Fatal familial insomnia (FFI) is a rare autosomal dominant neurodegenerative disorder characterized by rapidly progressive dementia, severe sleep disturbances, and autonomic dysfunction. The clinical manifestations of FFI can exhibit substantial variations, making it crucial to rule out other conditions, such as autoimmune encephalitis and Creutzfeldt-Jakob disease, during early diagnosis. In this study, we describe the case of a 58-year-old man who experienced persistent insomnia, autonomic symptoms, gait instability, and rapidly progressive dementia.

Introduction: The relationship between immune-related thyroid dysfunction (irTD) and survival rates in cancer patients remains unclear. Furthermore, the impact of variations in immunotherapy line numbers and pathological types among lung cancer patients on this relationship has not been fully elucidated. This study aims to evaluate the potential of irTD as a prognostic marker for immunotherapy in Chinese patients with lung cancer.

This study introduced a hydrogel dressing, termed SODex-gel, which was constructed by establishing Schiff base and hydrogen bonds with the precursors of oxidized dextran (ODex) and succinic dihydrazide (SD)-modified sodium alginate (SD--SA). Through comprehensive and studies, the adhesive properties, self-healing capabilities, hemostatic potential, and wound healing efficacy of the SODex-gel dressing were meticulously evaluated. The H NMR, FTIR, and TGA analyses confirmed the fabrication of the SODex-gel dressing and its constituent elements.

Sox genes encode a family of transcription factors that regulate multiple biological processes during metazoan development, including embryogenesis, tissue homeostasis, nervous system specification, and stem cell maintenance. The planarian Dugesia japonica contains a reservoir of stem cells that grow and divide continuously to support cellular turnover. However, whether SOX proteins retain these conserved functions in planarians remains to be determined.

Background: Attention Deficit Hyperactivity Disorder (ADHD) is a prevalent neurodevelopmental disorder in childhood, characterized by symptoms of inattention, hyperactivity, and impulsivity. Impaired inhibitory control is observed in the majority of individuals with ADHD. Understanding the relationship between inhibitory control and the developmental trajectory of ADHD is essential for informing clinical prognosis and guiding early interventions.

Objective: Limb ischemia-reperfusion injury caused by repeated tourniquet application usually leads to acute kidney injury, adversely affecting patient prognosis. This study aimed to investigate the renoprotective effect of remote ischemic preconditioning (RIPC) in patients undergoing extremity surgery with repeated tourniquet application.
Methods: 64 patients were enrolled and randomly divided into an RIPC group and a control group, with 32 patients in each.

Background: Cerebral autosomal dominant arteriopathy with subcortical infarcts and leukoencephalopathy (CADASIL) is one of the most common inherited cerebral small vessel diseases caused by the NOTCH3 gene mutation. This mutation leads to the accumulation of NOTCH3 extracellular domain protein (NOTCH3) into the cerebral arterioles, causing recurrent stroke, white matter lesions, and cognitive impairment. With the development of gene sequencing technology, cysteine-sparing mutations can also cause CADASIL disease, however, the pathogenicity and pathogenic mechanisms of cysteine-sparing mutations remain controversial.

This study proposes a public opinion monitoring model that combines the K-means clustering algorithm with Particle Swarm Optimization (PSO) to enhance the accuracy and effectiveness of public opinion monitoring on social media. The model's performance across various dissemination indicators is studied in detail. Through experiments conducted on social media datasets, the study comprehensively evaluates the model from four dimensions: dissemination speed, scope, depth, and sentiment dissemination effectiveness.

The Mpox virus (MPXV) has emerged as a formidable orthopoxvirus, posing an immense challenge to global public health. An understanding of the regulatory mechanisms of MPXV infection, replication and immune evasion will benefit the development of novel antiviral strategies. Despite the involvement of G-quadruplexes (G4s) in modulating the infection and replication processes of multiple viruses, their roles in the MPXV life cycle remain largely unknown.

The research aims to investigate the mechanical response of footfalls at different velocities to understand the mechanism of heel injury and provide a scientific basis for the prevention and treatment of heel fractures. A three-dimensional solid model of foot drop was constructed using anatomical structures segmented from medical CT scans, including bone, cartilage, ligaments, plantar fascia, and soft tissues, and the impact velocities of the foot were set to be 2 m/s, 4 m/s, 6 m/s, 8 m/s, and 10 m/s. Explicit kinetic analysis methods were used to investigate the mechanical response of the foot landing with different speeds to explore the damage mechanism of heel bone at different impact velocities.

China has implemented the "tiered medical services" policy since 2015, while there is a paucity of data evaluating the the current status of chronic obstructive pulmonary disease (COPD) management under the system. Characteristics and treatments from 11,905 COPD patients in 88 hospitals across different tiers in China were included and analyzed. We assessed the statistical significance of differences by one way analysis of variance (ANOVA) for continuous variables and with the chi-squared test for categorical variables.

Genome-wide association studies (GWASs) have reported multiple risk loci for schizophrenia (SCZ). However, the majority of the associations were from populations of European ancestry. Here we conducted a large-scale GWAS in Eastern Asian populations (29,519 cases and 44,392 controls) and identified ten Eastern Asian-specific risk loci, two of which have not been previously reported.

Cytokine storm is a life-threatening systemic hyper-inflammatory state caused by different etiologies, in which the bulk production of pro-inflammatory cytokines from activated macrophages has a central role. Integrated stress response (ISR) comprises several protective signaling pathways, leading to phosphorylation of eukaryotic initiation factor 2α (eIF2α) and repression of protein translation. Emerging evidence suggests that ISR induction may elicit anti-inflammatory effects.

Spinal cord injury (SCI) is a common neurological trauma that cannot be completely cured with surgical techniques and medications. In this study, we established a mouse SCI model and used an adeno-associated virus (AAV) to achieve the high expression of sonic hedgehog (Shh) at the injury site to further investigate the therapeutic effect and mechanism of Shh on SCI. The results of the present study show that Shh may promote motor function recovery.

Effective delivery of sufficient doxorubicin (DOX) molecules in tumors is hindered by the complex biological barriers. Herein, a DOX-loaded sodium alginate-based injectable hydrogel (DOX@MHB-conj-SA) was designed by the Michael addition reactions between the sulfydryl in cross-linkers and the double bonds in a derivative of sodium alginate. DOX@MHB-conj-SA was administrated to CT26 tumor-bearing mice via peritumoral injection for locoregional treatment of colorectal cancer by inducing apoptosis and pyroptosis.

Nasopharyngeal carcinoma (NPC) refers to a cancerous tumor that develops in the upper and side walls of the nasopharyngeal cavity. Typically, individuals are often diagnosed with the disease when it has already progressed significantly, and those with advanced NPC tend to have an unfavorable outlook in terms of response rate to targeted treatments and overall clinical survival. Various molecular mechanisms, including Myeloid-derived suppressor cells and factors like PD-L1, have been explored to enhance the outcome of NPC.

Plant microRNAs (miRNAs) are small non-coding RNA molecules that usually negatively regulate gene expression at the post-transcriptional level. Recent data reveal that plant miRNAs are not limited to individual plants but can transfer across different species, allowing for communication with the plant, animal, and microbial worlds in a cross-kingdom approach. This review discusses the differences in miRNA biosynthesis between plants and animals and summarizes the current research on the cross-species regulatory effects of plant miRNAs on nearby plants, pathogenic fungi, and insects, which can be applied to crop disease and pest resistance.
