October 1995

Department of Pharmacology and Toxicology, Faculty of Veterinary Medicine, Warsaw Agricultural University, ul. Nowoursynowska 166, 02-766 Warszawa, Poland.

Inorganic nitrite introduced into the living organism is rapidly converted into nitrate and nitric oxide (NO). It is known that nitrite decreases ammonia use and urea formation in vitro and it seems that nitrite also influences these processes in vivo. However, the mechanism underlying this effect is not known.

Polyamines in cow's and sow's milk.

Comp Biochem Physiol B Biochem Mol Biol

July 1995

Department of Animal Physiology, Faculty of Veterinary Medicine, Warsaw Agricultural University, Poland.

There were considerably interspecies, individual, lactation and age related variabilities in the concentration of spermidine and spermine in cow's and sow's milk. Concentration of spermidine was similar in cow's and sow's milk, whereas spermine level was higher in sow's milk. The level of spermine was higher than spermidine throughout the lactation in sow's milk, whereas spermine in cow's milk was secreted only at the beginning of lactation (collostrum and milk of the first month of lactation).

The effect of CCK-8 administered into the duodenal lumen and into the systemic blood on pancreatic secretion and duodenal migrating myoelectric complex (MMC) was studied in four calves. Simultaneous MMC recordings and collections of pancreatic juice were performed on valves that had been fasted overnight. Intraduodenal (0, 100, and 300 pmol/kg body wt) and intravenous (0, 30, and 100/pmol kg) infusions of CCK-8 were made for 5 min during the no spiking activity (NSA) phase of duodenal MMC associated with a nadir of periodic pancreatic secretion.

The aim of this study was to evaluate the effect of different leucocyte count from normal and inflamed uterus in excess of PMNs on the development of mouse embryos during the morula or blastocyst stages in vitro. Results showed that cultivation of blastocysts in the presence of leucocytes washed from the uterus, after mediation of specific or nonspecific endometritis at a concentration of 1 x 10(4), led to significant inhibition of embryo development. No similar observations were made with blastocysts cultivated in the presence of leucocytes obtained from the normal uterus at 2-4 days after spontaneous ovulation.

Bile samples, from slaughtered cattle harbouring between 120 and 280 adult lancet flukes, were used to investigate the range of somatic proteins inducing local antibody responses in naturally infected animals. Lancet fluke infections induced local (bile) antibody responses against Tris-buffered saline (TBS) soluble, sodium dodecyl sulphate (SDS) soluble and 2-mercaptoethanol (2-Me) soluble somatic proteins of adult Dicrocoelium dendriticum. IgA antibody isotypes predominated in the response against buffer-soluble somatic antigens, whereas SDS-soluble and 2-Me-soluble proteins induced similar level of both IgA and IgG1 antibodies.

Unlabelled: Transforming growth factor-beta 1 (TGF-beta 1) exerted growth-inhibitory effect on L1210 leukemic cell line, manifested by the decrease in viable and increase in dead cells. The cell death evoked by TGF-beta 1 was both necrotic and apoptotic, quantified by the trypan blue exclusion method and apoptotic index, respectively. The induction of programmed cell death by TGF-beta 1 was confirmed by gel electrophoresis of DNA, where the characteristic 'DNA ladder' resulting from the internucleosomal DNA cleavage was visualized.

Pituitary adenylate cyclase-activating polypeptide (PACAP) stimulates exocrine pancreas in conscious preruminating calves.

Comp Biochem Physiol C Pharmacol Toxicol Endocrinol

September 1994

Department of Animal Physiology, Faculty of Veterinary Medicine, Warsaw Agricultural University, Poland.

The effects of new hypothalamic peptides, PACAP-27 and PACAP-38, and secretin and VIP on the interdigestive pancreatic secretion and duodenal myoelectric activity during the asecretory phase of the pancreatic interdigestive cycle, compared with the milk ingestion phase, were examined in five calves. Peptides were infused for 5 min into the external jugular vein (0, 3, 10, 30 and 100 pmol/kg body wt during the asecretory phase of the pancreatic interdigestive cycle, and the pancreatic secretory response was compared with that obtained during milk ingestion. Intravenous infusion of PACAP-27 caused dose-related stimulation of pancreatic juice flow and bicarbonate and protein output; this effect was identical to infusion of secretin.

With the use of an enzyme-linked immunosorbent assay (ELISA), it was revealed that surface antigens of both adult and fourth-stage larvae (L4) of Ostertagia circumcincta induced high levels of serum IgG antibodies, while serum IgA antibody levels were low but increased significantly (P < 0.01) after infection. Immunofluorescence studies on the surface of viable L4 and adult nematodes showed that the IgG response was stage-specific only in animals vaccinated with adult surface extracts.

Plasma secretin and cholecystokinin (CCK) levels and the periodic secretions of the exocrine pancreas were studied simultaneously with the duodenal migrating myoelectric complexes (MMC) in six milk-fed calves which had been starved overnight. The experiments were performed first when the calves were 10 to 16 days old and subsequently when they were 36 to 45 days old. Plasma secretin and the secretion of pancreatic juice fluctuated periodically in phase with the duodenal MMC: plasma secretin, and the pancreatic secretion of water and protein were significantly higher during the phase of irregular spiking activity than during the phase of no spiking activity in both investigations.
