The identification of low-frequency variants remains challenging due to the inevitable high error rates of next-generation sequencing (NGS). Numerous promising strategies employ unique molecular identifiers (UMIs) for error suppression. However, their efficiency depends highly on redundant sequencing and quality control, leading to tremendous read waste and cost inefficiency.

This study presents a comprehensive framework to enhance Wikidata as an open and collaborative knowledge graph by integrating Open Biological and Biomedical Ontologies (OBO) and Medical Subject Headings (MeSH) keywords from PubMed publications. The primary data sources include OBO ontologies and MeSH keywords, which were collected and classified using SPARQL queries for RDF knowledge graphs. The semantic alignment between OBO ontologies and Wikidata was evaluated, revealing significant gaps and distorted representations that necessitate both automated and manual interventions for improvement.

  • The TORCHLIGHT trial tested the combination of the immune-checkpoint inhibitor toripalimab and nab-paclitaxel (nab-P) against a placebo with nab-P in patients with advanced triple-negative breast cancer (TNBC).
  • Results showed that the toripalimab group had a significantly longer progression-free survival (PFS) of 8.4 months compared to 5.6 months for the placebo group, particularly in PD-L1-positive patients.
  • The overall survival was also better for the toripalimab group (32.8 months vs. 19.5 months), and the safety profile was similar between groups, suggesting that toripalimab is a promising treatment for this cancer type.

Introduction: Research has shown an association between increased disclosure of HIV status by pregnant and breastfeeding women and improved clinical health and that of their infant. Increasing awareness about their male partner's HIV status will no doubt lead to even better outcomes at the population level. Male partner involvement is important for improving outcomes of prevention of mother-to-child transmission of HIV (MTCT) as it improves social support and commitment from both parents of the baby to ensure sustained good health.
