Background: Neoplastic lesions affecting peripheral nerves are rare in the general population and, most often, are benign peripheral nerve sheath tumors. However, a minority of lesions represent high-grade malignancies associated with a poor prognosis, such as malignant peripheral nerve sheath tumors (MPNSTs). Very rarely, these tumors represent peripheral non-nerve sheath tumors (PNNSTs), such as hematological neoplasms that impair nerve function.

Background: Liver uptake in [Ga]Ga-PSMA-11 PET is used as an internal reference in addition to clinical parameters to select patients for [Lu]Lu-PSMA-617 radioligand therapy (RLT). Due to increased demand, [Ga]Ga-PSMA-11 was replaced by [F]F-PSMA-1007, a more lipophilic tracer with different biodistribution and splenic uptake was suggested as a new internal reference. We compared the intra-patient tracer distribution between [Ga]Ga-PSMA-11 and [F]F-PSMA-1007.

HIV-1 Vpr Functions in Primary CD4 T Cells.

Viruses

March 2024

Institute for Medical Virology and Epidemiology of Viral Diseases, University Hospital Tuebingen, 72076 Tuebingen, Germany.

HIV-1 encodes four accesory proteins in addition to its structural and regulatory genes. Uniquely amongst them, Vpr is abundantly present within virions, meaning it is poised to exert various biological effects on the host cell upon delivery. In this way, Vpr contributes towards the establishment of a successful infection, as evidenced by the extent to which HIV-1 depends on this factor to achieve full pathogenicity in vivo.

Objectives: Antinuclear antibodies (ANA) of the IgE-type have been described in several connective tissue disorders (CTD) but not yet in systemic sclerosis (SSc). Aim of the study was, therefore, to establish an ELISA for the demonstration of IgE-autoantibodies to topoisomerase-I (topo-I) and the centromeric proteins A and B (CENP-A/B), to assess their prevalence and reactivity in SSc and to analyse their clinical relevance.

Methods: One hundred fifty-one patients with SSc and 88 with CREST-syndrome, 291 patients with other CTD, and 23 patients with fibromyalgia syndrome (FM) as a control collective were included into the study.

Patients with systemic sclerosis and low CD4 numbers after autologous stem cell transplantation have a favorable outcome.

Arthritis Res Ther

March 2024

Department of Hematology, Oncology, Clinical Immunology and Rheumatology, University Hospital Tuebingen, Otfried-Mueller-Strasse 10, 72076, Tuebingen, Germany.

Background: Treatment with high-dose chemotherapy followed by autologous hematopoietic stem cell transplantation (aHSCT) is an intensive treatment option for patients with severe forms of systemic sclerosis (SSc). Even though associated with a high treatment related mortality, the results in this high-risk population are generally favourable. The knowledge on the potential mechanism of action of this therapy and how it can improve patients with SSc is crucial to better select the right patients for aHSCT.

Article Synopsis
  • The study investigates the effectiveness of vessel-encoded arterial spin labeling (VE-ASL) in visualizing blood flow from individual arteries in the brain, compared to the traditional method of digital subtraction angiography (DSA).
  • A review of data from 20 VE-ASL and DSA sets involving 17 patients with Moyamoya angiopathy showed a high level of agreement between the two imaging techniques.
  • The findings suggest that VE-ASL is a reliable non-invasive alternative for assessing cerebral blood flow contributions, making it particularly useful both before and after revascularization surgery.

Background: Effective treatment options are limited for patients with advanced melanoma who have progressed on immune checkpoint inhibitors (ICI) and targeted therapies (TT). Preclinical models support the combination of ICI with TT; however, clinical trials evaluating the efficacy of triplet combinations in first-line setting showed limited advantage compared to TT only.

Methods: We conducted a retrospective, multicenter study, that included patients with advanced melanoma who were treated with BRAF/MEK inhibitors in combination with an anti-PD-(L)1 antibody (triplet therapy) after failure of at least one anti-PD-(L)1-based therapy and one TT in seven major melanoma centers between February 2016 and July 2022.

Stroke frequency, associated factors, and clinical features in primary systemic vasculitis: a multicentric observational study.

J Neurol

June 2024

Nuffield Department of Orthopaedics, Rheumatology and Musculoskeletal Sciences, Nuffield Orthopaedic Centre, Botnar Research Centre, University of Oxford, Oxford, UK.

Article Synopsis
  • The study investigates how common cerebrovascular events (CVEs) are in patients diagnosed with primary systemic vasculitis (PSV) and identifies factors linked to these events.
  • It involved analyzing data from over 4,800 adults with PSV, revealing that 3.50% experienced CVEs, primarily strokes and transient ischemic attacks (TIAs), with the highest rates in certain types of vasculitis like Behçet's disease.
  • The findings highlight that CVEs are prevalent among PSV patients, vary significantly depending on the type of vasculitis, and recur at a high rate, indicating that traditional vascular risk factors do not account for these events.

As our knowledge about the biology of Alzheimer's disease (AD) expands and we recognize the significance of early intervention for effective treatment, there is a shift in focus toward detecting the disease at an early stage. AD is characterized by the accumulation of misfolded amyloid-β (Aβ) and phosphorylated tau proteins in the brain, leading to the formation of senile plaques and neurofibrillary tangles. While a definitive diagnosis of AD can only be confirmed through autopsy by examining these pathological features, there are now reliable methods available for diagnosing the disease in living individuals.
