Department of Blood Transfusion, The general Hospital of PLA, Centre for Clinic Blood Transfusion Medicine of PLA, Beijing 100853, China.

In order to determine the tolerance ability of platelet to change of osmotic pressure in solution, the isotonic fresh platelets were exposed to a series of crystal salt solutions with osmotic pressure range from 47 to 611 mOsm for 15 minutes. Then the platelets were returned to isotonic condition and kept for 15 minutes. The expressions of phosphatidylserine and CD62p were assayed in platelets.

December 2001

Department of Hematology and Institute of Clinical Basic Medical Sciences, The General Hospital of PLA, Beijing 100853, China.

To simplify the traditional method for cryopreservation of peripheral blood stem cells(PBSCs) at -196 degrees C with rate-controlled freezing with 10% dimethyl sulfoxide(DMSO), the simplified method was carried out by freezing the cells to -80 degrees C in low-temperature freezer with the combination of 5% DMSO, 3% hydroxyethyl starch(HES) and 4% human serum albumin(HSA) as cryoprotectant. PBSCs were cryopreserved by different methods. Cell viability and recovery rate of mononuclear cells (MNC), CFU-GM and CD34(+) cells were compared.

The objective of the study is to explore the effect of Fas, FasL and Bcl-2 on the process of apoptosis induced by chemotherapeutic drugs through detecting the expression of Fas, FasL and Bcl-2 on murine lymphoma cell line RMA. Dexamethasone(DEX), etoposide (VP-16), arsenic trioxide As(2)O(3) and all trans-retinoic-acid (ATRA) were added to the RMA cells as well as to the cells preincubated with interleukin-2 (IL-2), interleukin-6 (IL-6) or granulocyte-macrophage colony-stimulating factor (GM-CSF), respectively. The effect on apoptosis was observed and the expression of Fas and FasL mRNA as well as the expression of Fas and Bcl-2 antigen were measured.

[CD62p expression in platelet during the preparation course of Cryopreservated platelet-rich plasma].

Zhongguo Shi Yan Xue Ye Xue Za Zhi

June 2002

Department of Blood Transfusion, The General Hospital of PLA, Centre for Clinical Blood Transfusion Medicine of PLA, Beijing 100853, China.

In order to explore the factors that affect CD62p expression in platelet during the whole course of cryopreservated platelets preparation, CD62p expression of platelet was evaluated by flow cytometry assay. The whole course of cryopreservated platelets preparation in order included whole blood collection, centrifugation for fresh platelet-rich plasma preparation, addition of dimethyl sulfoxide, and freeze in -80 degrees C refrigerator and thaw in 38 degrees C water bath. Result showed that the CD62p expressed slowly from whole blood collection to addition of dimethyl sulfoxide, but expressed abruptly after freeze and thaw and it occupied 82 per cent of the whole expression.

To explore the dynamic change of CD34(+) cell expressing adhesion molecules in bone marrow and peripheral blood during mobilization with combination of chemotherapy and G-CSF and its clinical significance, mononuclear cells of bone marrow and peripheral blood from malignant hematopathy cases before and after mobilization with G-CSF were labeled by CD45-CY-Chrome, PE conjugated anti-CD34, and FITC conjugated anti-CD44, anti-CD49d, anti-CD62L and anti-CXCR4. For three-color fluorescence analysis by flow cytometry was performed on a FACScalibur. Also the relationship between the number of subpopulations in different expressions of adhesion molecules infused and the time of recovery in different blood cells after transplantation was evaluated.

[Thermometry of intracellular ice crystal formation in cryopreserved platelets].

Zhongguo Shi Yan Xue Ye Xue Za Zhi

December 2002

Department of Blood Transfusion, The General Hospital of PLA, Beijing 100853, China.

The temperature of platelet intracellular ice crystal formation (IIF) is one of the most important physical parameters to instruct platelet cryopreservation. In this study, the range of temperatures for platelet IIF was measured by means of biological and physical methods. All platelet samples were graded cooling, and two samples of per 5 degrees C decrease were thawed by 2 different ways: 37 degrees C directly (T 37 degrees C) and 37 degrees C after keeping in liquid nitrogen (LN) for 2 hours.
