Fatigue is prevalent in immune-mediated inflammatory and neurodegenerative diseases, yet its assessment relies largely on patient-reported outcomes, which capture perception but not fluctuations over time. Wearable sensors, like inertial measurement units (IMUs), offer a way to monitor daily activities and evaluate functional capacity. This study investigates the relationship between sit-to-stand and stand-to-sit transitions and self-reported physical and mental fatigue in participants with Parkinson's, Huntington's, rheumatoid arthritis, systemic lupus erythematosus, primary Sjögren's syndrome and inflammatory bowel disease.

The state-of-the-art approach to open reduction and fixation (ORIF) of zygoma fracture fragments is based on manual skills. Achieving high accuracy can be challenging. Our feasibility study on deceased body donors with artificial zygomatic fractures investigated whether virtual repositioning of the fractures and the use of customised 3D-printed titanium osteosynthesis plates was similar in accuracy to the conventional manual procedure, and whether the method was applicable in a clinical setting.

Advances in cancer treatments have significantly improved their effectiveness, yet access to first-line therapies remains limited. A 2017 survey revealed that over 25 % of metastatic melanoma patients in Europe lacked access to recommended therapies. To address this, the European Association of Dermato-Oncology and the European Melanoma Registry conducted a follow-up study on the registration and reimbursement of first-line treatments.

Metastasis-directed therapy (MDT) for oligometastatic breast cancer (≤ 5 metastases) has shown little effect in specific scenarios of randomized trials. Therefore, we aimed to assess outcomes after metastasis-directed stereotactic radiotherapy (SRT) in various clinical scenarios. We conducted an international retrospective cohort study in thirteen centers including breast cancer patients receiving SRT to any metastatic site.

γδ T Cell-mediated Tumor Immunity is Tightly Regulated by STING and TGF-β Signaling Pathways.

Adv Sci (Weinh)

November 2024

Guangdong Provincial Key Laboratory of Tumor Interventional Diagnosis and Treatment, Zhuhai Institute of Translational Medicine, Zhuhai People's Hospital Affiliated with Jinan University, Jinan University, Zhuhai, 519000, P. R. China.

The STING pathway plays a critical role in tumor immunosurveillance. However, the precise mechanisms by which STING regulates gamma delta (γδ) T cell function during tumor progression remain unclear. Herein, we find that tumor-derived cyclic GMP-AMP (cGAMP) activates a distinct STING pathway by inducing TBK1-mediated phosphorylation of Eomes in γδ T cells during the early stage of tumor development is demonstrated.

Purpose: The aim of our study was to investigate the technical accuracy of susceptibility-weighted imaging (SWI) and quantitative susceptibility mapping (QSM) created to detect intramammary-like calcifications depending on different TEs, volume, and type of calcification samples at 1.5T.

Methods: Jello-embedded particles of blackboard chalk and ostrich eggshell ranging in size from 4 to 25 mm were used to simulate intramammary calcifications after testing different base substances and calcifications for their suitability to be used in breast phantoms.

Background: Humans are subjected to various environmental stressors (bacteria, viruses, pollution) throughout life. As such, an inherent relationship exists between the effect of these exposures with age. The impact of these environmental stressors can manifest through DNA methylation (DNAm).

Background: Lymphatic disorders such as protein-losing enteropathy, plastic bronchitis, and chylothorax are important complications of the Fontan circulation and ultimately result in an increased risk of early death. Several European centers are now performing lymphatic procedures. The aim of this study is to map the extent of these lymphatic disorders and treatments provided across European pediatric cardiology centers.

Deep learning-based image analysis offers great potential in clinical practice. However, it faces mainly two challenges: scarcity of large-scale annotated clinical data for training and susceptibility to adversarial data in inference. As an example, an artificial intelligence (AI) system could check patient positioning, by segmenting and evaluating relative positions of anatomical structures in medical images.

Interventional neuroradiologists carry out their minimally-invasive procedure by using X-rays within the setup of a Bi-plane Digital Subtraction Angiography. This work provides an immersive virtual reality (VR) environment where the physicians can perform an simulated catheter-based intervention. Since radiation is invisible, the risk of radiation exposure can be enhanced virtually.

The chromosomal translocation t(1;6)(p35.3;p25.2), recurrent in chronic lymphocytic leukaemia, leads to RCC1::IRF4 fusion.

Br J Haematol

December 2024

The Ernest and Helen Scott Haematological Research Institute, Leicester Cancer Research Centre, University of Leicester, Leicester, UK.

Article Synopsis
  • * A study of 10 cases revealed that this translocation connects the interferon regulatory factor 4 (IRF4) gene on chromosome 6 with the regulator of chromosome condensation 1 (RCC1) gene on chromosome 1, resulting in fusion transcripts.
  • * Despite the fusion, the expression levels of RCC1 and IRF4 proteins remained normal, and the cases also displayed typical mutations related to CLL, suggesting a linkage with the IGHV-unmutated subtype.

The mucus serves as a protective barrier in the gastrointestinal tract against microbial attacks. While its role extends beyond merely being a physical barrier, the extent of its active bactericidal properties remains unclear, and the mechanisms regulating these properties are not yet understood. We propose that inflammation induces epithelial cells to secrete antimicrobial peptides, transforming mucus into an active bactericidal agent.

Background: Rigid age limits in the current allocation system for post-mortem donor kidneys in Germany may have problematic effects. The new German national transplantion registry enables data analysis with respect to this question.

Methods: Using anonymized data from the German national transplantion registry, we extracted and evaluated information on the recipients and postmortem donors of kidneys that were allocated in Germany through Eurotransplant over the period 2006-2020.

The surgical management of anterior communicating artery aneurysms (AcomA) is challenging due to their deep midline position and proximity to complex skull base anatomy. This study compares the pterional craniotomy with the interhemispheric approach based on the specific aneurysm angulation. A total of 129 AcomA cases were analyzed, with 50 undergoing microsurgical clipping via either the pterional or interhemispheric approach.

Background: Endovascular treatment (ET) options for acute stroke due to distal middle cerebral artery occlusions are rapidly evolving, but data on outcome and safety are sparse. We therefore performed an analysis of patients undergoing ET for primary M3 occlusions in routine clinical practice in a nationwide registry.

Methods: Patients enrolled between 01/20 and 12/21 in the prospective, multicenter German Stroke Registry-Endovascular Treatment (GSR-ET) were screened for mechanical thrombectomy performed for primary M3 occlusion.

Influence of adjuvant therapies on organ-specific recurrence of cutaneous melanoma: A multicenter study on 1383 patients of the prospective DeCOG registry ADOReg.

Int J Cancer

November 2024

Department of Dermatology, Venereology, and Allergology, University Medical Center and Medical Faculty Mannheim, Heidelberg University, and Center of Excellence in Dermatology, Mannheim, Germany.

This study investigated whether adjuvant treatments in stage III cutaneous melanoma (CM) influenced patterns of recurrence. Patients with primary (n = 1033) or relapsed CM (n = 350) who received adjuvant therapies with Nivolumab (N), Pembrolizumab (P), or Dabrafenib and Trametinib (D + T) were extracted from the prospective multicenter real-world skin cancer registry ADOReg. Endpoints were progression-free survival (PFS), distant metastasis-free survival (DMFS), organ-specific DMFS, and overall survival (OS).
