The purpose of this study was to study the effect of inhibiting NLRC5 expression and function on CD4 + T cells, and islet and skin transplantation in mice. A murine skin graft model and islet cell transplantation model were established, and the expression of NLRC5 was compared in rejection and immune tolerance groups. Both calpain-2 (CALP2) and tumor necrosis factor-α (TNF-α) contribute to persistent bilateral hypersensitivity in animals subjected to L5 ventral root transection (L5-VRT), a model of selective motor fiber injury without sensory nerve damage. However, specific upstream mechanisms regulating TNF-α overexpression and possible relationships linking CALP2 and TNF-α have not yet been investigated in this model. Previous work from our laboratory showed that motor nerve injury by lumbar 5 ventral root transection (L5-VRT) led to interleukin-6 (IL-6) over-expression in bilateral spinal cord, and that intrathecal administration of IL-6 neutralizing antibody delayed the induction of mechanical allodynia in bilateral hind paws. However, early events and upstream mechanisms underlying spinal IL-6 expression following L5-VRT require elucidation. α-Synuclein (α-Syn) is hypothesized to have a critical role in sporadic and genetic cases of Parkinson's disease (PD) in which Lewy bodies, as the hallmark of PD, are formed from abnormal aggregates of α-Syn. To determine the role of α-Syn in the motor and cognitive dysfunction observed in PD, a Drosophila melanogaster model was established to investigate the electrophysiological and ethological changes caused by overexpression of α-Syn. The present data indicated that α-Syn overexpression reduced the synaptic transmission of cholinergic neurons by modulating the calcium channel currents in the projection neurons in the antennal lobe region of the Drosophila brain, as well as the learning and memory ability of the flies.
