Alzheimers Dement

December 2024

Federal University of Rio Grande do Sul, Porto Alegre, Rio Grande do Sul, Brazil.

Background: Alzheimer's disease (AD) is a progressive neurodegenerative disease and the most common form of dementia. Although AD is characterized by the accumulation of amyloid beta (Aβ) plaques and neurofibrillary tangles (NFTs), it's estimated that nearly half of AD cases might be attributed to modifiable risk factors and lifestyle-based interventions may offer promising preventative strategies to delay disease onset and progression. Polyphenolic derivatives easily found in foods like luteolin and curcumin have shown beneficial effects to counteract cognitive decline.

Alzheimers Dement

December 2024

Federal University of Rio Grande do Sul, Porto Alegre, Rio Grande do Sul, Brazil.

Background: Alzheimer's disease (AD) is a progressive and multifactorial neurodegenerative disease that still has no cure. Different pathological processes contribute to the disease's development, such as the presence of amyloid beta (Aβ) plaques, neurofibrillary tangles (NFTs), glutamatergic excitotoxicity, oxidative stress, and neuroinflammation. Chalcones are polyphenolic compounds of natural origin with a wide range of biological activities, and emerging studies have reported neurotrophic activity, anti-inflammatory and antioxidant effects, and the inhibition of Aβ aggregation.

Alzheimer's Disease Neuroimaging Initiative, http://adni.loni.usc.edu/, CA, USA.

Background: Several studies have shown that financial capacity constitutes a vital component of instrumental activities of daily living. However, there is insufficient research investigating the relationship between financial impairment, brain volume changes and cognitive decline in Alzheimer's disease (AD). Here, we examine the association between brain volume changes and financial capacity in cognitively unimpaired (CU) and mild cognitively impaired (MCI) individuals.

Alzheimers Dement

December 2024

Alzheimer's Disease Neuroimaging Initiative, http://adni.loni.usc.edu/, CA, USA.

Background: Amyloid and tau pathologies are the hallmarks of Alzheimer's disease (AD). Previous research indicated notable connections between financial capacity and AD biomarkers. Here, we aimed to understand whether financial capacity is affected by the cerebral accumulation of tau and amyloid.

Background: Anti-amyloid therapy appears to have an increased effect on reducing cognitive decline in amyloid- and tau-positive individuals. However, clinical trials inclusion criteria require solely amyloid positivity. Herein, we developed a machine-learning prediction model to identify tau positivity in amyloid-positive individuals using clinical variables.

Background: Screen failure due to amyloid negativity is yet a problem in clinical trials for anti-amyloid drugs. In this context, clinical characteristics of patients presenting with cognitive decline may decrease the screen failure ratio by increasing the odds of selecting individuals with brain amyloid pathology. Herein, we aimed at estimating amyloid and tau positivity in individuals using clinical variables in a machine learning model of prediction.

Indigenous university students' perceptions regarding nature, their daily lives and climate change: a photovoice study.

BMC Public Health

January 2025

Department of Population Health Sciences, School of Life Course & Population Sciences, King's College London, Franklin-Wilkins Building, Stamford Street London, SE1 9NH, UK.

Background: Climate change has severe health impacts, particularly for populations living in environmentally sensitive areas such as riversides, slopes, and forests. These challenges are exacerbated for Indigenous communities, who often face marginalisation and rely heavily on the land for their livelihoods. Despite their vulnerability, the perspectives of Indigenous populations on climate change and its impacts remain underexplored, creating a critical gap in the literature.

Background: Optimised use of kidney function information might improve cardiac risk prediction in noncardiac surgery.

Methods: In 35,815 patients from the VISION cohort study and 9219 patients from the POISE-2 trial who were ≥45 yr old and underwent nonurgent inpatient noncardiac surgery, we examined (by age and sex) the association between continuous nonlinear preoperative estimated glomerular filtration rate (eGFR) and the composite of myocardial injury after noncardiac surgery, nonfatal cardiac arrest, or death owing to a cardiac cause within 30 days after surgery. We estimated contributions of predictive information, C-statistic, and net benefit from eGFR and other common patient and surgical characteristics to large multivariable models.

  • Major depressive disorder (MDD) is linked to neuroinflammatory processes involving the NLRP3 inflammasome, and a study examined the impact of the NLRP3 rs10754558 polymorphism on MDD diagnoses among young adults.
  • A population-based study with 1,100 participants found no significant association between the NLRP3 genotype and MDD diagnosis, but a clinical trial showed that individuals with the GG genotype had poorer treatment outcomes compared to those with GC/CC genotypes.
  • Longitudinal analyses indicated that GG individuals experienced less improvement in depressive and anxiety symptoms over time, suggesting that this genotype may affect the effectiveness of psychotherapy for MDD.

Study of the Anti-MYC Potential of Lanostane-Type Triterpenes.

ACS Omega

December 2024

Department of Organic and Inorganic Chemistry, Science Center, Federal University of Ceará, Fortaleza, CE 60455-760, Brazil.

One of the most investigated molecular targets for anticancer therapy is the proto-oncogene , which is amplified and thus overexpressed in many types of cancer. Due to its structural characteristics, developing inhibitors for the target has proven to be challenging. In this study, the anti-MYC potential of lanostane-type triterpenes was investigated for the first time, using computational approaches that involved ensemble docking, prediction of structural properties and pharmacokinetic parameters, molecular dynamics (MD), and binding energy calculation using the molecular mechanics-generalized born surface area (MM-GBSA) method.
