Human liver models that are three-dimensional (3D) in architecture are indispensable for compound metabolism/toxicity screening, to model liver diseases for drug discovery, and for cell-based therapies; however, further development of such models is needed to maintain high levels of primary human hepatocyte (PHH) functions for weeks to months. Therefore, here we determined how microscale 3D collagen I presentation and fibroblast interaction affect the longevity of PHHs. High-throughput droplet microfluidics was utilized to generate reproducibly sized (∼300-μm diameter) microtissues containing PHHs encapsulated in collagen I ± supportive fibroblasts, namely, 3T3-J2 murine embryonic fibroblasts or primary human hepatic stellate cells (HSCs); self-assembled spheroids and bulk collagen gels (macrogels) containing PHHs served as controls.

Galectin-3, a β-galactosidase binding lectin, known to be involved in inflammatory processes may be associated with abdominal aortic aneurysm (AAA) incidence. We examined the prospective association between plasma galectin-3 and incident AAA in 9,704 participants of the Atherosclerosis Risk in Communities (ARIC) study cohort. We followed participants from 1996-1998 through 2011 (124,260 person-years) for incident AAA (n=325) defined by ICD codes from hospital records and death certificates.

To date, gene-based rare variant testing approaches have focused on aggregating information across sets of variants to maximize statistical power in identifying genes showing significant association with diseases. Beyond identifying genes that are associated with diseases, the identification of causal variant(s) in those genes and estimation of their effect is crucial for planning replication studies and characterizing the genetic architecture of the locus. However, we illustrate that straightforward single-marker association statistics can suffer from substantial bias introduced by conditioning on gene-based test significance, due to the phenomenon often referred to as "winner's curse.

Accelerated data acquisition with simultaneous multi-slice (SMS) imaging for functional MRI studies leads to interacting and opposing effects that influence the sensitivity to blood oxygen level-dependent (BOLD) signal changes. Image signal to noise ratio (SNR) is decreased with higher SMS acceleration factors and shorter repetition times (TR) due to g-factor noise penalties and saturation of longitudinal magnetization. However, the lower image SNR is counteracted by greater statistical power from more samples per unit time and a higher temporal Nyquist frequency that allows for better removal of spurious non-BOLD high frequency signal content.

Predictive processing (PP) is a paradigm in computational and cognitive neuroscience that has recently attracted significant attention across domains, including psychology, robotics, artificial intelligence and philosophy. It is often regarded as a fresh and possibly revolutionary paradigm shift, yet a handful of authors have remarked that aspects of PP seem reminiscent of the work of 18th century philosopher Immanuel Kant. To date there have not been any substantive discussions of how exactly PP links back to Kant.
