J Cell Mol Med

December 2024

Department of Nephrology, Leicester-Nantong Joint Institute of Kidney Science, Affiliated Hospital of Nantong University, Medical School of Nantong University, Nantong, China.

Article Synopsis
  • Acute kidney injury (AKI) can lead to chronic kidney disease, and this study explores a potential treatment combining erythropoietin-derived helix B surface peptide (HBSP) and caspase-3 small interfering RNA (CASP3siRNA) to prevent kidney damage and fibrosis after AKI.
  • The experiment involved inducing ischemia-reperfusion injury in mice and administering the treatments, which resulted in decreased kidney damage markers and improved kidney cell recovery, highlighting the effectiveness of the co-treatment.
  • The use of HBSP and CASP3siRNA together showed greater protective effects against kidney fibrosis by reducing inflammation and promoting healing during the early stages of recovery.

Superior ophthalmic vein thrombosis (SOVT) is a rare and potentially sight and life-threatening condition. Although broadly classified into septic and aseptic causes, its etiology is widely varied and presents clinically with a range of signs and symptoms, leading to diagnostic and therapeutic challenges. This case series describes three cases of radiologically confirmed SOVT, presenting in close succession at an ophthalmology department in a single center in Trinidad and Tobago.

Introduction: Understanding how RNA viral load changes (viral load kinetics) during acute infection in SARS-CoV-2 can help to identify when and which patients are most infectious. We seek to summarise existing data on the longitudinal RNA viral load kinetics of SARS-CoV-2 sampled from different parts of the respiratory tract (nose, nasopharynx, oropharynx, saliva and exhaled breath) and how this may vary with age, sex, ethnicity, immune status, disease severity, vaccination, treatment and virus variant.

Methods And Analysis: We will conduct a systematic review and meta-analysis, using studies identified through MEDLINE and EMBASE (via Ovid).

Since the first clinical use of ultrasound in the 1940s, significant advancements have been made in its applications. Color Doppler imaging and power Doppler imaging are considered the first and second generations of flow ultrasound assessment tools, respectively. Subsequently, the introduction of contrastenhanced ultrasound has significantly improved the assessment of arterial and venous vascular patterns in lesions and vessels.

Electronic address:

Article Synopsis
  • This review analyzes various mammography datasets used for AI development in breast cancer screening, focusing on their transparency, content, and accessibility.
  • A search identified 254 datasets, with only 28 being accessible; most datasets came from Europe, East Asia, and North America, raising concerns over poor demographic representation.
  • The findings highlight significant gaps in diversity within these datasets, underscoring the need for better documentation and inclusivity to enhance the effectiveness of AI technologies in breast cancer research.

Article Synopsis
  • A 56-year-old man with hypertrophic cardiomyopathy experienced jaundice, eight years after having a dual-chamber ICD implanted, but showed no signs of heart failure.
  • The diagnosis revealed congestive hepatopathy due to severe tricuspid regurgitation caused by the improperly positioned right ventricular lead from the ICD.
  • Treatment involved replacing the tricuspid valve and removing the ICD, leading to a successful recovery, highlighting the need to consider heart issues as a cause of liver dysfunction in similar patients.

Traditionally, patients with asymptomatic, advanced-stage follicular lymphoma were managed with a watchful-waiting approach until disease progression. The 'Watch and Wait' Phase-3 randomised international trial examined whether rituximab could delay the need for treatment and the effect on quality of life (QoL). In this article, we present the long-term results of the QoL aspect of the trial.

Background: Physical rehabilitation may improve health and wellbeing outcomes for some adults living with long COVID. However, individuals living with pre-existing multiple long-term conditions (MLTCs) and long COVID may have additional rehabilitation challenges. This scoping review aims to identify the available evidence describing physical rehabilitation interventions for adults living with long COVID, to systematically map the reporting of pre-existing MLTCs, and to describe the characteristics of physical rehabilitation interventions used in adults with both pre-existing long-term conditions (LTCs) and long COVID.

  • Sodium-glucose co-transporter 2 inhibitors (SGLT2i), particularly dapagliflozin, are believed to improve weight loss, cardiovascular health, and glycaemic control, though their effects on physical function are unclear.
  • The Dapagliflozin, Exercise Training and PhysicAl Function trial will compare the effects of dapagliflozin alone, dapagliflozin with exercise, and diet control on physical function in adults with type 2 diabetes and low physical fitness.
  • The study will analyze various outcomes over 24 weeks, including physical performance, cardiac function, and metabolic health, and has received ethical approval for its research methods.

Article Synopsis
  • The study assessed the effectiveness and safety of Direct Selective Laser Trabeculoplasty (DSLT) in patients with open angle glaucoma and ocular hypertension.
  • In a retrospective analysis of 15 eyes from 10 patients, success was achieved in 73.3% of cases, with a significant reduction in intraocular pressure (IOP) from 22.7 mmHg to 18.7 mmHg within four months.
  • No major complications were reported, and DSLT's safety and effectiveness were found to be comparable to traditional Selective Laser Trabeculoplasty (SLT), suggesting it could be a viable alternative.

There is increasing interest in the use of home-based monitoring in people with chronic lung diseases to improve access to care, support patient self-management, and facilitate the collection of information for clinical care and research. However, integration of home-based monitoring into clinical and research settings requires careful consideration of test performance and other attributes. There is no published guidance from professional respiratory societies to advance the science of home-based monitoring for chronic lung disease.

Sepsis-associated acute kidney injury (AKI) has high morbidity and mortality, but without cause-specific treatment. Erythropoietin derived Helix B surface peptide (HBSP) alleviates AKI, whereas its underlying mechanisms remain to be further explored. Here, the effects of HBSP on pyroptosis, apoptosis, macrophage polarization and repair were investigated in lipopolysaccharide (LPS)-induced AKI mouse model and cultured kidney epithelial cells.

Dominant negative variants in ITPR3 impair T cell Ca2+ dynamics causing combined immunodeficiency.

J Exp Med

January 2025

Molecular and Cellular Immunology, Great Ormond Street Institute of Child Health, University College London, London, UK.

The importance of calcium (Ca2+) as a second messenger in T cell signaling is exemplified by genetic deficiencies of STIM1 and ORAI1, which abolish store-operated Ca2+ entry (SOCE) resulting in combined immunodeficiency (CID). We report five unrelated patients with de novo missense variants in ITPR3, encoding a subunit of the inositol 1,4,5-trisphosphate receptor (IP3R), which forms a Ca2+ channel in the endoplasmic reticulum (ER) membrane responsible for the release of ER Ca2+ required to trigger SOCE, and for Ca2+ transfer to other organelles. The patients presented with CID, abnormal T cell Ca2+ homeostasis, incompletely penetrant ectodermal dysplasia, and multisystem disease.
