Determination of the depth of membrane penetration provides important information for studies of membrane protein folding and protein-lipid interactions. Here, we use a combination of molecular dynamics (MD) simulations and depth-dependent fluorescence quenching to calibrate the methodology for extracting quantitative information on membrane penetration. In order to investigate the immersion depth of the fluorescent label in lipid bilayer, we studied 7-nitrobenz-2-oxa-1,3-diazole (NBD) attached to the lipid headgroup in NBD-PE incorporated into POPC bilayer.

The aim of the study was to examine sperm maturation in sturgeon and to establish the localization of the maturation. We demonstrated that sperm maturation occurs in sturgeon outside the testes via dilution of sperm by urine. The process involves the participation of high molecular weight (>10kDa) substances and calcium ions.

Förster resonance energy transfer (FRET) between anthrylvinyl-labeled phosphatidylcholine (AV-PC) as a donor and newly synthesized benzanthrones (referred to here as A8, A6, AM12, AM15 and AM18) as acceptors has been examined to gain insight into molecular level details of the interactions between benzanthrone dyes and model lipid membranes composed of zwitterionic lipid phosphatidylcholine and its mixtures with anionic lipids cardiolipin (CL) and phosphatidylglycerol (PG). FRET data were quantitatively analyzed in terms of the model of energy transfer in two-dimensional systems taking into account the distance dependence of orientation factor. Evidence for A8 location in phospholipid headgroup region has been obtained.

We have observed that berberine prolonged life span and improved viability of pupae and climbing activity of imagoes of wild-type Drosophila melanogaster maintained at 23°C. As a continuation of our studies of berberine effect on life span, we were interested to evaluate the effect of berberine of life span in flies maintained at a higher temperature (28°C) known to accelerate aging in wild type flies. Considering that genetically or pharmacologically induced deficiency of TRP conversion into KYN prolonged life span in a Drosophila model, we compared the effects of berberine, a powerful inhibitor of kynurenine (KYN) formation from tryptophan (TRP), on life span in wild type and in Drosophila melanogaster mutants (vermilion) with deficient TRP-KYN metabolism maintained at 23°C and 28°C.

A series of novel fluorescent benzanthrone dyes have been tested for their ability to identify and characterize fibrillar aggregates of lysozyme prepared by protein denaturation in concentrated ethanol solution (F(eth)) or acidic buffer (F(ac)). Quantitative parameters of the dye association with native and fibrillar protein have been derived from the results of fluorimetric titration. The binding characteristics proved to be different for F(eth)- and F(ac)-bound benzanthrones, highlighting the dye sensitivity to the distinctions in fibril morphology.

Apolipoprotein A-I is amenable to a number of specific mutations associated with hereditary systemic amyloidoses. Amyloidogenic properties of apoA-I are determined mainly by its N-terminal fragment. In the present study Förster resonance energy transfer between tryptophan as a donor and Thioflavin T as an acceptor was employed to obtain structural information on the amyloid fibrils formed by apoA-I variant 1-83/G26R/W@8.

The discharging of dust particles in the afterglow of plasma with large dust density is studied. We used measured electron and metastable dependencies to calculate the rate describing collection of electrons by dust particles by solving the electron balance equation. This rate is compared with the rate calculated using the orbital motion limited (OML) theory.

Mathematical modeling is a convenient way for characterization of complex ecosystems. This approach was applied to study the dynamics of zooplankton in Lake Sevan (Armenia) at different stages of anthropogenic eutrophication with the use of a novel method called discrete modeling of dynamical systems with feedback (DMDS). Simulation demonstrated that the application of this method helps in characterization of inter- and intra-component relationships in a natural ecosystem.

Acanthobdellidans are unique in their organisation and phylogenetic relationships due to having transitional characters that combine features of oligochaetous and achaetous annelids. Alongside the relatively well-studied Acanthobdella peledina Grube, 1851, there is another member of the group, Paracanthobdella livanowi (Epshtein, 1966), with five rows of chaetae and an anterior sucker. It appears that the anterior sucker is weakly developed in small juveniles but acquires a deep cavity in adults.

Transmission of a p-polarized electromagnetic wave through a two-layer plasma structure with spatially nonuniform distributions of electron density in the layers is studied. The case, when the electromagnetic wave is obliquely incident on the structure and is evanescent in both plasma layers, is considered. The conditions for total transparency of the two-layer structure are found for the thin slab case and when the plasma inhomogeneity is weak.

Objectives: The purposes of this work were to detect the microscopic displacements accompanying isometric muscle tissue contraction under different external loads, to explore their spectral characteristics, and to develop physical models capable of explaining the experimental data obtained.
Methods: By means of a spectral Doppler musculoskeletal ultrasound method, spectral characteristics of local isometric skeletal muscle tissue contraction were studied. In this method, the phased fluctuation (Doppler) algorithm is used to estimate tissue displacements within the given sample volume in real time, and the spectral characteristics of recorded displacements are determined.

Up-regulation of kynurenine (KYN) pathway of tryptophan (TRP) was suggested as one of the mechanisms of aging and aging-associated disorders. Genetic and pharmacological impairment of TRP - KYN metabolism resulted in prolongation of life span in Drosophila models. Minocycline, an antibiotic with anti-inflammatory, antioxidant and neuroprotective properties independent of its antibacterial activity, inhibited KYN formation from TRP.

Ultrasound Doppler methods are widely used in clinical practice as prospective investigational tool to study the vascular system and soft biological tissues. Meanwhile, the most general relationship between the power Doppler spectra, spectral characteristics of the scattering fluctuations and the probing ultrasound field parameters for some clinical implementations are still unexplored. Based upon the continuum model of scattering inhomogeneities, a set of the closed-form expressions for the correlation functions and the spectra of Doppler response of soft tissues and blood have been derived.

Motivated by development of lithium-ion batteries, we study the structure and dynamics of LiBF(4) in pure and mixed solvents with various salt concentrations. For this purpose, we have developed force field models for ethylene carbonate, propylene carbonate, dimethyl carbonate, and dimethoxyethane. We find that Li(+) is preferentially solvated by the cyclic and more polar component of the mixtures, as the electrostatic interaction overcomes possible steric hindrances.

Successful use of fluorescence sensing in elucidating the biophysical properties of lipid membranes requires knowledge of the distribution and location of an emitting molecule in the bilayer. We report here that 2,6-bis(1H-benzimidazol-2-yl)pyridine (BBP), which is almost non-fluorescent in aqueous solutions, reveals a strong emission enhancement in a hydrophobic environment of a phospholipid bilayer, making it interesting for fluorescence probing of water content in a lipid membrane. Comparing the fluorescence behavior of BBP in a wide variety of solvents with those in phospholipid vesicles, we suggest that the hydrogen bonding interactions between a BBP fluorophore and water molecules play a crucial role in the observed "light switch effect".
