The multi-dimensional stigma of chronic pain: A narrative review.

Accumulating evidence suggests that stigma is a pervasive and pernicious psychosocial phenomenon that affects people living with chronic pain. In this narrative review, we describe the nature of stigma experienced by people with chronic pain and discuss its multifaceted determinants. These determinants include features of pain itself and intersectional factors, including comorbid conditions and social marginalization.

Optimising water storage for climate resilience: Geospatial targeting for small tanks rejuvenation in Sri Lanka.

In Sri Lanka, climate change and recurrent droughts pose significant threats to agricultural communities. Village water storage tanks have been used in countries in South Asia since the third millennium BC. According to the National Tanks Survey, Sri Lanka has some 23,000 small tanks of 80 ha or less; however, 21 percent are currently non-functional due to decades of neglect.

Generation of a human induced pluripotent stem cell line (CRICKi021-A) from a patient with Ullrich congenital muscular dystrophy carrying a pathogenic mutation in the COL6A1 gene.

Ullrich congenital muscular dystrophy (UCMD) represents the most severe subtype of collagen VI-related dystrophies (COL6-RDs), a spectrum of rare extracellular matrix disorders affecting skeletal muscle and connective tissue. Here, we generated an induced pluripotent stem cell (iPSC) line (CRICKi021-A) from a UCMD patient with de novo dominant-negative mutation in COL6A1 gene by reprogramming dermal fibroblasts using a non-integrating mRNA-based protocol. The resulting human iPSCs displayed normal morphology, expressed pluripotency-associated markers and differentiated into the three germ layers.

Mental health and well-being in adolescent and young adult refugees in Sweden: A cross-sectional study of accompanied and unaccompanied individuals.

Background: Currently, approximately 100 million people are displaced worldwide, including children and young adults. Previous studies showed exposure to violence and posttraumatic stress disorder (PTSD) are common in this sub-population. However, we still lack comprehensive data on well-being, mental health, and the ability to function.

Optimising recovery after caesarean delivery.

Caesarean delivery is the most performed inpatient surgery worldwide, with rates expected to rise. Optimising maternal recovery benefits not only the mother, but also the newborn and society. Enhanced Recovery After Caesarean delivery (ERAC) protocols standardize the approach to perioperative management of patients in order to accelerate early postoperative maternal rehabilitation.

Estimation of Central Aortic Pressure Waveforms by Combination of a Meta-Learning Neural Network and a Physics-Driven Method.

The accurate non-invasive detection and estimation of central aortic pressure waveforms (CAPW) are crucial for reliable treatments of cardiovascular system diseases. But the accuracy and practicality of current estimation methods need to be improved. Our study combines a meta-learning neural network and a physics-driven method to accurately estimate CAPW based on personalized physiological indicators.

Decoding Salience: A Functional Magnetic Resonance Imaging Investigation of Reward and Contextual Unexpectedness in Memory Encoding and Retrieval.

The present study investigated the neuromodulatory substrates of salience processing and its impact on memory encoding and behaviour, with a specific focus on two distinct types of salience: reward and contextual unexpectedness. 46 Participants performed a novel task paradigm modulating these two aspects independently and allowing for investigating their distinct and interactive effects on memory encoding while undergoing high-resolution fMRI. By using advanced image processing techniques tailored to examine midbrain and brainstem nuclei with high precision, our study additionally aimed to elucidate differential activation patterns in subcortical nuclei in response to reward-associated and contextually unexpected stimuli, including distinct pathways involving in particular dopaminergic modulation.

Using the hospital frailty risk score to assess oesophago-gastric cancer patient outcomes: a retrospective cohort study.

Background: The inclusion of clinical frailty in the assessment of patients planned for major surgery has proven to be an independent predictor of outcome. Since approximately half of all patients in the UK diagnosed with oesophagogastric (OG) cancer are over 75 years of age, assessment of frailty may be important in selection for surgery.

Materials And Methods: This retrospective cohort study applied the Hospital Frailty Risk Score to data obtained from the NHS Secondary Uses Service electronic database for patients aged 75 years or older undergoing oesophagectomy and gastrectomy between April 2017 and March 2020.

Effect of psilocybin versus escitalopram on depression symptom severity in patients with moderate-to-severe major depressive disorder: observational 6-month follow-up of a phase 2, double-blind, randomised, controlled trial.

Background: Psilocybin therapy (PT) produces rapid and persistent antidepressant effects in major depressive disorder (MDD). However, the long-term effects of PT have never been compared with gold-standard treatments for MDD such as pharmacotherapy or psychotherapy alone or in combination.

Methods: This is a 6-month follow-up study of a phase 2, double-blind, randomised, controlled trial involving patients with moderate-to-severe MDD.

The overlooked role of microbiota-gut-brain communication in child psychiatry: a call for integration in early intervention strategies.

