Pancreatic ductal adenocarcinoma (PDAC) displays a high degree of spatial subtype heterogeneity and co-existence, linked to a diverse microenvironment and worse clinical outcome. However, the underlying mechanisms remain unclear. Here, by combining preclinical models, multi-center clinical, transcriptomic, proteomic, and patient bioimaging data, we identify an interplay between neoplastic intrinsic AP1 transcription factor dichotomy and extrinsic macrophages driving subtype co-existence and an immunosuppressive microenvironment.

Introduction: For patients with psychosis, early, intensive therapeutic intervention is thought to improve long-term outcomes. Furthermore, patients with a first-episode psychosis (FEP) who experience a good early response to antipsychotic medication show a clinical and functional benefit over the longer term if they continue low-dose antipsychotic treatment. Lurasidone is an atypical antipsychotic agent which is approved in Europe for the treatment of schizophrenia in adults and adolescents (13-17 years).

In early-stage Alzheimer's disease (AD) amyloid-β (Aβ) deposition can induce neuronal hyperactivity, thereby potentially triggering activity-dependent neuronal secretion of phosphorylated tau (p-tau), ensuing tau aggregation and spread. Therefore, cortical excitability is a candidate biomarker for early AD detection. Moreover, lowering neuronal excitability could potentially complement strategies to reduce Aβ and tau buildup.

Background: Due to its significant unilateral predominance, tennis can provoke functional and morphological asymmetries that develop over time and may result in undesired morphological alterations.
Hypothesis/purpose: The goals of this study were a) to assess glenohumeral range of motion and muscular stiffness in young tennis players with and without a history of shoulder pain and b) to examine interlimb asymmetries in these variables in both groups. It was hypothesized that players with a history of shoulder pain would show a reduced glenohumeral internal rotation (IR) and total arc of motion (TAM) and increased stiffness in internal rotator muscles compared to those without shoulder pain.

Air pollution is a major cardiovascular risk factor leading to higher rates of heart failure and myocardial infarction (MI), but its effects on functional recovery after an MI remain unknown. Cardiac rehabilitation is a cornerstone of post-MI care and leads to better performance and quality of life, but its benefits may be hampered in heavily polluted environments. To assess the effect of different pollutants on post-MI rehabilitation, we included 137 post-MI patients from 7 Spanish hospitals that were enrolled in a cardiac rehabilitation program who underwent two cardiopulmonary exercise tests (CPET) within a 12-week period.

Physical exercise has been shown to induce epigenetic modifications with various health implications, directly affect DNA methylation (DNAm), as well as reverse the epigenetic age. Hence, we aimed to identify differential methylation changes and assess the epigenetic age in the saliva of 7-9-year-old school children following a 3-month integrated neuromuscular training (INT), as well as to explore if any of the methylation changes are in core genes. Core genes are defined as genes of high relevance and essential importance within the human genome.

Static cold storage remains the traditional standard for liver graft preservation prior to transplantation in both clinical and experimental settings. The use of polyethylene glycol 35 solutions, such as Institut Georges Lopez-2 (IGL2) preservation solution, for protecting against mitochondrial damage during cold static preservation necessitates combination with hypothermic oxygenated perfusion to enhance liver graft performance. This study presents a preliminary comparative evaluation of "danger signals" indicating hepatocellular injury (transaminases, lactate content), mitochondrial damage (glutamate dehydrogenase release), and cytokine release in liver perfusates from suboptimal grafts (fatty livers) subjected to 24-hour cold storage.

Solid magnetic liposomes (ML, nanocomposites comprising lipid bilayers that incorporate magnetic nanoparticles) may be used in wastewater remediation: the lipid bilayer creates an environment where organic pollutants preferentially partition instead of water and the manipulation of ML with an external magnet enables an easy recovery from water. This study aimed to assess the system's potential for water remediation, focusing on ML ability to remove common pollutants in industrial wastewater. Specifically, alkylphenol ethoxylates (APEO) were used as the archetype for organic pollutants.

Nowadays, consent to use donor bodies for medical education and research is obtained from the body donors and their families before the donation. Recently, the International Federation of Associations of Anatomists (IFAA) published guidelines that could restrict the appearance of cadaveric images in commercial anatomical resources such as textbooks and other educational products. These guidelines state that the donor must expressly consent to using such images for this purpose.

Maintaining homeostasis is essential for continued health, and the progressive decay of homeostatic processes is a hallmark of ageing. Daily environmental rhythms threaten homeostasis, and circadian clocks have evolved to execute physiological processes in a manner that anticipates, and thus mitigates, their effects on the organism. Clocks are active in almost all cell types; their rhythmicity and functional output are determined by a combination of tissue-intrinsic and systemic inputs.

Intestinal epithelial cells are segregated into proliferative crypts and differentiated regions. This organization relies on specific signals, including Wnt3a, which regulates cell proliferation within crypts, and Eph/Ephrin, which dictates cell positioning along the crypt-villus axis. However, studying how the spatial distributions of these signals influences crypt-villus organization is challenging both in vitro and in vivo.
