Characterization of accurate compositions and total abundance of homologous drug-metabolizing enzymes, such as UDP glucuronosyltransferases (UGTs), is important for predicting the fractional contribution of individual isoforms involved in the metabolism of a drug for applications in physiologically based pharmacokinetic (PBPK) modeling. Conventional targeted proteomics utilizes surrogate peptides, which often results in high technical and interlaboratory variability due to peptide-specific digestion leading to data inconsistencies. To address this problem, we developed a novel conserved-plus-surrogate peptide (CPSP) approach for determining the accurate compositions and total or cumulative abundance of homologous UGTs in commercially available pooled human liver microsomes (HLM), human intestinal microsomes (HIM), human kidney microsomes (HKM), and human liver S9 (HLS9) fraction.

Background: Environmental exposures are commonly estimated using spatial methods, with most epidemiological studies relying on home addresses. Passively collected smartphone location data, like Google Location History (GLH) data, may present an opportunity to integrate existing long-term time-activity data.
Objectives: We aimed to evaluate the potential use of GLH data for capturing long-term retrospective time-activity data for environmental health research.

Aspirin overdose is still a common cause of presentation to the emergency department and is commonly seen in the setting of one-time, accidental or intentional ingestion of large amounts of salicylate-containing compounds, though can occur with long-term ingestion of super-therapeutic doses of medications. Salicylate toxicity has a predictable progression from early respiratory alkalosis to late metabolic acidosis. We present the case of a 14-year-old girl who intentionally ingested a handful of Aspirin and despite appropriate therapy, developed transient proximal tubule dysfunction.

Attention may be important for actively maintaining posture during computer tasks, resulting in a dual-task tradeoff, where maintaining posture through extrinsic feedback imposes cognitive load. Mindfulness may make intrinsic postural feedback (which imposes less cognitive load) more available. Therefore, we hypothesized that the use of biofeedback would improve posture and negatively impact game performance; additionally, higher levels of mindfulness would be associated with lower game performance costs in the biofeedback condition.

As a promising amendment, biochar has excellent characteristics and can be used as a remediation agent for diverse types of soil pollution. Biochar is mostly made from agricultural wastes, forestry wastes, and biosolids (eg, sewage sludge), but not all the biochar has the same performance in the improvement of soil quality. There is a lack of guidelines devoted to the selection of biochar to be used for different types of soil pollution, and this can undermine the remediation efficiency.

Hydrothermal pretreatment of waste activated sludge (WAS) could eliminate the rate limiting step of anaerobic digestion (AD) -hydrolysis. However, the high organic loading rate may cause acid accumulation, thus leading to an unstable system. This study compared the effect of different hydrochar (HC2-260°C and HC3-320°C) and biochar (BC5-500°C and BC7-700°C) on AD of hydrothermal pretreated WAS (HPS).

Anaerobic digestion (AD) of waste activated sludge (WAS) has been widely used, while it poses problems including low methane yield and production rate. Hydrochar is produced by hydrothermal liquefaction of biomass; however, little is known about the role of hydrochar in promoting AD of WAS. The present study showed that hydrochar increased the methane production rate by 30.

Members of the SH3- and ankyrin repeat (SHANK) protein family are considered as master scaffolds of the postsynaptic density of glutamatergic synapses. Several missense mutations within the canonical SHANK3 isoform have been proposed as causative for the development of autism spectrum disorders (ASDs). However, there is a surprising paucity of data linking missense mutation-induced changes in protein structure and dynamics to the occurrence of ASD-related synaptic phenotypes.

The mechanism of arsenic (As) immobilization in soils is crucial for improving photosynthetic pigments and antioxidants in food crops. The effects of soil amendments with arbuscular mycorrhizal fungi (AMF), biochar (BC), selenium (Se), sulfur (S) and Si-gel on the concentrations of chlorophyll, carotenoid, proline, malondialdehyde (MDA), and the activity of ascorbate peroxidase (APX), guaiacol peroxidase (POD), and catalase (CAT) were studied in BARI pea under As stress. Soil amendments with AMF, Se, Si-gel and S enhanced chlorophyll a and total chlorophyll contents by 31-35% and 60-75%, respectively.

We present a unique case of a patient simultaneously diagnosed with solitary fibrous tumor (SFT) and unrelated adenocarcinoma of the lung, both proven with separate pathology. It was subsequently found that the SFT had metastasized to the brain by additional pathology, and not the predicted adenocarcinoma. SFTs are a rare mesenchymal neoplasm that accounts for less than 2% of all reported soft tissue tumors.

Aim: We tested the hypothesis that patients with succinic semialdehyde dehydrogenase (SSADH) deficiency on taurine would have decreased cortical excitability as measured by transcranial magnetic stimulation (TMS) and improved cognition, due to taurine's partial GABA(A and B) receptor agonist effects and rescue in the null mouse model from status epilepticus and premature lethality.
Method: Biomarkers including neuropsychological testing, TMS, and CSF metabolites were studied in a cohort of patients on and off three months' taurine treatment.
Results: Seven patients (5M/2F; age range 12-33 years) were enrolled in this open-label crossover study.
