[Deepening the understanding of the diagnostic system and therapeutic strategy for burn sepsis].

Zhonghua Shao Shang Za Zhi

May 2021

Department of Burns and Plastic Surgery, the 924th Hospital of PLA, Guilin 541002, China.

Burn patients with large area of skin defect are prone to cause wound infection, severe fluid loss, and hypermetabolism, etc, which lead to sequential dysfunction of multiple systems and easily induce severe systemic infections and sepsis. At present, sepsis has been the leading causes of death in severe burn patients, and its early diagnosis and timely treatment are critical for successful treatment of patients. As burn sepsis has unique pathophysiological characteristics, the diagnostic criteria for general sepsis lack specificity for burn sepsis.

Anterior cruciate ligament (ACL) reconstruction was realized using a combination of bone mesenchymal stem cells (BMSCs) and silk-collagen scaffold, and an in vivo evaluation of this combination was performed. By combining type I collagen and degummed silk fibroin mesh, silk-collagen scaffolds were prepared to simulate ligament components. BMSCs isolated from bone marrow of rabbits were cultured for a homogenous population and seeded on the silk-collagen scaffold.

[Research advances on application of single-cell RNA sequencing in islet cell biology].

Zhonghua Shao Shang Za Zhi

December 2020

Department of Burns and Plastic Surgery, the Fourth Medical Center of PLA General Hospital, Beijing 100048, China.

After the first single-cell RNA sequencing of human islet in 2016, a large number of analysis of RNA sequencing at single-cell level on mouse and human islets emerged, which has brought new progress to the research of islet cell biology. It has been proved that single-cell RNA sequencing can characterize rare types of endocrine cells, find the cellular heterogeneity of typical endocrine cells and new cell subtypes, analyze the differences of endocrine cells among species, and describe different states of various types of islet cells in developmental processes and metabolic diseases more accurately. Although the detection efficiency of single-cell RNA sequencing for low abundance transcripts is relatively low at present, with the improvement of technology and the progress of analytical methods, this technology will undoubtedly become a powerful tool to explore the cellular heterogeneity, development, and biological characteristics of metabolic diseases.

Background: Circular RNAs (circRNAs) serve for a genre of considerable modulatory molecules that have been largely researched in human cancers. However, the contribution of circRNA cysteine-rich transmembrane bone morphogenetic protein regulator 1 (circCRIM1) to osteosarcoma (OS) is completely unclear.

Methods: All the RNA levels were examined via quantitative real-time polymerase chain reaction (qRT-PCR).

Gastric cancer (GC) remains poor prognosis and survival issues due to the resistance of chemotherapies, such as cisplatin. The long non-coding RNA small nucleolar RNA host gene 7 (lncRNA-SNHG7) is known as an oncogenic molecule in diverse cancers. Here, we demonstrate that SNHG7 was significantly upregulated in gastric cancer and positively correlated with cisplatin resistance of gastric cancer cells that SNHG7 was significantly upregulated in cisplatin resistant cells.

Human bone marrow‑derived mesenchymal stem cells (HBMSCs) have the potential of multidirectional differentiation and self‑renewal, which is important for the formation of human bone. It has been reported that long non‑coding RNAs (lncRNAs) serve important roles in HBMSC osteogenic differentiation. The current study aimed to investigate the roles of long intergenic non‑protein coding RNA 00707 (LINC00707) and microRNA (miR)‑103a‑3p in the osteogenic differentiation of HBMSCs.

Article Synopsis
  • LncRNAs play a crucial role in various cellular processes and are emerging as potential targets for cancer therapy, specifically focusing on colon cancer in this study.
  • CASC2 is found to be significantly reduced in colon cancer tissues, and its overexpression hinders the growth of HT-29 cancer cells by promoting apoptosis and autophagy through changes in specific protein levels.
  • CASC2 regulates TRIM16 via the microRNA-214 pathway, and both CASC2 overexpression and TRIM16 silencing inhibit cancer cell growth, suggesting that targeting CASC2 could be a promising strategy for colon cancer treatment.
