During meiosis, programmed DNA double-strand breaks (DSBs) are formed by the topoisomerase-like enzyme, Spo11, activating the DNA damage response (DDR) kinase Mec1ATR via the checkpoint clamp loader, Rad24RAD17. At single loci, loss of Mec1 and Rad24 activity alters DSB formation and recombination outcome, but their genome-wide roles have not been examined in detail. Here, we utilise two strategies-deletion of the mismatch repair protein, Msh2, and control of meiotic prophase length via regulation of the Ndt80 transcription factor-to help characterise the roles Mec1 and Rad24 play in meiotic recombination by enabling genome-wide mapping of meiotic progeny.

Bacterial swarming is a complex phenomenon in which thousands of self-propelled rod-shaped cells move coherently on surfaces, providing an excellent example of active matter. However, bacterial swarming is different from most studied examples of active systems because single isolated cells do not move, while clusters do. The biophysical aspects underlying this behavior are unclear.

Article Synopsis
  • * It includes data from 413 patients (142 HBV-HDV and 271 HBV monoinfected) and finds that those with HBV-HDV are generally younger and experience a significantly higher incidence of HCC over 5 years compared to HBV-only patients.
  • * The results indicate that HDV is an independent risk factor for the development of HCC in patients with cirrhosis, highlighting its important role regardless of HBV

Purpose: The recently released EANM/SNMMI guideline, endorsed by several important clinical and imaging societies in the field of breast cancer (BC) care (ACR, ESSO, ESTRO, EUSOBI/ESR, EUSOMA), emphasized the role of [F]FDG PET/CT in management of patients with no special type (NST) BC. This review identifies and summarizes similarities, discrepancies and novelties of the EANM/SNMMI guideline compared to NCCN, ESMO and ABC recommendations.

Methods: The EANM/SNMMI guideline was based on a systematic literature search and the AGREE tool.

Relapsed/refractory T-cell acute lymphoblastic leukemia (ALL)/lymphoma (LBL) represent a significant unmet medical need. WU-CART-007 is a CD7-targeting, allogeneic, fratricide-resistant chimeric antigen receptor T cell product generated from healthy donor T cells. WU-CART-007 was evaluated in a phase 1/2 study with a 3 + 3 dose-escalation design followed by cohort expansion in relapsed/refractory T-ALL/LBL.

Unlabelled: With the increasing rate of infections caused by multidrug-resistant organisms (MDRO), selecting appropriate empiric antibiotics has become challenging. We aimed to develop and externally validate a model for predicting the risk of MDRO infections in patients with cirrhosis.

Methods: We included patients with cirrhosis and bacterial infections from two prospective studies: a transcontinental study was used for model development and internal validation (n = 1302), and a study from Argentina and Uruguay was used for external validation (n = 472).

Accumulation and depletion of E. coli in surfaces mediated by curvature.

Phys Rev E

May 2024

Departamento de Física, FCFM, Universidad de Chile, Av. Beauchef 850, 8370458 Santiago, Chile.

Can topography be used to control bacteria accumulation? We address this question in the model system of smooth-swimming and run-and-tumble Escherichia coli swimming near a sinusoidal surface, and show that the accumulation of bacteria is determined by the characteristic curvature of the surface. For low curvatures, cells swim along the surface due to steric alignment and are ejected from the surface when they reach the peak of the sinusoid. Increasing curvature enhances this effect and reduces the density of bacteria in the curved surface.

Prevalence and location of coronary artery disease in anomalous aortic origin of coronary arteries.

Coron Artery Dis

December 2024

Department of Cardiology, Assistance Publique-Hôpitaux de Paris, Hôpital Bichat, Paris.

Article Synopsis
  • The study examines the occurrence and location of coronary artery disease (CAD) in adults with anomalous aortic origin of a coronary artery (AAOCA), which is not well documented.
  • Researchers analyzed data from 390 patients, focusing on the presence of CAD through imaging techniques like coronary angiography.
  • The findings revealed that CAD is more prevalent in the proximal segment of retroaortic AAOCA compared to other types of AAOCA, with no CAD found in the proximal segment of interarterial AAOCA, and the reasons for these differences remain unclear.

Introduction: There is much literature about the role of 2-[F]FDG PET/CT in patients with breast cancer (BC). However, there exists no international guideline with involvement of the nuclear medicine societies about this subject.

Purpose: To provide an organized, international, state-of-the-art, and multidisciplinary guideline, led by experts of two nuclear medicine societies (EANM and SNMMI) and representation of important societies in the field of BC (ACR, ESSO, ESTRO, EUSOBI/ESR, and EUSOMA).

Ecoanthropologie, Centre National de la Recherche Scientifique/Muséum National d'Histoire Naturelle, University Paris Diderot, Sorbonne Paris Cité, Musée de l'Homme, Paris, France.

Lethal intergroup encounters occur in many species because of sexual selection. While documented in mountain gorillas, they are absent in western gorillas as, instead, it is predicted by their higher feeding (frugivory) and mate competition (single-vs. multi-male groups).

Single-molecule characterization of Sen1 translocation properties provides insights into eukaryotic factor-dependent transcription termination.

Nucleic Acids Res

April 2024

Molecular Motors and Machines group, Ecole normale supérieure, Institut de Biologie de l'Ecole normale supérieure (IBENS), CNRS, INSERM, PSL Research University, 75005 Paris, France.

Article Synopsis
  • Sen1 is a key helicase involved in terminating transcription in yeast (Saccharomyces cerevisiae), and this study investigates its motor properties using single-molecule experiments.
  • The research reveals that Sen1 has high translocation rates and processivity on single-stranded DNA, but altering its structural domains affects these properties while also impacting transcription termination efficiency.
  • Overall, the study highlights the importance of Sen1's structural interactions with RNA polymerase II for successful termination of transcription in eukaryotic cells.
