Is host heparanase required for the rapid spread of heparan sulfate binding viruses?

Virology

March 2019

Cancer & Vascular Biology Group, ACRF Department of Cancer Biology and Therapeutics, The John Curtin School of Medical Research, The Australian National University, Canberra, Australia.

Vaccinia virus (VACV), like many other viruses, binds to cell surface heparan sulfate (HS) prior to infecting cells. Since HS is ubiquitously expressed extracellularly, it seemed likely that VACV-HS interaction may impede virus spread, with host heparanase, the only known mammalian endoglycosidase that can degrade HS, potentially overcoming this problem. In support of this hypothesis, we found that, compared to wild type, mice deficient in heparanase showed a 1-3 days delay in the spread of VACV to distant organs, such as ovaries, following intranasal inoculation, or to ovaries and spleen following intramuscular inoculation.

Background: Control of lymphatic filariasis (LF) in most of the sub-Saharan African countries is based on annual mass drug administration (MDA) using a combination of ivermectin and albendazole. Monitoring the impact of this intervention is crucial for measuring the success of the LF elimination programmes. This study assessed the status of LF infection in Rufiji district, southeastern Tanzania after twelve rounds of MDA.

Glycophorin A and glycophorin B are red blood cell surface proteins and are both receptors for the parasite Plasmodium falciparum, which is the principal cause of malaria in sub-Saharan Africa. DUP4 is a complex structural genomic variant that carries extra copies of a glycophorin A-glycophorin B fusion gene and has a dramatic effect on malaria risk by reducing the risk of severe malaria by up to 40%. Using fiber-FISH and Illumina sequencing, we validate the structural arrangement of the glycophorin locus in the DUP4 variant and reveal somatic variation in copy number of the glycophorin B-glycophorin A fusion gene.

Introduction: Liver cirrhosis affects health-related quality of life (HRQoL) even in its early stages. Morbidity is especially high when the disease decompensates and self-care actions become essential. Nurse involvement in secondary prevention in other chronic diseases has contributed to better symptom control, less need of inpatient care and improved HRQoL.

Pion-Pole Contribution to Hadronic Light-By-Light Scattering in the Anomalous Magnetic Moment of the Muon.

Phys Rev Lett

September 2018

Helmholtz-Institut für Strahlen- und Kernphysik (Theory) and Bethe Center for Theoretical Physics, University of Bonn, 53115 Bonn, Germany.

The π^{0} pole constitutes the lowest-lying singularity of the hadronic light-by-light (HLBL) tensor, and thus, it provides the leading contribution in a dispersive approach to HLBL scattering in the anomalous magnetic moment of the muon (g-2)_{μ}. It is unambiguously defined in terms of the doubly virtual pion transition form factor, which in principle, can be accessed in its entirety by experiment. We demonstrate that, in the absence of a direct measurement, the full spacelike doubly virtual form factor can be reconstructed very accurately based on existing data for e^{+}e^{-}→3π, e^{+}e^{-}→e^{+}e^{-}π^{0}, and the π^{0}→γγ decay width.

Article Synopsis
  • High blood pressure is a significant risk factor for cardiovascular disease and is influenced by genetics, but it can be modified through lifestyle changes.
  • This research is the largest genetic study on blood pressure, involving over 1 million individuals of European descent, identifying 535 new genetic loci related to blood pressure traits like systolic and diastolic pressure.
  • The study reveals new biological pathways for regulating blood pressure, which could lead to better prevention strategies for cardiovascular disease in the future.

Swedish asthma guidelines recommend treating asthma with the lowest effective dose possible. These recommendations are both hard to follow and seldom considered enough in asthma care today. As an example, combination therapy with inhaled corticosteroids (ICS) and long acting beta agonists (LABA) should only be considered if asthma control is not achieved with low to medium dose ICS daily.

In pursuit of developing fast and accurate species-level molecular identification methods, we tested six DNA barcodes, namely ITS2, KLa, ITS2+K, ITS2+La, K+La and ITS2+K+La, for their capacity to identify frequently consumed but geographically isolated medicinal species of Fabaceae and Poaceae indigenous to the desert of Cholistan. Data were analysed by BLASTn sequence similarity, pairwise sequence divergence in TAXONDNA, and phylogenetic (neighbour-joining and maximum-likelihood trees) methods. Comparison of six barcode regions showed that ITS2 has the highest number of variable sites (209/360) for tested Fabaceae and (106/365) Poaceae species, the highest species-level identification (40%) in BLASTn procedure, distinct DNA barcoding gap, 100% correct species identification in BM and BCM functions of TAXONDNA, and clear cladding pattern with high nodal support in phylogenetic trees in both families.

The semi-arid Amboseli landscape, southern Kenya, is characterised by intermittent groundwater-fed wetlands that form sedimentary geoarchives recording past ecosystem changes. We present a 5000-year environmental history of a radiocarbon dated sediment core from Esambu Swamp adjacent to Amboseli National Park. Although radiocarbon dates suggest an unconformity or sedimentary gap that spans between 3800 and 500 cal year BP, the record provides a unique insight into the long-term ecosystem history and wetland processes, particularly the past 500 years.

Despite great interest and decades of research, the musculoskeletal relationships of the masticatory system in primates are still not fully understood. However, without a clear understanding of the interplay between muscles and bones it remains difficult to understand the functional significance of morphological traits of the skeleton. Here, we aim to study the impacts of the masticatory muscles on the shape of the cranium and the mandible as well as their co-variation in strepsirrhine primates.

We introduce multi-trait analysis of GWAS (MTAG), a method for joint analysis of summary statistics from genome-wide association studies (GWAS) of different traits, possibly from overlapping samples. We apply MTAG to summary statistics for depressive symptoms (N  = 354,862), neuroticism (N = 168,105), and subjective well-being (N = 388,538). As compared to the 32, 9, and 13 genome-wide significant loci identified in the single-trait GWAS (most of which are themselves novel), MTAG increases the number of associated loci to 64, 37, and 49, respectively.

Electronic stopping of slow protons in ZnO, VO_{2} (metal and semiconductor phases), HfO_{2}, and Ta_{2}O_{5} was investigated experimentally. As a comparison of the resulting stopping cross sections (SCS) to data for Al_{2}O_{3} and SiO_{2} reveals, electronic stopping of slow protons does not correlate with electronic properties of the specific material such as band gap energies. Instead, the oxygen 2p states are decisive, as corroborated by density functional theory calculations of the electronic densities of states.

High-elevation ecosystems, such as those on Mount Kenya are undergoing significant changes, with accelerated glacial ice losses over the twentieth century creating new space for alpine plants to establish. These ecosystems respond rapidly to climatic variability and within decades of glacial retreat, Afroalpine pioneering taxa stabilize barren land and facilitate soil development, promoting complex patches of alpine vegetation. Periglacial lake sediment records can be used to examine centennial and millennial scale variations in alpine and montane vegetation compositions.