Emerging research has highlighted the significant role of microbiota-gut-brain communication in child psychiatric disorders, including autism spectrum disorder (ASD) and anxiety disorders. Despite this, mainstream psychiatric interventions for children continue to focus predominantly on neurological and psychological therapies, neglecting the critical influence of gut microbiota on brain development and behavior. This commentary underscores the need for greater integration of microbiota-targeted therapies, such as dietary interventions, prebiotics, and probiotics, into early psychiatric intervention strategies.

An overview of proactive monitoring and management of respiratory issues in ataxia-telangiectasia in a specialist and shared care pediatric clinic.

Ataxia-telangiectasia (A-T) is an ultrarare autosomal recessive disorder and occurs in all racial and ethnic backgrounds. Clinically, children and young people with A-T are affected by sinopulmonary infections, neurological deterioration with concomitant bulbar dysfunction, increased sensitivity to ionizing radiation, immunodeficiency, a decline in lung function, chronic liver disease, endocrine abnormalities, cutaneous and deep-organ granulomatosis, and early death. Pulmonary complications become more frequent in the second decade of life and are a leading cause of death in individuals with A-T.

Enhancing oncolytic virotherapy by extracellular vesicle mediated microRNA reprograming of the tumour microenvironment.

Background: There has been limited success of cancer immunotherapies in the treatment of ovarian cancer (OvCa) to date, largely due to the immunosuppressive tumour microenvironment (TME). Tumour-associated macrophages (TAMs) are a major component of both the primary tumour and malignant ascites, promoting tumour growth, angiogenesis, metastasis, chemotherapy resistance and immunosuppression. Differential microRNA (miRNA) profiles have been implicated in the plasticity of TAMs.

The response of health systems to the needs of migrants and refugees in the COVID-19 pandemic: a comparative case study between Mexico, Colombia and Peru.

Background: Protecting the health of migrants and refugees during the pandemic was a significant challenge in the Latin American region. We aimed to describe and contrast the response of the health systems of Mexico, Colombia and Perú to migrants' and refugees' health needs during the COVID-19 pandemic, and to situate the response in the context of the migration and health policies of each country.

Methods: We conducted case studies of the three countries.

Background: Negative perceptions of mental health professionals can deter individuals from seeking mental healthcare. Given the high burden of mental health globally, it is essential to understand attitudes towards mental health professionals. Social media platforms like Twitter/X provide valuable insights into the views of the general population.

The 2023 fatal dengue outbreak in Bangladesh highlights a paradigm shift of geographical distribution of cases.

In 2023, Bangladesh experienced its largest and deadliest outbreak of the Dengue virus (DENV), reporting the highest-ever recorded annual cases and deaths. Historically, most of the cases were recorded in the capital city, Dhaka. We aimed to characterize the geographical transmission of DENV in Bangladesh.

Depressive symptoms in older adults are associated with changes in stress-related markers, functional connectivity and brain volume.

Background: Subclinical depressive symptoms increase the risk of developing Alzheimer's disease (AD). The neurobiological mechanisms underlying this link may involve stress system dysfunction, notably related to the hippocampus which is particularly sensitive to AD. We aimed to investigate the links between blood stress markers and changes in brain regions involved in the stress response in older adults with or without subclinical depressive symptoms.

The effect of post-exercise heat exposure (passive heat acclimation) on endurance exercise performance: a systematic review and meta-analysis.

Background: "Active" heat acclimation (exercise-in-the-heat) can improve exercise performance but the efficacy of "passive" heat acclimation using post-exercise heat exposure is unclear. Therefore, we synthesised a systematic review and meta-analysis to answer whether post-exercise heat exposure improves exercise performance.

Methods: Five databases were searched to identify studies including: (i) healthy adults; (ii) an exercise training intervention with post-exercise heat exposure via sauna or hot water immersion (treatment group); (iii) a non-heat exposure control group completing the same training; and (iv) outcomes measuring exercise performance in the heat (primary outcome), or performance in thermoneutral conditions, V̇Omax, lactate threshold, economy, heart rate, RPE, core temperature, sweat rate, and thermal sensations.

Gut microbiome signatures of vegan, vegetarian and omnivore diets and associated health outcomes across 21,561 individuals.

As plant-based diets gain traction, interest in their impacts on the gut microbiome is growing. However, little is known about diet-pattern-specific metagenomic profiles across populations. Here we considered 21,561 individuals spanning 5 independent, multinational, human cohorts to map how differences in diet pattern (omnivore, vegetarian and vegan) are reflected in gut microbiomes.

Autogene cevumeran with or without atezolizumab in advanced solid tumors: a phase 1 trial.

Effective targeting of somatic cancer mutations to enhance the efficacy of cancer immunotherapy requires an individualized approach. Autogene cevumeran is a uridine messenger RNA lipoplex-based individualized neoantigen-specific immunotherapy designed from tumor-specific somatic mutation data obtained from tumor tissue of each individual patient to stimulate T cell responses against up to 20 neoantigens. This ongoing phase 1 study evaluated autogene cevumeran as monotherapy (n = 30) and in combination with atezolizumab (n = 183) in pretreated patients with advanced solid tumors.
