Introduction: To analyze the molecular pathogenesis of severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2), a small animal model such as mice is needed: human angiotensin converting enzyme 2 (hACE2), the receptor of SARS-CoV-2, needs to be expressed in the respiratory tract of mice.
Methods: We conferred SARS-CoV-2 susceptibility in mice by using an adenoviral vector expressing hACE2 driven by an elongation factor 1α (EF1α) promoter with a leftward orientation.
Results: In this model, severe pneumonia like human COVID-19 was observed in SARS-CoV-2-infected mice, which was confirmed by dramatic infiltration of inflammatory cells in the lung with efficient viral replication.

In this study, we employed platinum-assisted surface-assisted laser desorption/ionization mass spectrometry imaging (MSI) (Pt-SALDI-MSI) to detect and visualize the spatial distribution of antioxidant additives and organic dyes in polystyrene films undergoing photodegradation. In traditional matrix-assisted laser desorption/ionization mass spectrometry (MALDI-MS), matrix-derived ion peaks often obscure signals from low-molecular-weight analytes. Pt-SALDI-MSI, which utilizes inorganic nanoparticles instead of an organic matrix, enables the interference-free analysis of low-molecular-weight compounds, thereby addressing the limitation of traditional MALDI-MS.

Background: Adult spinal reconstructive surgery that requires multilevel spinal fusion is highly invasive and requires two-stage surgery using lateral lumbar interbody fusion (LLIF) and/or percutaneous pedicle screw (PPS) fixation to make it less invasive. However, it is still difficult to make spinal osteotomy less invasive, and the high complication rate is an issue.
Observations: The authors present the surgical techniques of a two-stage Schwab grade 4 spinal osteotomy using LLIF, which could reduce surgical invasiveness and enable good correction and anterior spinal column reconstruction for lumbar kyphosis, and also report a case treated with this procedure.

Introduction: Limited research has extensively analyzed neurodegenerative disease-related protein deposition patterns in the hippocampus.
Methods: This study examined the distribution of proteins in hippocampal subregions across major neurodegenerative diseases and explored their relation to each other. The area density of phosphorylated tau (p-tau), amyloid beta (Aβ), α-synuclein, and phosphorylated TDP-43 protein deposits together with pyramidal cell density in each hippocampal subregion, including CA1-4, prosubiculum (ProS), and subiculum was assessed in 166 cases encompassing various neurodegenerative diseases.

Piceatannol, a stilbene compound, undergoes a comprehensive phase II metabolism mediated by UDP-glucuronosyltransferases (UGTs) and sulfotransferases (SULTs) in humans. Despite their well-documented beneficial effects on health, their detailed pharmacokinetic fate, including the metabolite structure and properties, is poorly understood. Thus, we determined the structure of seven glucuronides and six sulfates transformed from piceatannol and its methylated derivatives in recombinant yeast cells expressing UGTs or SULTs.

Background: To evaluate whether sodium zirconium cyclosilicate (SZC) enables the up-titration of spironolactone without increasing the risk of hyper- and hypokalemia in patients with heart failure with reduced and mildly reduced ejection fraction (HFrEF and HFmrEF) and moderate/severe chronic kidney disease (CKD) who developed hyperkalemia during treatment with suboptimal spironolactone dose.
Methods: The REGISTA-K is a randomized, double-blind, placebo-controlled, multicenter trial that examined the efficacy and safety of SZC in up-titrating spironolactone without the occurrence of hyperkalemia or hypokalemia. A total of 266 patients with HFrEF and HFmrEF and hyperkalemia will be randomized in a 1:1 ratio to receive either SZC or placebo after treating hyperkalemia with SZC at 25 sites in Japan.

Inhibition of xanthine oxidoreductase (XOR) was shown to ameliorate the stroke susceptibility in the stroke-prone spontaneously hypertensive rat (SHRSP), suggesting hyperuricemia had a pathological role in this rat model. In this study, we thus aimed to explore mechanisms inducing hyperuricemia in SHRSP. XOR is known to have two forms, xanthine dehydrogenase (XDH) as the prototype and xanthine oxidase (XO) as the converted form through cleavage of a peptide bond or through formation of disulfide bonds in the enzyme.

Most members of the genus Orthonairovirus, represented by Crimean-Congo hemorrhagic fever virus and Nairobi sheep disease virus, are tick-borne, and some have become a public health concern in recent years. Here, we report the isolation and genetic and biological characterization of a new orthonairovirus, designated as "Iwanai Valley virus" (IWVV), from Ixodes ovatus ticks in Hokkaido, Japan. The amino acid sequence of the viral nucleoprotein (NP) was found to be 34-45% identical to those of known orthonairoviruses.

Introduction: Osimertinib, a third-generation epidermal growth factor receptor (EGFR) tyrosine kinase inhibitor, is a first-line therapy for advanced or metastatic non-small cell lung cancer (NSCLC) with EGFR mutations, including both sensitizing and T790M resistance mutations. Its real-world efficacy against uncommon EGFR mutations remains under-researched.
Methods: The REIWA study, a multicentric, prospective, observational study conducted in Japan from September 2018 to August 2020, enrolled patients with advanced or recurrent EGFR mutation-positive NSCLC receiving osimertinib.

Asialo-GM1 (ASGM1) has been identified as a cell surface marker of murine NK cells. Although polyclonal anti-asialo-GM1 antibodies (anti-ASGM1 pAb) have been widely used for studying natural killer (NK) cell functions in vivo, the technical challenges have existed in their specificity for NK cell depletion. Furthermore, the exact expression of ASGM1 on the NK cell lineage and other immune cells has not been characterized due to the lack of appropriate reagents.
