Introduction: Oral anticoagulants (OAC) reduce stroke/systemic embolism and mortality risks in atrial fibrillation (AF). However, there is an inherent bleeding risk with OAC, where intracranial hemorrhage (ICH) is the most feared, disabling, and lethal complication of this therapy. Therefore, the optimal management of OAC-associated ICH is not well defined despite multiple suggested strategies.

Article Synopsis
  • The inflammatory immune response can contribute to the development of colorectal cancer (CRC) if not properly regulated.
  • A study using transcriptomics analysis found a significant connection between inflammation and the expression of granzyme A (GzmA) in human CRC.
  • In mouse models, inhibiting GzmA reduced gut inflammation and prevented CRC, highlighting its role in promoting cancer through mechanisms involving NF-κB and STAT3 activation, suggesting GzmA could be a potential therapeutic target for CRC.

Hematopoietic Transplant and Cellular Therapy Unit, Instituto Murciano de Investigación Biosanitaria IMIB-Arrixaca, Virgen de la Arrixaca University Hospital, University of Murcia, Murcia, Spain.

Article Synopsis
  • - Allogeneic hematopoietic stem cell transplantation (allo-HCT) is a key treatment for severe blood cancers and genetic disorders, but acute graft-versus-host disease (aGvHD) is a major complication that increases mortality and limits treatment use.
  • - Current medications for preventing and treating aGvHD have mixed results and serious side effects, indicating a need for better alternatives.
  • - Recent studies suggest that defibrotide may effectively prevent aGvHD by reducing immune cell infiltration and improving survival rates in mouse models, as well as showing promising results in human cell studies by inhibiting key interactions involved in aGvHD.

Background:  The ABC (age, biomarkers, and clinical history)-stroke and ABC-bleeding are biomarker-based scores proposed to predict stroke and bleeding, but non-specificity of biomarkers is common, predicting different clinical events at the same time. We assessed the predictive performance of the ABC-stroke and ABC-bleeding scores, for outcomes beyond ischemic stroke and major bleeding, in a cohort of atrial fibrillation (AF) patients.

Methods:  We included AF patients stable on vitamin K antagonists for 6 months.

Article Synopsis
  • Sepsis triggers a strong inflammatory response followed by a weakened immune system, often leading to metabolic issues and mitochondrial problems, especially in immunocompromised individuals.
  • In patients with intra-abdominal sepsis, their immune cells (monocytes) show reduced activation of the NLRP3 inflammasome when triggered by the P2X7 receptor, correlating with higher mortality rates.
  • The P2X7 receptor is linked to mitochondrial dysfunction in sepsis patients, and its activation can cause further mitochondrial damage, which inhibits NLRP3 inflammasome activation, suggesting a key role of this receptor in sepsis-related complications.
