ESMO Open

October 2023

Laboratory of Clinical and Experimental Pathology, FHU OncoAge, Biobank Côte d'Azur BB-0033-00025, Louis Pasteur Hospital, IRCAN, Université Côte d'Azur, Nice, France.

Article Synopsis
  • Testing for EGFR mutations is crucial for metastatic non-squamous non-small-cell lung cancer, but practical challenges like access to testing and limited tissue samples hinder routine biomarker testing in European labs.
  • A validated online survey conducted among 64 expert labs revealed varying turnaround times and testing practices, with a significant portion only conducting EGFR testing upon clinician request.
  • Despite these issues, testing rates remained stable during the COVID-19 pandemic, but overall there is significant variability in testing methods and quality assurance across different European laboratories.

Validation of the clinical utility of microRNA as noninvasive biomarkers of cardiac allograft rejection: A prospective longitudinal multicenter study.

J Heart Lung Transplant

November 2023

University of Paris, INSERM UMR 970, Paris Translational Research Centre for Organ Transplantation, Paris, France; Kidney Transplant Department, Necker Hospital, Assistance Publique-Hôpitaux de Paris, Paris, France.

Article Synopsis
  • A prospective study was conducted to examine the association between three specific circulating microRNAs (10a, 92a, 155) and cardiac rejection in heart transplant recipients across 11 centers from August 2016 to March 2018.
  • The study involved 461 patients and 831 biopsies, of which 79 indicated rejection; however, the interim analysis found no significant association between the microRNAs and rejection diagnosis.
  • Ultimately, the research concluded that monitoring these microRNAs does not provide clinical utility for heart transplant recipients, leading to the decision to halt further analysis for futility.

Immunoblots remain the gold standard for HIV-1/HIV-2 infection confirmation. However, their ability to differentiate HIV-1 from HIV-2 infection on an antigenically diversified HIV-1 and HIV-2 panel remain uncommon. We performed a multicenter study on 116 serum samples accounting for most of the diversity of HIV-1 (9 different subtypes in group M, 17 circulating recombinant forms (CRFs), and 3 group O) and HIV-2 (groups A and B), evaluating seven confirmatory assays (six commercially available assays and one in-house assay) with genotyping as the reference.

Cátedra de Salud Respiratoria (University of Barcelona), Respiratory Institute (Hospital Clinic Barcelona), IDIBAPS and CIBERES, Barcelona, Spain.

The severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) pandemic has caused severe illness and mortality for millions worldwide. Despite the development, approval and rollout of vaccination programmes globally to prevent infection by SARS-CoV-2 and the development of coronavirus disease 2019 (COVID-19), treatments are still urgently needed to improve outcomes. Early in the pandemic it was observed that patients with pre-existing asthma or COPD were underrepresented among those with COVID-19.

Squamous Cell Carcinoma of the Anal canal (SCCA) is a rare disease associated with a Human Papillomavirus (HPV) infection in most cases, predominantly the HPV16 genotype. About 15% of SCCA are diagnosed in metastatic stage and some will relapse after initial chemoradiotherapy (CRT). Treatment of patients by Docetaxel, Cisplatin and 5-fluorouracil (DCF) has been recently shown to improve their complete remission and progression-free survival.

The Hippo pathway effector YAP is dysregulated in malignant pleural mesothelioma (MPM). YAP's target genes include the secreted growth factor amphiregulin (AREG), which is overexpressed in a wide range of epithelial cancers and plays an elusive role in MPM. We assayed the expression of YAP and AREG in MPM pathology samples and that of AREG additionally in plasma samples of patients from the randomized phase 3 IFCT-0701 Mesothelioma Avastin Cisplatin Pemetrexed Study (MAPS) using immunohistochemistry and ELISA assays, respectively.

Trauma in Rapes and Assaults.

Children (Basel)

December 2021

Centre Adolescents FSEF Henri-Danon-Boileau, University Hospital Bichat, 75018 Paris, France.

Psychological trauma primarily affects children and adolescents; it mostly results from physical and sexual maltreatment. In the Medico-Judicial Unit Center for Sexual Violence Victims in Tours, France, which I joined in 1992 for research and to give treatment, underage patients represented about three-quarters of patients. At the same time, a national survey was conducted in collaboration with Marie Choquet's "Adolescent Health" group (INSERM), which targeted several thousand adolescents representing the general population.

Immunotherapy, the promise for present and future of malignant pleural mesothelioma (MPM) treatment.

Ther Adv Med Oncol

December 2021

Service d'oncologie thoracique and Early Phase Unit CIC1425, Hôpital Bichat-Claude Bernard, 46 rue Henri Huchard, FR-75018 Paris, France.

Due to occupational asbestosis exposure, the incidence of malignant pleural mesothelioma (MPM) has continuously increased over the last 30 years, with a plateau anticipated around the year 2030 in Western countries. Molecular MPM carcinogenesis involves alterations of NF2, RASSF1, LATS2WT1, p16, as well as BAP-1tumor-suppressor genes, which usually regulate apoptosis, cell invasion, motility, cell division, chromatin remodeling, as well as control of DNA repair. In few selected patients, debulking surgery consisting of pleurectomy-decortication is carried out, resulting in unsatisfactory long-term results.
