Article Synopsis
  • - Tubo-ovarian high-grade serous carcinoma (HGSC) is a highly lethal form of cancer that often responds to platinum-based chemotherapy due to common issues with DNA damage repair pathways.
  • - Current mechanisms behind platinum resistance in HGSC are complex and not well understood, leading to a lack of effective biomarkers or treatments to improve patient outcomes.
  • - The study uses advanced single-cell mass cytometry to analyze protein responses in HGSC cells, identifying eight specific protein modules linked to carboplatin resistance and sensitivity, which could help in better categorizing and treating drug resistance in patients.

Background: Twice-yearly subcutaneous lenacapavir has been shown to be efficacious for prevention of HIV infection in cisgender women. The efficacy of lenacapavir for preexposure prophylaxis (PrEP) in cisgender men, transgender women, transgender men, and gender-nonbinary persons is unclear.

Methods: In this phase 3, double-blind, randomized, active-controlled trial, we randomly assigned participants in a 2:1 ratio to receive subcutaneous lenacapavir every 26 weeks or daily oral emtricitabine-tenofovir disoproxil fumarate (F/TDF).

Background And Purpose: Prolonged venous transit (PVT+) is a marker of venous outflow; it is defined as the presence or absence of time-to-maximum ≥10 seconds timing in either the superior sagittal sinus or torcula. This novel perfusion imaging-based metric has been associated with higher odds of mortality and lower odds of functional recovery. This study aims to assess the relationship between PVT on admission perfusion imaging and length of hospital stay in large vessel occlusion strokes successfully reperfused with mechanical thrombectomy.

Background: Current asthma guidelines, including those of the European Respiratory Society (ERS) and American Thoracic Society (ATS), suboptimally predict asthma remission, disease severity, and health-care utilisation. We aimed to establish a novel approach to assess asthma severity based on asthma health-care burden data.

Methods: We analysed prospectively collected data from the Severe Asthma Research Program III (SARP III; USA) and the European Unbiased Biomarkers for the Prediction of Respiratory Disease Outcomes (U-BIOPRED; 11 European countries) to calculate a composite burden score based on asthma exacerbations and health-care utilisation, which was modified to include the use of short-acting beta agonists (SABAs) to reflect asthma symptom burden.

: Pretreatment CTA-based Cortical Vein Opacification Score (COVES) has been shown to predict good functional outcomes at 90 days in patients with acute ischemic stroke secondary to large vessel occlusion (AIS-LVO). This is thought to be related to its ability to measure collateral status (CS). However, its association with the reference standard test, the DSA-based American Society of Interventional and Therapeutic Neuroradiology (ASITN) collateral score, has yet to be established.

  • Coagulation factor XI (FXI) inhibitors show potential as new anticoagulants but may worsen heart failure and diastolic dysfunction according to an animal study.
  • In the ARIC study, involving 4,471 older participants, researchers found that lower plasma FXI levels were linked to a higher incidence of heart failure over a median follow-up of 9 years, especially in those aged 75 and older.
  • The study also noted that while lower FXI levels were associated with increased prevalence of diastolic dysfunction, they correlated with a higher likelihood of atrial fibrillation at the baseline, but not with new cases of atrial fibrillation during the follow-up.

  • - Rates of cocaine use disorder (CUD) are increasing among men who have sex with men (MSM), with a lack of effective FDA-approved medication and limited success from psychosocial interventions.
  • - Interviews with 16 MSM participants showed that they were eager to reduce cocaine use and considered medication as a valuable option, facing fewer barriers with pharmacotherapy than with traditional therapies.
  • - The study identified key factors that influence adherence to medication, such as reminders aiding in pill-taking, while stigma and side effects posed challenges, informing future treatment development for CUD in this community.

Despite the abundance of somatic structural variations (SVs) in cancer, the underlying molecular mechanisms of their formation remain unclear. In the present study, we used 6,193 whole-genome sequenced tumors to study the contributions of transcription and DNA replication collisions to genome instability. After deconvoluting robust SV signatures in three independent pan-cancer cohorts, we detected transcription-dependent, replicated-strand bias, the expected footprint of transcription-replication collision (TRC), in large tandem duplications (TDs).

Deep phenotypic profiling of neuroactive drugs in larval zebrafish.

Nat Commun

Institute for Neurodegenerative Diseases, University of California, San Francisco, San Francisco, CA, USA.

Behavioral larval zebrafish screens leverage a high-throughput small molecule discovery format to find neuroactive molecules relevant to mammalian physiology. We screen a library of 650 central nervous system active compounds in high replicate to train deep metric learning models on zebrafish behavioral profiles. The machine learning initially exploited subtle artifacts in the phenotypic screen, necessitating a complete experimental re-run with rigorous physical well-wise randomization.

Frontotemporal dementia (FTD) is one of the leading causes of young-onset dementia before age 65, typically manifesting as abnormal behavior (in behavioral variant FTD) or language impairment (in primary progressive aphasia). Although FTD affects all populations across the globe, knowledge regarding the pathophysiology and genetics derives primarily from studies conducted in North America and Western Europe. Globally, biomedical research for FTD is hindered by variable access to diagnosis, discussed in this group's earlier article, and by reduced access to expertise, funding, and infrastructure.

Prospective cohort studies are useful for studying how biomolecular status affects risk of adverse health outcomes. Less well known is that the longer the follow-up time, the lower the association (or "apparent effect") due to "regression dilution". Here, we evaluate how follow-up interval from baseline to "event" affects the relationship between baseline serum 25-hydroxyvitamin D [25(OH)D] concentration and the later incidence of stroke and major cardiovascular events (MACEs).

polymorphisms (G2019S/N2081D) that increase susceptibility to Parkinson's disease and Crohn's disease (CD) lead to LRRK2 kinase hyperactivity and suppress autophagy. This connection suggests that LRRK2 kinase inhibition, a therapeutic strategy being explored for Parkinson's disease, may also benefit patients with CD. Paneth cell homeostasis is tightly regulated by autophagy, and their dysfunction is a precursor to gut inflammation in CD.

Induction of a distinct macrophage population and protection from lung injury and fibrosis by Notch2 blockade.

Nat Commun

November 2024

Department of Research Biology, Genentech Research and Early Development, South San Francisco, CA, 94080, USA.

Article Synopsis
  • Macrophages are diverse cells that inhabit all body tissues, with specific types residing in organs and additional subtypes recruited during injury.
  • A specific population of recruited macrophages, marked by certain gene expressions, has been linked to fibrosis in various injury and cancer models.
  • Blocking Notch2 increases these macrophages in the lungs, but evidence suggests they actually help reduce fibrosis rather than cause it, highlighting their potential protective role during lung injuries.

This study was designed to assess the quantitative performance of endogenous drug-drug interaction (DDI) biomarkers (N1-methylnicotinamide (1-NMN), N1-methyladenosine (mA), and creatinine) for the organic cation transporters, OCT2 and MATE1/2K in the kidney. Ten healthy volunteers received cimetidine (400 and 800 mg, single dose) or dolutegravir (50 mg, twice a day) together with metformin (500 mg). Cimetidine and dolutegravir were considered to act mainly as MATE1/2K and OCT2 inhibitors, respectively.
