A field resource for the glioma cerebrospinal fluid proteome: impacts of resection and location on biomarker discovery.

Background: While serial sampling of glioma tissue is rarely performed prior to recurrence, cerebrospinal fluid (CSF) is an underutilized longitudinal source of candidate glioma biomarkers for understanding therapeutic impacts. However, the impact of key variables to consider in longitudinal CSF samples for monitoring biomarker discovery, including anatomical location and post-surgical changes, remains unknown.

Methods: Aptamer-based proteomics was performed on 147 CSF samples from 74 patients, 71 of whom had grade 2-4 astrocytomas or grade 2-3 oligodendrogliomas.

A Hitchhiker's Guide Toward CSF Biomarkers for Neuro-Oncology.

Cerebrospinal fluid (CSF) has emerged as a valuable liquid biopsy source for glioma biomarker discovery and validation. CSF produced within the ventricles circulates through the subarachnoid space, where the composition of glioma-derived analytes is influenced by the proximity and anatomical location of sampling relative to tumor, in addition to underlying tumor biology. The substantial gradients observed between lumbar and intracranial CSF compartments for tumor-derived analytes underscore the importance of sampling site selection.

Distinguishing Neural Correlates of Prediction Errors on Perceptual Content and Detection of Content.

Accounting for why discrimination between different perceptual contents is not always accompanied conscious detection of that content remains a challenge for predictive processing theories of perception. Here, we test a hypothesis that detection is supported by a distinct inference within generative models of perceptual content. We develop a novel visual perception paradigm that probes such inferences by manipulating both expectations about stimulus content (stimulus identity) and detection of content (stimulus presence).

Biomarkers.

Background: Alzheimer's disease is a devastating neurodegenerative disorder with a complex pathogenesis. One main pathological feature utilised in diagnosis is neurodegeneration or neuronal injury, which is reflected in reductions in cerebral glucose metabolism measured by [18F]Fluorodeoxyglucose ([18F]FDG) positron emission tomography (PET). Here we evaluated the involvement of glial reactivity measured with magnetic resonance spectroscopy (MRS) and cerebral blood flow measured with arterial spin labelling (ASL) on [18F]FDG PET as a measure of cerebral glucose metabolism.

Everting Sutures for Entropion Repair: Recurrence Rate and Quality of Life Metrics.

Purpose: The study aimed to assess the efficacy and recurrence rate of everting sutures as a sole procedure in entropion repair, its impact on quality of life during the coronavirus disease pandemic, and to determine if this procedure remains an option to offer after the pandemic.

Design: Retrospective clinical cohort study.

Methods: Retrospective study of 58 patients who had everting sutures for entropion repair over a 1-year period.

Several states have recently implemented laws that allow pharmacists to prescribe contraceptives to women without the need to see a physician. We study the effect of these state laws on fertility rates.

DNA aptamers that modulate biological activity of model neurons.

There is an urgent need for agents that promote health and regeneration of cells and tissues, specifically to treat diseases of the aging nervous system. Age-associated nervous system degeneration and various diseases are driven by many different biochemical stresses, often making it difficult to target any one disease cause. Our laboratory has previously identified DNA aptamers with apparent regenerative properties in murine models of multiple sclerosis by selecting aptamers that bind oligodendrocyte membrane preparations.

Organizational on-boarding practices and shifts in alcohol misuse among at-risk college recruits.

With a significant proportion of college students in many countries engaging in risky drinking behavior, this study examines the tendency of such young adults to 'mature out' of such behavior in their first year of employment after graduating, and the degree to which three mainstream organizational on-boarding experiences may expedite such 'maturing out'. Focusing on newcomers' experiences with alcohol-oriented job orientation, job empowerment, and organizational efforts to facilitate the development of supportive peer relationships, we test hypotheses regarding the direct effects of time on the change in alcohol misuse among those reporting misuse in college, as well as the degree to which individual on-boarding experiences account for the variance in young adults' maturing out trajectories over the course of their first year of employment. Findings generated from data collected from over 400 young adults over multiple waves offer important theoretical and practical implications regarding how and why particular onboarding tactics may be more or less effective in influencing newcomers' health-related behaviors.

Longitudinal glioma monitoring via cerebrospinal fluid cell-free DNA.

Purpose: Current methods for glioma response assessment are limited. This study aimed to assess the technical and clinical feasibility of molecular profiling using longitudinal intracranial CSF from patients with gliomas.

Experimental Design: Adults with gliomas underwent longitudinal intracranial CSF collection via Ommaya reservoirs or ventriculoperitoneal shunts.

