DNA-methylation age and accelerated epigenetic aging in blood as a tumor marker for predicting breast cancer susceptibility.

Aging (Albany NY)

December 2024

Department of Molecular, Cell and Developmental Biology, Life Sciences Division, University of California, Los Angeles, CA 90095, USA.

Background: DNA methylation (DNAm)-based marker of aging, referred to as 'epigenetic age' or 'DNAm age' is a highly accurate multi-tissue biomarker for aging, associated with age-related disease risk, including cancer. Breast cancer (BC), an age-associated disease, is associated with older DNAm age and epigenetic age acceleration (age accel) at tissue levels. But this raises a question on the predictability of DNAm age/age accel in BC development, emphasizing the importance of studying DNAm age in pre-diagnostic peripheral blood (PB) in BC etiology and prevention.

  • Ductal carcinoma in-situ (DCIS) is a non-invasive breast cancer type that makes up about 25% of breast cancer cases, but it often leads to unnecessary aggressive treatment despite many cases never progressing to invasive cancer.
  • A study analyzed 197 breast tissue samples to explore molecular changes in DCIS, using techniques like mRNA expression and DNA analysis to compare progressing versus non-progressing cases.
  • The research found significant molecular differences among DCIS subtypes and between DCIS and invasive breast cancer, highlighting the complexity of DCIS and the need for more tailored approaches to assess risk and treatment.

Article Synopsis
  • The study investigates pyroptosis, a form of programmed cell death, in the context of status epilepticus (SE), a condition marked by prolonged seizures, and how it relates to neurodegeneration.
  • Researchers induced SE in mice using pilocarpine and found increased levels of inflammatory markers and TRPM7 expression, identifying TRPM7 as a key player in neuronal damage and hyperexcitability.
  • The findings suggest that targeting TRPM7 and NLRP3 could be a potential treatment strategy for SE, as inhibiting these pathways alleviated neuronal hyperexcitability and pyroptosis.

Genetic evidence against involvement of TRPC proteins in SOCE, ROCE, and CRAC channel function.

Proc Natl Acad Sci U S A

December 2024

Institute of Biomedical Research, School of Biomedical Sciences, Catholic University of Argentina, Buenos Aires C1107AFF, Argentina.

Article Synopsis
  • Researchers used genetically engineered mice and cell lines to study how the depletion of endoplasmic reticulum (ER) calcium stores activates specific calcium entry channels (SOCE) that primarily rely on Orai1 molecules.
  • They discovered that Orai1 is the dominant calcium entry pathway compared to Orai2 and Orai3, and this process does not require functional TRPC molecules, as shown by experiments using cells with inactive TRPC genes.
  • The study also found that even though the TRPC genes were disrupted, both store-depletion-activated calcium entry and receptor-operated calcium entry (ROCE) still relied on Orai1, leading to the establishment of a new strain of mice called TRPC heptaKO mice, which are

Advancement of AI has opened new possibility for accurate diagnosis and prognosis using digital histopathology slides which not only saves hours of expert effort but also makes the estimation more standardized and accurate. However, preserving the AI model performance on the external sites is an extremely challenging problem in the histopathology domain which is primarily due to the difference in data acquisition and/or sampling bias. Although, AI models can also learn spurious correlation, they provide unequal performance across validation population.
