Objective: Neuronal cell death and neuroinflammation are characteristic features of epilepsy, but it remains unclear whether neuronal cell death as such is causative for the development of epileptic seizures. To test this hypothesis, we established a novel mouse line permitting inducible ablation of pyramidal neurons by inserting simian diphtheria toxin (DT) receptor (DTR) cDNA into the Ccl17 locus. The chemokine CCL17 is expressed in pyramidal CA1 neurons in adult mice controlling microglial quiescence.

Controlled environment agriculture is a promising solution to address climate change and resource limitations. Light, the primary energy source driving photosynthesis and regulating plant growth, is critical in optimizing produce quality. However, the impact of specific light spectra during night interruption on improving phytochemical content and produce quality remains underexplored.

Two-dimensional conjugated metal-organic frameworks (2D c-MOFs) are emerging as promising electrode materials for electrochemical energy storage devices. However, a viable path to realize superior dual-ion storage in 2D c-MOFs has remained elusive. Here, we report the synthesis of Cu(N-OHPTP) 2D c-MOFs (x=0,1,2; OHPTP=octahydroxyphenanthrotriphenylene) with precise aromatic carbon-nitrogen arrangements, based on the π-conjugated OHPTP ligand incorporated with one or two nitrogen atoms.

We evaluated Aβ levels in cerebrospinal fluid (Aβ CSF) and Aβ levels in positron emission tomography (Aβ PET) biomarkers in a real-world memory-clinic setting to develop an efficient algorithm for clinical use.
Methods: Patients were evaluated for AD-related Aβ pathology from two independent cohorts (Ludwig Maximilian University [LMU], = 402, and Medical University of Vienna [MUV], = 144).

Tau PET has attracted increasing interest as an imaging biomarker for 4-repeat (4R)-tauopathy progressive supranuclear palsy (PSP). However, the translation of in vitro 4R-tau binding to in vivo tau PET signals is still unclear. Therefore, we performed a translational study using a broad spectrum of advanced methodologies to investigate the sources of [F]PI-2620 tau PET signals in individuals with 4R-tauopathies, including a pilot PET autopsy study in patients.

In Europe, various conservation programs adopted to maintain or restore biodiversity have experienced differing levels of success. However, a synthesis about major factors for success of biodiversity-related conservation programs across ecosystems and national boundaries, such as incentives, subsidies, enforcement, participation, or spatial context, is missing. Using a balanced scorecard survey among experts, we analyzed and compared factors contributing to success or failure of three different conservation programs: two government programs (Natura 2000 and the ecological measures of the Water Framework Directive) and one conservation program of a non-governmental organization (NGO; Rewilding Europe), all focusing on habitat and species conservation.

Macrophages are integral components of the innate immune system that colonize organs early in development and persist into adulthood through self-renewal. Their fate, whether they are replaced by monocytes or retain their embryonic origin, depends on tissue type and integrity. Macrophages are influenced by their environment, a phenomenon referred to as developmental programming.

Drug development for children presents unique challenges and is highly regulated. Novel approaches, such as the use of extrapolation to address, for example, the need to avoid unethical studies, whilst supporting robust evidence generation have been developed in support of benefit/risk considerations by regulatory authorities. This is only one step in the decision-making process towards access, which in Europe also includes health technology assessment (HTA) bodies.

Patients with sepsis differ in their clinical presentations and immune dysregulation in response to infection, but the fundamental processes that determine this heterogeneity remain elusive. Here, we aim to understand which types of immune dysregulation characterize patients with sepsis. To that end, we investigate sepsis pathogenesis in the context of two transcriptional states: one represents the immune response to eliminate pathogens (resistance, R) and the other is associated with systemic inflammation (SI).

Amyloid-PET quantification through the tracer-independent Centiloid (CL) scale has emerged as an essential tool for the accurate measurement of amyloid-β (Aβ) pathology in Alzheimer's disease (AD) patients. The AMYPAD consortium set out to integrate existing literature and recent work from the consortium to provide clinical context-of-use recommendations for the CL scale. Compared to histopathology, visual reads, and cerebrospinal fluid, CL quantification accurately reflects the amount of AD pathology.

Background And Purpose: Endocannabinoids are lipid mediators, which elicit complex biological effects that extend beyond the central nervous system. Tissue concentrations of endocannabinoids increase in atherosclerosis, and for the endocannabinoid N-arachidonoyl-ethanolamine (anandamide, AEA), this has been linked to an anti-inflammatory function. In this study, we set out to determine the anti-inflammatory mechanism of action of AEA, specifically focusing on vascular smooth muscle cells.
