Article Synopsis
  • Chronic obstructive pulmonary disease (COPD) traditionally requires a fixed spirometric ratio of FEV/FVC < 0.70 post-bronchodilator, while variable obstruction (VO) is when this ratio improves with treatment but is < 0.70 pre-treatment.
  • A study analyzed data from the SPIROMICS cohort to compare participants with VO to those without obstruction, looking at lung function, health status, and exacerbation rates.
  • Results showed that participants with VO had a significantly higher risk (6.2 times) of developing COPD in the future, prompting a suggestion to revise COPD definitions to include pre-bronchodilator obstruction.

Protein functional effector sncRNAs (pfeRNAs) are approximately 30-60 nucleotides (nt), of which the extraction method from plasma has not yet been reported. Silver staining in a high-resolution polyacrylamide gel suggested that the majority of plasma sncRNAs extracted by some broadly used commercial kits were sncRNAs from 100 nt upwards. Additionally, TRIzol's protocol is for long RNA but not sncRNA recovery.

Plasminogen activator inhibitor type-2 (PAI-2), a member of the serpin family, is dramatically upregulated during pregnancy and in response to inflammation. Although PAI-2 exists in glycosylated and non-glycosylated forms in vivo, the majority of in vitro studies of PAI-2 have exclusively involved the intracellular non-glycosylated form. This study shows that exposure to inflammation-associated hypochlorite induces the oligomerisation of PAI-2 via a mechanism involving dityrosine formation.

Long-term impairment in T cell-mediated adaptive immunity is a major clinical obstacle following treatment of blood disorders with hematopoietic stem cell transplantation. Although T cell development in the thymus has been extensively characterized, there are significant gaps in our understanding of prethymic processes that influence early T cell potential. We have uncovered a Notch/IL-21 signaling axis in bone marrow common lymphoid progenitor (CLP) cells.

DDX41 is needed for pre- and postnatal hematopoietic stem cell differentiation in mice.

Stem Cell Reports

April 2022

Department of Microbiology and Immunology, University of Illinois at Chicago College of Medicine, 835 South Wolcott Avenue, E705 MSB (MC 790), Chicago, IL 60612, USA. Electronic address:

Article Synopsis
  • DDX41 is identified as a crucial tumor suppressor that is often mutated in human blood-related cancers, but its role in blood cell formation was previously unclear.
  • Research using knockout mice showed that DDX41 is essential for the survival and differentiation of hematopoietic stem and progenitor cells, especially for developing myeloid lineage cells.
  • Analysis of gene expression in DDX41-deficient cells revealed significant changes in genes related to blood cell differentiation and highlighted issues with gene splicing, demonstrating DDX41's key role in blood cell development through gene regulation.

Comparison of Using Intraoperative Computed Tomography-Based 3-Dimensional Navigation and Fluoroscopy in Anterior Cervical Diskectomy and Fusion for Cervical Spondylosis.

World Neurosurg

May 2022

Division of Neurosurgery, Southern Illinois University School of Medicine, Springfield, Illinois, USA; Neurological Surgery, Springfield Clinic, Springfield, Illinois, USA.

Objective: Anterior cervical diskectomy and fusion (ACDF) is a highly successful procedure to treat spinal cord or nerve root compression; however, complications can still occur. With advancements in imaging, 3-dimensional (3D) reconstruction allows real-time instrument tracking in a surgical field relative to the patient's anatomy. Here, we compare plate positioning and short-term outcomes when using 3D navigation to fluoroscopy in ACDF for degenerative spine disease.

Dual-specificity tyrosine phosphorylation-regulated kinase 1 A (DYRK1A) is a serine/threonine kinase that belongs to the DYRK family of proteins, a subgroup of the evolutionarily conserved CMGC protein kinase superfamily. Due to its localization on chromosome 21, the biological significance of DYRK1A was initially characterized in the pathogenesis of Down syndrome (DS) and related neurodegenerative diseases. However, increasing evidence has demonstrated a prominent role in cancer through its ability to regulate biologic processes including cell cycle progression, DNA damage repair, transcription, ubiquitination, tyrosine kinase activity, and cancer stem cell maintenance.

Black Babies Matter.

Clin Perinatol

March 2022

Division of Neonatology, Stroger Hospital of Cook County, University of Illinois at Chicago College of Medicine, 1969 Ogden Avenue, Chicago, IL 60612, USA.

Despite dramatic advancements in neonatal intensive care since the 1960s, African-American infants still have more than a two-fold higher first-year mortality rate than non-Latinx White infants. Our essay examines the impact of upstream factors closely linked to the historical and contemporary context of structural racism in the United States on the African-American women's birth outcome disadvantage. In the process, we propose a paradigm to address the racial health inequity in adverse birth outcome by considering the interplay of racism and social class.

Conformation-dependent anti-Aβ monoclonal antibody signatures of disease status and severity in urine of women with preeclampsia.

Pregnancy Hypertens

June 2022

Department of Obstetrics and Gynecology, University of Illinois at Chicago College of Medicine, Chicago, IL 60611, United States; Department of Obstetrics and Gynecology & Reproductive Sciences, Yale University School of Medicine, New Haven, CT 06520, United States. Electronic address:

Prior research has shown that urine of women with preeclampsia (PE) contains amyloid-like aggregates that are congophilic (exhibit affinity for the amyloidophilic dye Congo red) and immunoreactive with A11, a polyclonal serum against prefibrillar β-amyloid oligomers, thereby supporting pathogenic similarity between PE and protein conformational disorders such as Alzheimer's and prion disease. The objective of this study was to interrogate PE urine using monoclonal antibodies with previously characterized A11-like epitopes. Over 100 conformation-dependent monoclonals were screened and three (mA11-09, mA11-89, and mA11-205) selected for further confirmation in 196 urine samples grouped as follows: severe features PE (sPE, n = 114), PE without severe features (mPE, n = 30), chronic hypertension (crHTN, n = 14) and normotensive pregnant control (P-CRL, n = 38).
