  • Recent studies on bronchiolitis show that traditional medical therapies have not proven effective, leading to a focus on nutritional support and oxygen therapy instead.
  • High-flow nasal cannulas (HFNC) have gained popularity despite evidence lacking significant benefits over standard oxygen therapy (SOT), prompting a need for more clinical data.
  • A recent quasi-experimental analysis found that HFNC did not reduce the length of oxygen therapy or hospitalization, suggesting it may not be necessary as a first-line treatment for moderate bronchiolitis.

Progression trajectories from prodromal to overt synucleinopathies: a longitudinal, multicentric brain [F]FDG-PET study.

NPJ Parkinsons Dis

October 2024

Department of Neuroscience, Rehabilitation, Ophthalmology, Genetics, Maternal and Child Health (DINOGMI), University of Genoa, Genoa, Italy.

The phenoconversion trajectory from idiopathic/isolated Rapid eye movement (REM) sleep behavior disorder (iRBD) towards either Parkinson's Disease (PD) or Dementia with Lewy Bodies (DLB) is currently uncertain. We investigated the capability of baseline brain [F]FDG-PET in differentiating between iRBD patients eventually phenoconverting to PD or DLB, by deriving the denovoPDRBD-related pattern (denovoPDRBD-RP) from 32 de novo PD patients; and the denovoDLBRBD-RP from 30 de novo DLB patients, both with evidence of RBD at diagnosis. To explore [F]FDG-PET phenoconversion trajectories prediction power, we applied these two patterns on a group of 115 iRBD patients followed longitudinally.

Article Synopsis
  • Checkpoint immunotherapy struggles against non-immunogenic tumors like neuroblastoma due to low MHC class I expression and neoantigen burden, but inhibiting ERAP1 can enhance anti-tumor immune responses.
  • A novel strategy combining genetic knockout of ERAP1 with the HDAC inhibitor entinostat was developed, leading to increased immunogenicity and making neuroblastoma more responsive to PD-1 therapy.
  • Experimental methods included CRISPR/Cas9 gene editing, flow cytometry, and mass spectrometry to demonstrate that ERAP1 inhibition improves immune cell activity and increases MHC class I and PD-L1 expression in neuroblastoma cells, ultimately slowing tumor growth.
