Angiogenesis and immune protection are essential at the onset of tumorigenesis. Angiogenesis serves to nourish the tumor, and prevention of immune defenses, for example, by dendritic cells (DCs), allows tumor growth. In this study, we investigated whether there are factors with dual functions that are both angiogenic and immunomodulatory and represent a therapeutic target.

Article Synopsis
  • Alzheimer's disease (AD) is a devastating condition characterized by poor outcomes, with no effective treatments currently available, largely due to processes like neuroinflammation damaging nerve cells.
  • The disease’s pathology involves the activation of immune cells like astrocytes and microglia, leading to the accumulation of beta-amyloid and tau proteins, and may be exacerbated by infections from bacteria and viruses, which can trigger harmful inflammatory responses in the brain.
  • Current research focuses on developing antibodies to clear beta-amyloid plaques and identifying early biomarkers for AD, as the preclinical phase of the disease could provide an important opportunity for effective intervention.

Lung cancer remains the leading cause of cancer death worldwide, with the majority of cases diagnosed in an advanced stage. Early-stage disease non-small cell lung cancer (NSCLC) has a better outcome, nevertheless the 5-year survival rates drop from 60% for stage IIA to 36% for stage IIIA disease. Early detection and optimized perioperative systemic treatment are frontrunner strategies to reduce this burden.

Parkinson's Disease: A Tale of Many Players.

Med Princ Pract

October 2023

Netherlands Institute for Brain Research, Royal Academy of Arts and Sciences, Amsterdam, The Netherlands.

In 2020, more than 9 million patients suffering from Parkinson's disease (PD) were reported worldwide, and studies predict that the burden of this disease will grow substantially in industrial countries. In the last decade, there has been a better understanding of this neurodegenerative disorder, clinically characterized by motor disturbances, impaired balance, coordination, memory difficulties, and behavioral changes. Various preclinical investigations and studies on human postmortem brains suggest that local oxidative stress and inflammation promote misfolding and aggregation of alpha-synuclein within Lewy bodies and cause nerve cell damage.

Imaging the TGFβ type I receptor in pulmonary arterial hypertension.

EJNMMI Res

March 2023

Department Radiology and Nuclear Medicine(s), (Amsterdam Cardiovascular Sciences), Amsterdam UMC, VU University Medical Center, de Boelelaan 1117, Amsterdam, The Netherlands.

Transforming growth factor β (TGFβ) activity is perturbed in remodelled pulmonary vasculature of patients with pulmonary arterial hypertension (PAH), cancer, vascular diseases and developmental disorders. Inhibition of TGFβ, which signals via activin receptor-like kinase 5 (ALK5), prevents progression and development of experimental PAH. The purpose of this study was to assess two ALK5 targeting positron emission tomography (PET) tracers ([C]LR111 and [F]EW-7197) for imaging ALK5 in monocrotaline (MCT)- and Sugen/hypoxia (SuHx)-induced PAH.
