We develop a technology based on competitive adsorption between drug molecules and water, specifically designed to address the critical issue of poor drug solubility. By specially engineering silica nanosurfaces with ultrahigh densities of silanol, we significantly enhance their affinity for both drug molecules and water, with a notably greater increase in water affinity. Such surfaces can effectively adsorb a variety of drug molecules under dry conditions.

We introduce a quantum-classical generative model for small-molecule design, specifically targeting KRAS inhibitors for cancer therapy. We apply the method to design, select and synthesize 15 proposed molecules that could notably engage with KRAS for cancer therapy, with two holding promise for future development as inhibitors. This work showcases the potential of quantum computing to generate experimentally validated hits that compare favorably against classical models.

Quality control (QC) in mass spectrometry (MS)-based proteomics is mainly based on data-dependent acquisition (DDA) analysis of standard samples. Here, we collect 2754 files acquired by data independent acquisition (DIA) and paired 2638 DDA files from mouse liver digests using 21 mass spectrometers across nine laboratories over 31 months. Our data demonstrate that DIA-based LC-MS/MS-related consensus QC metrics exhibit higher sensitivity compared to DDA-based QC metrics in detecting changes in LC-MS status.

The conversion of nitrate-rich wastewater and biomass-derived blocks into high-value products using renewably generated electricity is a promising approach to modulate the artificial carbon and nitrogen cycle. Here, a new synthetic strategy of WO sub-nanoclusters is reported and supported on carbon materials as novel efficient electrocatalysts for nitrate reduction and its coupling with α-keto acids. In acidic solutions, the NH-NHOH selectivity can also optimized by adjusting the potential, with the total FE exceeding 80% over a wide potential range.

Long-term durable remission in patients with B cell malignancies following chimeric antigen receptor (CAR)-T cell immunotherapy remains unsatisfactory, often due to antigen escape. Malignant B cell transformation and oncogenic growth relies on efficient ATP synthesis, although the underlying mechanisms remain unclear. Here, we report that YTHDF2 facilitates energy supply and antigen escape in B cell malignancies, and its overexpression alone is sufficient to cause B cell transformation and tumorigenesis.

He proved a formula, known today as the Bowen's formula, that relates the Hausdorff dimension of a conformal repeller to the zero of a pressure function defined by a single conformal map. Aging compromises antitumor immunity, but the underlying mechanisms remain elusive. Here, we report that aging impairs the generation of CD8 tissue resident memory T (T) cells in nonlymphoid tissues in mice, thus compromising the antitumor activity of aged CD8 T cells, which we also observed in human lung adenocarcinoma. We further identified that the apoptosis regulator BFAR was highly enriched in aged CD8 T cells, in which BFAR suppressed cytokine-induced JAK2 signaling by activating JAK2 deubiquitination, thereby limiting downstream STAT1-mediated T reprogramming.

Triple-negative breast cancers (TNBC) are treated primarily by chemotherapy and lack clinically validated therapeutic targets. In particular, inhibitors of the PI3K/AKT/mTOR pathway, abnormally activated in many breast cancers, failed to achieve clinical efficacy in TNBC due to the development of adaptive drug resistance, which is largely driven by the transcriptomic plasticity of TNBC. Expression of CDK8/19 Mediator kinases that control transcriptional reprogramming correlates with relapse-free survival and treatment failure in breast cancer patients, including TNBC.

Continuously monitoring human airway conditions is crucial for timely interventions, especially when airway stents are implanted to alleviate central airway obstruction in lung cancer and other diseases. Mucus conditions, in particular, are important biomarkers for indicating inflammation and stent patency but remain challenging to monitor. Current methods, reliant on computational tomography imaging and bronchoscope inspection, pose risks due to radiation and lack the ability to provide continuous real-time feedback outside of hospitals.
