Framework for the Use of External Controls to Evaluate Treatment Outcomes in Precision Oncology Trials.

JCO Precis Oncol

January 2024

National Health and Medical Research Council Clinical Trials Centre, University of Sydney, Camperdown, NSW, Australia.

Advances in genomics have enabled anticancer therapies to be tailored to target specific genomic alterations. Single-arm trials (SATs), including those incorporated within umbrella, basket, and platform trials, are widely adopted when it is not feasible to conduct randomized controlled trials in rare biomarker-defined subpopulations. External controls (ECs), defined as control arm data derived outside the clinical trial, have gained renewed interest as a strategy to supplement evidence generated from SATs to allow comparative analysis.

Article Synopsis
  • Scientists are worried about wrong size estimates (called effect sizes) in small studies about how different drugs affect the brain, so they designed a new model to fix this.
  • They looked at differences in brain features between people who are addicted to certain drugs and those who aren’t, using data from 21 studies with almost 2,000 participants.
  • The study found that smaller studies often had bigger changes in their results when adjusted, showing that using information from other studies can help make results more accurate.

Background: Viruses have high mutation rates, facilitating rapid evolution and the emergence of new species, subspecies, strains and recombinant forms. Accurate classification of these forms is crucial for understanding viral evolution and developing therapeutic applications. Phylogenetic classification is typically performed by analyzing molecular differences at the genomic and sub-genomic levels.

Mycobacterial bioenergetics is a validated target space for antitubercular drug development. Here, we identify BB2-50F, a 6-substituted 5-(N,N-hexamethylene)amiloride derivative as a potent, multi-targeting bioenergetic inhibitor of Mycobacterium tuberculosis. We show that BB2-50F rapidly sterilizes both replicating and non-replicating cultures of M.

The impact of whole of patient nursing assessment frameworks on hospital inpatients: A scoping literature review.

J Adv Nurs

September 2024

Susan Wakil School of Nursing and Midwifery, Faculty of Medicine and Health, University of Sydney, Sydney, New South Wales, Australia.

Introduction: A comprehensive patient assessment is essential for safe patient care. Patient assessment frameworks for nurses are generally restricted to patients who already have altered vital signs and are at risk of deterioration, or to specific risks or body systems such as falls, pressure injury and the Glasgow Coma Score. Comprehensive and structured evidence-based nursing assessment frameworks that consider the whole patient and extend beyond vital signs, specific risks and single systems are not routinely used in inpatient settings but are important to establish early risks for patient deterioration.

Article Synopsis
  • Colo-uterine fistulas are uncommon in medical literature, and their occurrence due to complications from tubo-ovarian abscesses related to chronic sigmoid diverticulitis is particularly rare.
  • A case study of a 68-year-old woman revealed her struggle with ongoing left iliac fossa pain that was initially diagnosed as complicated sigmoid diverticulitis, but subsequent imaging showed a growing mass indicating further complications.
  • Surgical intervention was required, leading to the discovery of a colo-uterine fistula, ultimately resulting in the successful resection of the affected tissues and a positive recovery outcome for the patient.

Reducing and eliminating seclusion and restraint in inpatient settings has been a key area of focus in mental health policy and research for many years. To address this issue, numerous programmes aimed at minimising the use of these practices have been developed over the past two decades, with varying degrees of success. This article reports on research focused on the implementation of a localised, multilevel complex intervention that targeted both organisational and individual factors related to the use of seclusion and restraint.
