Intermediate repeat expansions of TBP and STUB1: Genetic modifier or pure digenic inheritance in spinocerebellar ataxias?

Genet Med

February 2023

Sorbonne University, Paris Brain Institute (ICM- Institut du Cerveau), INSERM, CNRS, Assistance Publique-Hôpitaux de Paris (AP-HP), Paris, France. Electronic address:

Purpose: CAG/CAA repeat expansions in TBP are responsible for spinocerebellar ataxia (SCA) type 17 (SCA17). We previously detected cosegregation of STUB1 variants causing SCA48 with intermediate alleles of TBP in 2 families. This cosegregation questions the existence of SCA48 as a monogenic disease.

Dual-energy computed tomography (DECT) has the potential to improve contrast and reduce artifacts and the ability to perform material decomposition in advanced imaging applications. The increased number of measurements results in a higher radiation dose, and it is therefore essential to reduce either the number of projections for each energy or the source x-ray intensity, but this makes tomographic reconstruction more ill-posed.We developed the multi-channel convolutional analysis operator learning (MCAOL) method to exploit common spatial features within attenuation images at different energies and we propose an optimization method which jointly reconstructs the attenuation images at low and high energies with mixed norm regularization on the sparse features obtained by pre-trained convolutional filters through the convolutional analysis operator learning (CAOL) algorithm.

Background: The aim of this work was to investigate the ability of building prognostic models in non-small cell lung cancer (NSCLC) using radiomic features from positron emission tomography and computed tomography with 2-deoxy-2-[fluorine-18]fluoro-d-glucose (F-FDG PET/CT) images based on a "rough" volume of interest (VOI) containing the tumor instead of its accurate delineation, which is a significant time-consuming bottleneck of radiomics analyses.

Methods: A cohort of 138 patients with stage II-III NSCLC treated with radiochemotherapy recruited retrospectively ( = 87) and prospectively ( = 51) was used. Two approaches were compared: firstly, the radiomic features were extracted from the delineated primary tumor volumes in both PET (using the automated fuzzy locally adaptive Bayesian, FLAB) and CT (using a semi-automated approach with 3D Slicer™) components.

Characteristics of Early-Onset vs Late-Onset Colorectal Cancer: A Review.

JAMA Surg

September 2021

Centre for Colorectal Disease, St Vincent's University Hospital, Dublin, Ireland.

Article Synopsis
  • The incidence of early-onset colorectal cancer (diagnosed in individuals under 50) is on the rise globally, with distinct clinical and molecular characteristics compared to late-onset cases.
  • Factors contributing to this increase may include diet, obesity, antibiotic use, and changes in gut microbiome, with most cases occurring sporadically rather than being genetically inherited.
  • There is a need for tailored preventive and therapeutic approaches for early-onset colorectal cancer, as younger patients often present with more severe disease stages but similar outcomes to older patients after treatment.

Depending on the analytical tool applied, the hallmarks of chronic myeloid leukemia (CML) are the Philadelphia Chromosome and the resulting mRNA fusion transcript BCR-ABL1. With an incidence of 1 per 1 million of children this malignancy is very rare in the first 20 years of life. This article aims to; (i) define the disease based on the WHO nomenclature, the appropriate ICD 11 code and to unify the terminology, (ii) delineate features of epidemiology, etiology, and pathophysiology that are shared, but also differing between adult and pediatric patients with CML, (iii) give a short summary on the diseases to be considered as a differential diagnosis of pediatric CML, (iv) to describe the morphological, histopathological and immunophenotypical findings of CML in pediatric patients, (v) illustrate rare but classical complications resulting from rheological problems observed at diagnosis, (vi) list essential and desirable diagnostic criteria, which hopefully in the future will help to unify the attempts when approaching this rare pediatric malignancy.

Chronic myeloid leukemia (CML) in minors is a rare disease which can be effectively treated by tyrosine kinase inhibitors (TKIs) since the year 2000. A majority of pediatricians will encounter one or two CML patients in the course of their careers and will typically have to rely on written information along with their own intuition to provide care. Knowledge of response to TKIs and of age-specific side effects has an impact on the design of pediatric CML trials in many ways aiming to contribute toward greater predictability of clinical improvements.

MPI_AGE is a European Union co-funded research project aimed to use the Multidimensional Prognostic Index (MPI), a validated Comprehensive Geriatric Assessment (CGA)-based prognostic tool, to develop predictive rules that guide clinical and management decisions in older people in different European countries. A series of international studies performed in different settings have shown that the MPI is useful to predict mortality and risk of hospitalization in community-dwelling older subjects at population level. Furthermore, studies performed in older people who underwent a CGA before admission to a nursing home or receiving homecare services showed that the MPI successfully identified groups of persons who could benefit, in terms of reduced mortality, of specific therapies such as statins in diabetes mellitus and coronary artery disease, anticoagulants in atrial fibrillation and antidementia drugs in cognitive decline.
