Treatment of non-small cell lung cancer (NSCLC) has drastically changed in recent years owing to the robust anticancer effects of immune checkpoint inhibitors (ICI). However, only 20% of the patients with NSCLC benefit from ICIs, highlighting the need to uncover the mechanisms mediating resistance. By analyzing the overall survival (OS) and mutational profiles of 424 patients with NSCLC who received ICI treatments between 2015 and 2021, we determined that patients carrying a loss-of-function mutation in neurotrophic tyrosine kinase receptor 1 (NTRK1) had a prolonged OS when compared with patients with wild-type NTRK1.

PD-1 Targeted Antibody Discovery Using AI Protein Diffusion.

Technol Cancer Res Treat

November 2024

Tuple LLC, Charlotte, NC, USA.

The programmed cell death protein 1 (PD-1, CD279) is an important therapeutic target in many oncological diseases. This checkpoint protein inhibits T lymphocytes from attacking other cells in the body and thus blocking it improves the clearance of tumor cells by the immune system. While there are already multiple FDA-approved anti-PD-1 antibodies, including nivolumab ( from Bristol-Myers Squibb) and pembrolizumab ( from Merck), there are ongoing efforts to discover new and improved checkpoint inhibitor therapeutics.

Unified modeling of photothermal and photochemical damage.

Front Ophthalmol (Lausanne)

August 2024

Department of Mathematics and Statistics, University of North Carolina at Charlotte, Charlotte, NC, United States.

Correlating damage outcomes to a retinal laser exposure is critical for diagnosis and choosing appropriate treatment modalities. Therefore, it is important to understand the causal relationships between laser parameters, such as wavelength, power density, and length of exposure, and any resulting injury. Differentiating photothermal from photochemical processes in an retinal model using cultured retinal pigment epithelial cells would be a first step in achieving this goal.

Genomic factors shaping codon usage across the Saccharomycotina subphylum.

G3 (Bethesda)

November 2024

Department of Bioinformatics and Genomics, University of North Carolina at Charlotte, North Carolina Research Campus, Kannapolis, NC 28081, USA.

Codon usage bias, or the unequal use of synonymous codons, is observed across genes, genomes, and between species. It has been implicated in many cellular functions, such as translation dynamics and transcript stability, but can also be shaped by neutral forces. We characterized codon usage across 1,154 strains from 1,051 species from the fungal subphylum Saccharomycotina to gain insight into the biases, molecular mechanisms, evolution, and genomic features contributing to codon usage patterns.
