Purpose: Dynamic susceptibility contrast (DSC) MRI is commonly part of the clinical brain tumor imaging protocol. Usually, a preload of contrast agent is administered to minimize contrast-leakage T effects. However, recent studies have indicated that with adaptation of scan parameters (in particular, low flip angle), a preload is not required.

Evaluating and reporting LET and RBE-weighted dose in proton therapy for glioma - The Dutch approach.

Radiother Oncol

January 2025

Department of Radiotherapy, Erasmus MC Cancer Institute, Erasmus University Medical Center, Rotterdam, The Netherlands; HollandPTC, Delft, The Netherlands.

Background And Purpose: With proton therapy, the relative biological effectiveness (RBE) accounts for increased DNA damage caused by higher linear energy transfer (LET) compared to photons. However, the LET and hence the RBE varies along the proton range, particularly at the Bragg peak, introducing challenges in proton treatment planning for brain tumors. The aim of this paper is to standardize evaluating and reporting LET and RBE in proton therapy for patients with grade 2 and 3 IDH mutant gliomas among the Dutch proton therapy centers.

Background: Pre-clinical studies demonstrate that delivering a high dose at a high dose rate result in less toxicity while maintaining tumor control, known as the FLASH effect. In proton therapy, clinical trials have started using 250 MeV transmission beams and more trials are foreseen. A novel aspect of FLASH treatments, compared to conventional radiotherapy, is the importance of dose rate next to dose and geometry.

Background: It remains unclear how COVID-19 has disproportionately affected the mental health of different vulnerable groups. This study explores how mental health inequalities changed between 2014 (pre-COVID-19) and 2021 (during COVID-19) in the Netherlands across intersectional social strata defined by interplays of educational attainment, income level, gender, and age.

Methods: Using 2014 and 2021 self-reported cohort data on health and living conditions of the adult population of Eindhoven and surroundings (N = 1,157), a Multilevel Analysis of Individual Heterogeneity and Discriminatory Accuracy (MAIHDA) was applied to explore intersectional inequalities in mental health in 2014, 2021, and in mental health changes (2014-2021).

Epigenetics of cardiomyopathies: the next frontier.

Heart Fail Rev

January 2025

Thoraxcenter, Department of Cardiology, Cardiovascular Institute. Erasmus MC - University Medical Center Rotterdam, Office RG-431Dr. Molewaterplein 40, 3015 GD, Rotterdam, The Netherlands.

Cardiomyopathies (CMP) are a diverse group of myocardial diseases that cause structural, functional, and pathological changes to the heart. Alterations at the molecular level associated with the clinical phenotype and progression of CMPs cannot be solely explained by the genetic mutations, even in inherited cardiomyopathies. Epigenetics and environmental factors are likely to significantly modify the clinical manifestations of CMPs, resulting in variable clinical expression and different age-related penetrance.

Article Synopsis
  • Risk-reducing salpingo-oophorectomy (RRSO) significantly lowers the risk of high-grade serous carcinoma (HGSC) in women carrying BRCA1/2 genetic variants, but some still develop HGSC after the procedure.
  • A study involving 2,519 women found that the 20-year cumulative incidence of HGSC was low (1.5% for BRCA1 and 0.2% for BRCA2 carriers), with particular risk factors identified, such as older age at RRSO and incomplete embedding of RRSO specimens.
  • Implementing strict guidelines for timely RRSO and ensuring complete specimen embedding could further minimize the risk of developing HGSC in these patients

DNA methylation (DNAm) is the most commonly measured epigenetic mechanism in human populations, with most studies using Illumina arrays to assess DNAm levels. In 2023, Illumina updated their DNAm arrays to the EPIC version 2 (EPICv2), building on prior iterations, namely the EPIC version 1 (EPICv1) and 450K arrays. Whether DNAm measurements are stable across these three generations of arrays has yet not been investigated, limiting the ability of researchers-especially those with longitudinal data-to compare and replicate results across arrays.
