Background: Assessment of health-related quality of life (HRQoL) in patients with cardiovascular disease (CVD) is impaired by limitations of current patient-reported outcome measures (PROMs). We developed the first cardiovascular disease (CVD) specific electronic PROM for which health items were derived by a fully patient-centered method. This paper reports on the measurement of HRQoL in CVD patients by a novel developed electronic patient-centred PROM based on a preference-based measurement model.

ZIC1 is a context-dependent medulloblastoma driver in the rhombic lip.

Nat Genet

January 2025

Department of Laboratory Medicine and Pathobiology, University of Toronto, Toronto, Ontario, Canada.

Transcription factors are frequent cancer driver genes, exhibiting noted specificity based on the precise cell of origin. We demonstrate that ZIC1 exhibits loss-of-function (LOF) somatic events in group 4 (G4) medulloblastoma through recurrent point mutations, subchromosomal deletions and mono-allelic epigenetic repression (60% of G4 medulloblastoma). In contrast, highly similar SHH medulloblastoma exhibits distinct and diametrically opposed gain-of-function mutations and copy number gains (20% of SHH medulloblastoma).

Repeated muscle micro-trauma may cause severe muscle damage. Diffusion tensor imaging (DTI) exhibits sensitivity to microstructural changes in skeletal muscle. We hypothesize that longer diffusion times enhance sensitivity to micro-trauma and that membrane permeability increases with micro-trauma.

Background: Liquid biopsies offer less burdensome sensitive disease monitoring. Bone marrow (BM) metastases, common in various cancers including neuroblastoma, is associated with poor outcomes. In pediatric high-risk neuroblastoma most patients initially respond to treatment, but in the majority the disease recurs with only 40% long-term survivors, stressing the need for more sensitive detection of disseminated disease during therapy.

User Perspectives and Community-based Interventions, Department of Public Health, University of Southern Denmark, Odense, Denmark.

Background: Young adult cancer survivors, defined as individuals aged 18-39 who have completed primary curative treatment, face numerous age-specific biopsychosocial late effects that impact health-related quality of life negatively. Rehabilitation can enhance participation in life roles, work, leisure activities and health-related quality of life. However, there is a lack of age-specific cancer rehabilitation for this population, leaving many young adults with diminished self-efficacy in managing their challenges, resulting in unmet needs.

Background: Air pollution is a major risk factor for cardiovascular diseases and contributes to health disparities, particularly among minority ethnic groups, who often face higher exposure levels. Knowledge on whether the effect of air pollution on cardiovascular diseases differs between ethnic groups is crucial for identifying mechanisms underlying health disparities, ultimately informing targeted public health strategies and interventions. We explored differences in associations between air pollution and ischemic stroke and ischemic heart disease (IHD) for the six largest ethnic groups in the Netherlands.

Purpose: Placement of zygomatic implants in the most optimal prosthetic position is considered challenging due to limited bone mass of the zygoma, limited visibility, length of the drilling path and proximity to critical anatomical structures. Augmented reality (AR) navigation can eliminate some of the disadvantages of surgical guides and conventional surgical navigation, while potentially improving accuracy. In this human cadaver study, we evaluated a developed AR navigation approach for placement of zygomatic implants after total maxillectomy.

Article Synopsis
  • The oestrous cycle significantly affects breast cancer's response to neoadjuvant chemotherapy (NAC), with treatment during the dioestrus stage resulting in less effectiveness compared to the oestrus stage.
  • In mouse models and human premenopausal cohorts, dioestrus is associated with increased chemoresistance, characterized by more cells undergoing epithelial-to-mesenchymal transition, narrower tumor blood vessels, and higher macrophage levels.
  • Understanding the impact of the oestrous cycle on treatment response could lead to improved timing for chemotherapy, enhancing patient outcomes.

Arterial calcification in the heart-brain axis and cognitive performance over time.

Alzheimers Dement

December 2024

Department of Radiology and Nuclear Medicine, Erasmus MC, Rotterdam, The Netherlands.

Article Synopsis
  • - The study explored the relationship between arteriosclerosis, measured by arterial calcification in the heart-brain axis, and cognitive performance over time, using data from the Rotterdam Study with 2368 participants.
  • - Findings showed that higher levels of arterial calcification were linked to poorer initial cognitive performance and a faster decline in cognitive abilities across multiple domains, particularly influenced by calcification in the intracranial carotid artery.
  • - The results suggest that arterial calcification may drive cognitive decline through changes in blood flow before any neurovascular damage occurs, emphasizing the role of vascular health in maintaining cognitive function.

Background: Our aim is to investigate the association of treatment with survival in patients with diffuse intrinsic pontine glioma (DIPG) by examining 6 historical treatment paths.

Methods: We retrospectively analyzed data from 409 patients with radiologically centrally reviewed DIPG, sourced from the German Society of Pediatric Oncology and Hematology HIT-HGG trial database and the SIOPE-DIPG/DMG Registry. Survival outcomes were estimated using the Kaplan-Meier method, and univariable and multivariable Cox proportional hazard models were estimated to study treatment effects.

Background: To prevent the transfer of embryos affected by monogenic conditions and/or chromosomal defects, preimplantation genetic testing (PGT) requires trophectoderm biopsy and cryopreservation. In 2-6% of biopsies, the diagnosis may be inconclusive due to DNA amplification failure or low-quality results. In these cases, a round of re-warming, re-biopsy, and re-cryopreservation is required to obtain a genetic diagnosis.

Article Synopsis
  • Chronic subdural hematoma (CSDH) treatment often involves burr-hole drainage and either subdural or subgaleal drain insertion, with surgeon preference influencing the choice.
  • A study analyzed 700 CSDH cases from 2017 to 2022, finding no significant differences in recurrence, reoperation rates, or complications between subdural and subgaleal drains.
  • The results suggest that both drainage methods are equally effective, indicating a need for a larger, multicenter randomized controlled trial to further explore their outcomes.

Introduction: To the best of our knowledge, clinically significant endogenous ethylene glycol production has never been reported in humans, very seldom reported in other animals or microorganisms, and then only under rare and specific conditions. We describe the detailed investigations we undertook in two adult monozygotic twin sisters to ascertain whether they were producing endogenous ethylene glycol.

Methods: Two previously healthy monozygotic adult twin sisters presented with recurrent episodes of apparent ethylene glycol poisoning beginning at age 35, requiring chronic hemodialysis to remove ethylene glycol and its metabolites as well as to restore metabolic homeostasis.
