Uncertainty in parameter estimates from fitting within-host models to empirical data limits the model's ability to uncover mechanisms of infection, disease progression, and to guide pharmaceutical interventions. Understanding the effect of model structure and data availability on model predictions is important for informing model development and experimental design. The semisynthetic derivatization of natural products is crucial for their continued development as antibiotics. While commercial pleuromutilin derivatives depend on amines for solubility, we demonstrate the high activity and solubility of oligoethylene glycol-substituted pleuromutilins achieved via a one-pot deprotection/attachment approach using thiolates protected as thioesters. The bifunctional linker synthesis is versatile and can be broadly applied to other chemistries.

The well-posedness of the initial-boundary value problem for higher-order quadratic nonlinear Schrödinger equations on the half-line is studied by utilizing the Fokas solution formula for the corresponding linear problem. Using this formula, linear estimates are derived in Bourgain spaces for initial data in spatial Sobolev spaces on the half-line and boundary data in temporal Sobolev spaces suggested by the time regularity of the linear initial value problem. Then, the needed bilinear estimates are derived and used for showing that the iteration map defined via the Fokas solution formula is a contraction in appropriate solution spaces.

Objectives: To implement, examine the feasibility of, and evaluate the performance of quantitative ultrasound (QUS) with a handheld point-of-care US (POCUS) device for assessing liver fat in adults.
Materials And Methods: This prospective IRB-approved, HIPAA-compliant pilot study enrolled adults with overweight or obesity. Participants underwent chemical-shift-encoded magnetic resonance imaging to estimate proton density fat fraction (PDFF) and, within 1 mo, QUS with a POCUS device by expert sonographers and novice operators (no prior US scanning experience).

Each year, millions of animals enter animal shelters across the United States and are met with a variety of potential stressors that can negatively impact their experience, including noise, confinement, and social isolation. Foster care, a unique form of human-animal interaction, is increasingly understood to be an effective tool for improving welfare by allowing animals to escape the stressors of the shelter, providing an environment that allows for greater social interaction, and offering opportunities for improved health and behavior. This review includes 42 published articles, reports, master's theses, and doctoral dissertations that have previously evaluated companion animal foster care programs.

Environmental health studies are of great interest in research to evaluate the mortality-temperature relationship by adjusting spatially correlated random effects as well as identifying significant change points in temperature. However, this relationship is often not expressed using parametric models, which makes identifying change points an even more challenging problem. Animals within social groups respond to costs and benefits of sociality by adjusting the proportion of time they spend in close proximity to other individuals in the group (cohesion). Accuracy of binding free energy calculations utilizing implicit solvent models is critically affected by parameters of the underlying dielectric boundary, specifically, the atomic and water probe radii. Here, a multidimensional optimization pipeline is used to find optimal atomic radii, specifically for binding calculations in the implicit solvent. To reduce overfitting, the optimization target includes separate, weighted contributions from both binding and hydration free energies.
