Lancet Psychiatry

January 2025

Developmental Evidence synthesis, Prediction, Implementation lab, Centre for Innovation in Mental Health, Faculty of Environmental and Life Sciences, University of Southampton, Southampton, UK; Hampshire and Isle of Wight NHS Foundation Trust, Southampton, UK; Clinical and Experimental Sciences (CNS and Psychiatry), Faculty of Medicine, University of Southampton, Southampton, UK; Hassenfeld Children's Hospital at NYU Langone, New York University Child Study Center, New York City, NY, USA; DiMePRe-J-Department of Precision and Regenerative Medicine-Jonic Area, University of Bari Aldo Moro, Bari, Italy.

Background: Randomised controlled trials (RCTs) evaluating ADHD medications often use strict eligibility criteria, potentially limiting generalisability to patients in real-world clinical settings. We aimed to identify the proportion of individuals with ADHD who would be ineligible for medication RCTs and evaluate differences in treatment patterns and clinical and functional outcomes between RCT-eligible and RCT-ineligible individuals.

Methods: We used multiple Swedish national registries to identify individuals with ADHD, aged at least 4 years at the age of diagnosis, initiating pharmacological treatment between Jan 1, 2007, and Dec 31, 2019, with follow-up up to Dec 31, 2020.

Background: Mucopolysaccharidosis (MPS) type 1 S and type 2 are rare lysosomal storage disorders characterized by impaired enzyme production, resulting in glycosaminoglycans accumulation within lysosomes. Enzyme Replacement Therapy (ERT) with laronidase and idursulfase are first line treatments, respectively. However, infusion-related hypersensitivity reactions (HR) may lead to ERT discontinuation.

Non-motor fluctuations in Parkinson's disease: underdiagnosed, yet important.

J Mov Disord

December 2024

Department of Basic and Clinical Neuroscience, Institute of Psychiatry, Psychology and Neuroscience, King's College London, London, UK.

Article Synopsis
  • Non-motor fluctuations (NMF) in Parkinson's disease (PD) substantially impact patients' quality of life but remain largely unrecognized and poorly understood despite being identified for over 20 years.
  • NMF, which can overlap with motor fluctuations (MF), are categorized into neuropsychiatric, sensory, and autonomic subtypes, leading to variability in their prevalence and severity due to differences in patient populations and assessment methods.
  • There is a pressing need for high-quality research to better understand the complex nature of NMF, improve diagnostic accuracy, and enhance treatment options for PD patients in clinical settings.

The potential of MAO inhibitors as chemotherapeutics in cancer: A literature survey.

Eur J Med Chem

February 2025

Department of Pharmacy-Pharmaceutical Sciences, University of Bari Aldo Moro, Via E. Orabona 4, 70125, Bari, Italy. Electronic address:

Drug resistance in cancer is determined by genetic mutations and adaptations of tumor cells to drug treatments, raising a challenge in the treatment of cancer. Factors such as prolonged drug exposure, genetic variability among patients, and tumor heterogeneity have been established as contributors to rising incidence of drug resistance, prompting ongoing research into alternative therapies and combination treatments to overcome this challenge. Monoamine oxidases (MAOs), including both isoforms MAO-A and MAO-B, are mitochondrial enzymes responsible for the catabolism of monoamine neurotransmitters such as dopamine, norepinephrine, and serotonin.

Pharmacological management of gambling disorder: A systematic review and network meta-analysis.

Compr Psychiatry

December 2024

Department of Psychiatry, Faculty of Medicine, University of Southampton, UK and Hampshire and Isle of Wight Healthcare NHS Foundation Trust, Southampton, UK.

Article Synopsis
  • Clinical guidelines for medications treating gambling disorder are unclear, prompting a network meta-analysis to compare their efficacy and tolerability.
  • The study reviewed 22 randomized controlled trials, ultimately analyzing 16 with nearly 1,000 participants, focusing on gambling symptom severity and quality of life as efficacy outcomes.
  • Nalmefene showed the most promise in reducing gambling severity and improving quality of life, although it and naltrexone had higher dropout rates due to side effects, indicating lower tolerability compared to placebo.

In the last decade, it has been recognized that tumors do not exist in isolation but interact with surrounding cells, blood vessels, immune cells, and extracellular matrix components. This understanding has shifted the focus from tumor cells alone to the broader context in which they exist, known as tumor microenvironment (TME). The TME is highly heterogeneous, consisting of various cell types, mainly cancer cells, immune cells, and stromal cells.

Mismatch negativity (MMN) is widely considered a candidate diagnostic biomarker for schizophrenia (SCZ). Although blunted MMN responses have been reliably observed in psychosis, the evidence for MMN deficits in other disorders, such as major depressive disorder (MDD), is mixed. This study explores whether MMN alterations in amplitude or latency are unique to SCZ or extend to non-psychotic MDD patients.
