Background And Purpose: Behavioral variant frontotemporal dementia (bvFTD) and primary psychiatric disorders (PPD), such as mood, psychotic, and autism spectrum disorders, share similar clinical characteristics of behavior and social cognition. Better understanding of clinical progression in bvFTD and PPD is essential for adequate disease monitoring and trial design.

Methods: In this longitudinal study (N = 89), patients with bvFTD and PPD with at least one follow-up assessment were included from the Social Brain Project of the Alzheimer Center Amsterdam.

Article Synopsis
  • The European LeukemiaNet (ELN) previously created genetic risk classifications using data from younger adults who underwent intensive chemotherapy.
  • New research has emerged analyzing patients who receive less-intensive therapies, highlighting the need for tailored classifications.
  • This has led to a proposal for a new ELN genetic risk classification specifically designed for patients undergoing these alternative treatment options.

  • Symptomatic dermographism (SD) is a type of skin condition that causes hives and itching when the skin is stroked, and this study used magnetoencephalography (MEG) to explore the brain's role in this condition.
  • Twelve SD patients and fourteen control participants were tested for itch response and brain activity after exposure to histamine and skin stroking, revealing that SD patients reported greater itch levels than controls.
  • The study found no major brain activity differences between groups but observed notable changes in brain wave patterns, suggesting that SD may involve abnormal itch processing linked to the central nervous system.

Article Synopsis
  • - Obesity affects over 650 million people worldwide and is linked to significant health issues, making it a global health concern.
  • - Research suggests that the gut microbiome might play a role in weight regulation, but there's limited human evidence to confirm this connection compared to studies done on rodents.
  • - The review proposes that while more research is needed to link the gut microbiome to human obesity, certain modified probiotics and high doses of microbial metabolites could help in managing obesity.

Objective: To test the hypothesis that self-rated personality disorder (PD) symptoms are a significant and clinically relevant predictor of treatment outcomes in a naturalistic treatment setting specialized in trauma-focused treatment using a single-group pretest-posttest design.

Method: Treatment-seeking patients reporting clinical levels of posttraumatic stress disorder (PTSD) symptoms filled out questionnaires at intake and after treatment. The primary outcome was change in PTSD severity after treatment, measured by the PTSD Checklist for DSM-5 (PCL-5).

Background The association between common carotid artery intima-media thickness (CCA-IMT) and incident carotid plaque has not been characterized fully. We therefore aimed to precisely quantify the relationship between CCA-IMT and carotid plaque development. Methods and Results We undertook an individual participant data meta-analysis of 20 prospective studies from the Proof-ATHERO (Prospective Studies of Atherosclerosis) consortium that recorded baseline CCA-IMT and incident carotid plaque involving 21 494 individuals without a history of cardiovascular disease and without preexisting carotid plaque at baseline.

Department of Medical Microbiology and Infection Control, Amsterdam UMC, Location Vrije Universiteit Medical Center, De Boelelaan 1108, 1081 HZ Amsterdam, The Netherlands.

Finding new anti-tuberculosis compounds with convincing in vivo activity is an ongoing global challenge to fight the emergence of multidrug-resistant Mycobacterium tuberculosis isolates. In this study, we exploited the medium-throughput capabilities of the zebrafish embryo infection model with Mycobacterium marinum as a surrogate for M. tuberculosis.

Advances in Thiopurine Drug Delivery: The Current State-of-the-Art.

Eur J Drug Metab Pharmacokinet

November 2021

Department of Gastroenterology and Hepatology, AGEM Research Institute, Amsterdam University Medical Center, Location Vrije Universiteit Medical Center, Amsterdam, The Netherlands.

Thiopurines (mercaptopurine, azathioprine and thioguanine) are well-established maintenance treatments for a wide range of diseases such as leukemia, inflammatory bowel disease (IBD), systemic lupus erythematosus (SLE) and other inflammatory and autoimmune diseases in general. Worldwide, millions of patients are treated with thiopurines. The use of thiopurines has been limited because of off-target effects such as myelotoxicity and hepatotoxicity.

Somatic copy number alterations can be detected in cell-free DNA (cfDNA) by shallow whole genome sequencing (sWGS). PCR is typically included in library preparations, but a PCR-free method could serve as a high-throughput alternative. To evaluate a PCR-free method for research and diagnostics, archival peripheral blood or bone marrow plasma samples, collected in EDTA- or lithium-heparin-containing tubes, were collected from patients with non-small-cell lung cancer (n = 10 longitudinal samples; 4 patients), B-cell lymphoma (n = 31), and acute myeloid leukemia (n = 15), or from healthy donors (n = 14).

Carbohydrates in allergy: from disease to novel immunotherapies.

Trends Immunol

July 2021

Department of Experimental Immunology, Amsterdam UMC, location Academic Medical Center (AMC), Amsterdam, The Netherlands; Amsterdam Infection and Immunity Institute, Amsterdam, The Netherlands. Electronic address:

Respiratory allergic disorders are a global public health problem that are responsible for substantial morbidity and healthcare expenditure. Despite the availability of allergen immunotherapy (AIT), its efficacy is suboptimal and regimens are lengthy, with a significant risk of potentially severe side effects. Studies on the recognition of allergens by immune cells through carbohydrate-lectin interactions, which play a crucial role in immune modulation and pathogenesis of allergy, have paved the way for improvements in AIT.

Posttraumatic stress disorder (PTSD) is a serious and relatively common mental disorder causing a high burden of suffering. Whereas evidence-based treatments are available, dropout and non-response rates remain high. PTSD and Cluster C personality disorders (avoidant, dependent or obsessive-compulsive personality disorder; CPD) are highly comorbid and there is evidence for suboptimal treatment effects in this subgroup of patients.

Alterations in dopamine signalling have been implied in autism spectrum disorder (ASD), and these could be associated with the risk of developing a psychotic disorder in ASD adults. Negative social experiences and feelings of social defeat might result in an increase in dopamine functioning. However, few studies examined dopamine functioning in vivo in ASD.
