the main biological characteristics of MSCs cultured in conditions for spermatogonia in vitro.

Methods: The tibias and femurs were dissected from 5 - 6-week old mice and the marrow in the tibias and femurs was flushed out with medium. MSCs were isolated, cultured, purified in vitro by Percoll density gradient centrifugation combined with adherent method and identified by dynamic observation of stem cell characteristics by transmission electron microscope, HE staining, and immunohistochemical detection of cell markers.

Objective: To detect the role of Vascular endothelial growth factor C (VEGF-C) in the interaction of tongue squamous cell carcinoma (TSCC) with the peri-carcinoma lymphatics.

Methods: TSCC cells were implanted on the chorioallantoic membrane (CAM), in which, group A was transfected by plasmid pREP7 including VEGF-C, group B was transfected by plasmid pREP7, and group C was Tca8113. Lymphatic vessels were stained by 5'-Nase.

To investigate the distribution of HLA-E alleles and linkage between HLA-E and HLA-A or -B loci in Chinese Han in Guangdong area, HLA-E alleles were detected by using PCR-SSP in 150 unrelated healthy individuals from Guangzhou area; HLA-A, -B antigens typing in 106 individuals was carried out with NIH standard microlymphocytoxic method. Analysis of linkage was performed between HLA-E and HLA-A, -B. The results showed that three alleles of HLA-E could be detected in this population.

Objective: To investigate in vitro methods of inducing mouse embryonic stem cell(s) (ESC) into hepatocytes.

Methods: E14 mouse ESC were cultivated in suspension and plated to form aggregates, the embryoid bodies. They were allowed to outgrow on the plated culture with the stepwise addition of growth factors-- acidic fibroblast growth factor (aFGF), hepatocyte growth factor (HGF) and oncostatin M (OSM) into the culture medium.

Objective: To explore the change of T cell subsets in patients suffered from hepatocellular carcinoma (HCC) before and after hepatectomy, and study the value of Roferon-A (interferon alpha-2a) combined with hepatic artery chemoembolization (HACE) and portal vein chemotherapy (PVC) after radical resection of HCC for preventing recurrence.

Methods: On 75 HCC patients, PVC and HACE were respectively given at 2 weeks and 4 weeks after radical tumor resection. In 2nd week after surgery, 33 cases of them accepted Roferon-A treatment for 1 week.

Background & Objective: Overexpression of cyclin D1 was shown in many tumors. The excessive expression of cyclin D1 is an important cause of many tumors. But there are still some controversies of whether the overexpression of cyclin D1 exists in brain gliomas.

Background & Objective: Matrix metalloproteinases(MMPs) play critical roles in the process of tumorigenesis, tumor invasion and metastasis. Extracellular matrix metalloproteinase inducer (EMMPRIN) derived from cancer cell can induce stromal fibroblasts to synthesize MMPs, which advances the invasion and metastasis of cancer. This study was designed to investigate the roles of EMMPRIN in the tumorigenesis, invasion and metastasis of bladder carcinoma by detecting the expression of EMMPRIN in human normal and carcinomatous bladder tissues.

Background & Objective: Brain gliomas seldom undergo extracranial metastasis. Local recurrence is the main reason of tumor patient's death. Therefore, it is important to detect tumor biological features through determination of gene expression.

[Research on physiological function of CD47].

Zhongguo Shi Yan Xue Ye Xue Za Zhi

August 2003

Department of Hematology, The Second Affiliated Hospital of Sun Yat-sen University, Guangzhou 510120, China.

CD47, also known as integrin-associated protein (IAP), is an ubiquitously expressed 50 kD cell membrane glycoprotein. This article reviews that as an integrin-associated molecule, CD47 regulates a series of integrin-dependent cell functions; CD47 regulates cell adhesiveness, migration and activation and platelet aggregation through binding to thrombospondins; CD7 is the extracellular ligand for human signal-regulatory proteins, thus it causes negative regulatory effect; CD47 induces costimulatory signals on activation of T cell, T cell apoptosis and T cell anergy and enhances the efficiency of TCR signaling; CD47 is a member of Rh antigen complex. CD47 is involved in stroma-supported erythropoiesis.

Objective: To investigate whether genetic variation in cocaine and amphetamine-regulated transcript (CART) gene might contribute to the genesis of type 2 diabetes.

Methods: Screening for mutations in the entire coding region for the CART gene were performed with polymerase chain reaction-single strand conformation polymorphism (PCR-SSCP) in 180 normoglycemic control subjects and 221 patients with type 2 diabetes.

Results: (1) Adenine deletion was identified at position 1,457 nucleotide located at untranslation area of exon 3.

This study was undertaken to establish a murine model for unrelated allogeneic umbilical cord blood transplantation (UCBT). The characteristics and percentage of hematopoietic stem/progenitor cells between near-term fetal and neonatal murine peripheral blood (FNPB) and bone marrow (BM) were evaluated by flow cytometry and semisolid methylcellulose culture. BABL/c (H-2(d)) recipient mice conditioned with high dose CTX were transplanted with FNPB form C57BL/6 (H-2(b)) mice and the survival rate, hematopoietic and immunological reconstruction, graft versus host disease (GVHD) and engraftment level were observed.
