Protein degradation, a major eukaryotic response to cellular signals, is subject to numerous layers of regulation. In yeast, the evolutionarily conserved GID E3 ligase mediates glucose-induced degradation of fructose-1,6-bisphosphatase (Fbp1), malate dehydrogenase (Mdh2), and other gluconeogenic enzymes. "GID" is a collection of E3 ligase complexes; a core scaffold, RING-type catalytic core, and a supramolecular assembly module together with interchangeable substrate receptors select targets for ubiquitylation.

Department of Interventional Therapy, Zhongshan Hospital, University of Fudan, Shanghai 200032, China.

Primary gastric cancer (PGC) is the fourth most common malignant human cancer and the second leading cause of death worldwide. The majority of the subjects of PGC is diagnosed at a late stage, resulting in poor prognosis and therapeutic outcome, largely attributable to dissemination of tumor cells into circulation as circulating tumor cells (CTCs) and their formation of distal tumor. Curcumin is an active ingredient from the rhizome of the plant Curcuma longa.

Herein we report superior dye-adsorption performance for flower-like nanostructure composed of two dimensional (2D) MoS nanosheets by a facile hydrothermal method, more prominent adsorption of cationic dye compared with anodic dye indicates the dye adsorption performance strongly depends on surface charge of MoS nanosheets. The adsorption mechanism of dye is analyzed, the kinetic data of dye adsorption fit well with the pseudo-second-order model, meanwhile adsorption capability at different equilibrium concentrations follows Langmuir model, indicating the favorability and feasibility of dye adsorption. The regenerable property for MoS with full adsorption of dye molecules by using alkaline solution were demonstrated, showing the feasibility of reuse for the MoS, which is promising in its practical water treatment application.

Hallux valgus is a common disorder of the forefoot that results from medial deviation of the first metatarsal and lateral deviation and/or rotation of the great. The management of hallux valgus is usually focused on the malalignment of the first ray, however, some patients report that pain at the plantar aspect of second and/or third metatarsal head(s) is more severe than the first metatarsophalangeal joint. In order to alleviate the metatarsalgia, we developed an oblique sliding osteotomy to manage the second metatarsal.

G-CSF and neuroprotection: a therapeutic perspective in cerebral ischaemia.

Biochem Soc Trans

December 2006

Institute of Neurology, Huashan Hospital, Institute of Brain Science, University of Fudan, 12 Middle Wulumuqi Road, 200040 Shanghai, People's Republic of China.

In several experimental studies of cerebral ischaemia, G-CSF (granulocyte colony-stimulating factor) exerted neuroprotective effects through different mechanisms, including mobilization of haemopoietic stem cells, anti-apoptosis, neuronal differentiation, angiogenesis and anti-inflammation. Hence, G-CSF not only inhibits neuron death, but also generates 'new' neural tissue formation. A small pilot trial reports on the safety and feasibility of G-CSF therapy in stroke patients.

A novel type of PTD, common helix-loop-helix motif, could efficiently mediate protein transduction into mammalian cells.

Biochem Biophys Res Commun

September 2006

State Key Laboratory of Genetic Engineering, School of Life Science, University of Fudan, Shanghai 200433, China.

Protein transduction domains (PTDs), such as HIV TAT PTD, have been widely used as delivery tools into living cells. Here we reported for the first time that the helix-loop-helix (HLH) domain of basic helix-loop-helix (bHLH) family was a novel type of PTD. Efficient internalization has been obtained with HLH domains derived from bHLH proteins, NeuroD/BETA2, Neurogenin3, and Mitf, in various cell types including stable cell lines and primary cells.

There is increasing evidence that neuronal cell death induced by seizures occurs via extrinsic (death receptors) and intrinsic (mitochondria) pathways. Caspase-8 cleaves Bid, which releases cytochrome c, bridging the "extrinsic" and "intrinsic" pathways. Cleavage of Bid may amplify caspase-8-induced neuronal cell death following seizures.

Separation of polyethylene glycols and their amino-substituted derivatives by high-performance gel filtration chromatography at low ionic strength with refractive index detection.

J Chromatogr A

August 2004

Department of Pharmaceutics, Fudan-Pharmco Targeting Drug Research Center, School of Pharmacy, University of Fudan, Shanghai 200032, China.

The chromatographic analysis of polyethylene glycols (PEG) and their amino-substituted derivatives, with an average molecular mass of 2000 and 3350 is described. The PEG derivatives were perfectly separated by TSK-GEL G4000PW(XL) column, which was widely used in high-performance size-exclusion chromatography (SEC), at low ionic strength with refractive index detection. It was shown that ion-exchange interactions were mainly involved in the retention mechanism of these compounds on TSK-GEL G4000PW(XL) column, since retention volume decreased when salt concentration were added to the mobile phase and varied with the pH of the eluent.
