Upper Permian rocks of the former supercontinent Gondwana record climax late Paleozoic terrestrial vertebrate faunas that were dominated numerically and ecologically by therapsid synapsids. Older faunal elements of earlier Paleozoic faunas, such as captorhinid reptiles, are rare and scattered components of the first amniote faunas to inhabit high-latitude regions. Here we describe a new genus and species of moradisaurine captorhinid that represents an archaic faunal element of the high-fibre herbivore fauna of the late Permian of what is now peninsular India.

Desiccation is a fundamental challenge confronted by all terrestrial organisms, particularly insects. With a relatively small body size and large surface-to-volume ratio, insects are susceptible to rapid evaporative water loss and dehydration. To counter these physical constraints, insects have acquired specialized adaptations, including a hydrophobic cuticle that acts as a physical barrier to transpiration.

Modern-day terrestrial amphibians pale in comparison to their monstrous ancient relatives, the Late Carboniferous and Early Permian trematopid temnospondyls. With a skeleton that clearly indicated a terrestrial mode of life and armed with an impressive set of large, recurved marginal dentition and palatal fangs for holding their prey-this group of terrestrial temnospondyls roamed North America and Central Europe as a top predator. Lack of substantial informative fossil material has previously limited our understanding of trematopid diversity and ontogeny.

Here we describe a design strategy that allows the construction of proteomimetics against challenging targets without a priori characterization of the target surface. Fragment screening is a popular strategy of generating viable chemical starting points especially for challenging targets. Although fragments provide a better coverage of chemical space and they have typically higher chance of binding, their weak affinity necessitates highly sensitive biophysical assays. Here, we introduce a screening concept that combines evolutionary optimized fragment pharmacophores with the use of a photoaffinity handle that enables high hit rates by LC-MS-based detection.

Multiplexed assays of variant effect (MAVEs) have emerged as a powerful approach for interrogating thousands of genetic variants in a single experiment. The flexibility and widespread adoption of these techniques across diverse disciplines have led to a heterogeneous mix of data formats and descriptions, which complicates the downstream use of the resulting datasets. To address these issues and promote reproducibility and reuse of MAVE data, we define a set of minimum information standards for MAVE data and metadata and outline a controlled vocabulary aligned with established biomedical ontologies for describing these experimental designs.

Focal recording is an extracellular method for studying synaptic transmission at the larval neuromuscular junction (NMJ) designed for the study of synaptic activity of one or a few synaptic boutons rather than the ensemble activity of all the boutons as occurs with intracellular recording or two-electrode voltage-clamp. This is a useful technique for investigating the properties of different motor neurons that innervate the same muscle, applying statistical analysis to discrete synaptic events, and investigating the heterogeneity of synaptic release properties among boutons. A compound microscope with epifluorescent imaging capability is very helpful but not essential; any GFP strain that labels the nerve terminal or synaptic boutons can be used to locate the boutons.

This preparation has become the "gold standard" for studying neuronal excitability as well as synaptic transmission. Although it is particularly valuable in revealing membrane potential changes, intracellular recording has a number of limitations. Primarily, it does not offer information on the kinetics of membrane currents associated with ion channels or synaptic receptors responsible for the potential change. Furthermore, the resting potential of the body wall muscle varies naturally such that the driving force also varies considerably, making it difficult to accurately compare the amplitude of miniature synaptic potentials (minis) or evoked excitatory junction potentials (EJPs).

While urban trees can be important determinants of human health and wellbeing in world cities, the specific influence of nearby urban trees upon human wellbeing has not been adequately explored. While many studies have associated urban greenery abundance with wellbeing scores, many measures of urban greenery do not specify the type of vegetation or the impact of co-location. Here we fill this gap by associating self-reported measures of the presence of nearby trees (tree in front of one's home) with validated subjective wellbeing (SWB) scores.

Relational boredom is an important cognitive-emotional experience that is understudied in the relationship maintenance literature. In three dyadic studies, we investigated accuracy and bias in partners' perceptions of each other's relational boredom, and how accurate and biased boredom perceptions were associated with relationship quality. Results revealed that, overall, partners tended to overestimate-but accurately track-each other's relational boredom across the features that comprise relational boredom and across time.

Human leukocyte antigen (HLA)-G is an immune checkpoint molecule that is highly expressed in papillary thyroid carcinoma (PTC). The gene presents several functional polymorphisms distributed across the coding and regulatory regions (5'URR: 5' upstream regulatory region and 3'UTR: 3' untranslated region) and some of them may impact HLA-G expression and human malignancy.