Vasculitic neuropathy is caused by inflammatory destruction of nerve blood vessels resulting in nerve ischemia. Nerve vasculitis can be divided into two categories based on vessel size - large arteriole vasculitis (≥75 µm) and microvasculitis (<75 µm). Herein, we characterize the clinical features of nerve large-arteriole vasculitis compared to nerve microvasculitis.

A Retrospective Audit of Osteoporosis Management Following Fragility Fractures in a Hospital Setting.

Background Fragility fractures, often caused by osteoporosis, are a major public health concern among the growing population of the United Kingdom (UK). In addition to being a major source of illness and mortality, the rising incidence of osteoporosis places a heavy strain on healthcare systems if it is not adequately managed. In order to lower the risk of additional fractures, current guidelines place a strong emphasis on the timely evaluation and treatment of fragility fractures.

The Efficacy of Biodegradable Temporising Matrix for Upper Limb Reconstruction: A Systematic Review and Meta-Analysis.

The objective of this systematic review and meta-analysis is to assess the efficacy of the biodegradable temporising matrix (BTM) (NovoSorb; PolyNovo Biomaterials Pty Ltd, Port Melbourne, Victoria, Australia) in the reconstruction of complex upper extremity wounds. The authors conducted a systematic review and meta-analysis as per the Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) guidelines assessing the efficacy of BTM in complex upper extremity wound reconstruction. The primary outcome measures were successful BTM integration and the proportion of wounds healed.

Expert Perspective: Management of Relapses in Giant Cell Arteritis.

Giant cell arteritis (GCA) is a relapsing large-vessel vasculitis with risk of serious ischemic manifestations including vision loss and vascular damage in the form of large-artery stenosis, aneurysms and dissections. Approximately 50% of patients treated with glucocorticoid (GC) monotherapy and 30% of patients receiving adjunctive therapy with tocilizumab experience disease relapses, often during the first 2 years after diagnosis. Although most relapses in GCA do not involve life- or organ-threatening presentations and can be controlled successfully, frequent relapses may lead to increased use of GC and consequent treatment-related morbidity, in addition to risk of further vascular damage.

The effects of plasma exchange and glucocorticoids on early kidney function among patients with ANCA-associated vasculitis in the PEXIVAS trial.

Therapeutic plasma exchange (PLEX) is an adjunctive treatment for patients with anti-neutrophil cytoplasmic antibody (ANCA)-associated vasculitis and kidney involvement. Little is known about the effect of PLEX on early changes in kidney function. This post-hoc analysis of the PEXIVAS trial investigated the effects of PLEX on changes in kidney function within 12 months.

Treatment trends of systemic lupus erythematosus from 2007 to 2023 in the USA.

Objective: To characterise the changing trends in the pharmacological management of SLE in the USA between 2007 and 2023 as new treatment options emerged.

Methods: In a retrospective cohort study using data from OptumLabs Data Warehouse, we characterised the annual prevalent (ie, all) and incident (ie, new) use of antimalarials, glucocorticoids and immunosuppressive medications among patients with SLE from 2007 to 2023 and assessed for changing trends over time.

Results: We identified 19 122 adults with SLE; they were 51.

Deposition simulations of realistic dosages in patient-specific airways with two- and four-way coupling.

Inhalers spray over 100 million drug particles into the mouth, where a significant portion of the drug may deposit. Understanding how the complex interplay between particle and solid phases influence deposition is crucial for optimising treatments. Existing modelling studies neglect any effect of particle momentum on the fluid (one-way coupling), which may cause poor prediction of forces acting on particles.

In 2007, Caspi et al. found evidence in two cohorts for an interaction between genetic variation in the FADS2 gene (a gene involved in fatty acid metabolism) and breastfeeding on IQ.

How to measure peak expiratory flow rate.

Peak expiratory flow rate (PEFR) is an important test in the diagnosis and monitoring of respiratory conditions, especially asthma. PEFR measures the speed at which air is forcefully exhaled from full inspiration, expressed in litres per minute (L/min). It involves using a peak flow meter - a simple, affordable, portable and widely available device that can be used in clinic or home settings.

MS is a disease characterised by demyelination of the central nervous system resulting in decreased quality of life, increased anxiety, depression, fatigue, and cognitive dysfunction. Attention is frequently impaired in MS. A previous study demonstrated impairment specifically in the attentional alerting domain.

Quality of life, healthcare usage and finances of UK cancer survivors five years post-diagnosis: a matched controlled study.

Purpose: Assessing the long-term impact of cancer on people's lives is challenging due to confounding issues such as aging and comorbidities. We aimed to investigate this impact by comparing the outcomes of cancer survivors with a matched control cohort.

Methods: This was a cross-sectional survey of breast, colorectal and ovarian cancer survivors approximately 5 years post-diagnosis and a cohort of age, sex and social deprivation-matched controls who had never had a cancer diagnosis.
