Oxygen radicals--biochemical basis for their efficacy.

In the last decade it has become quite clear that oxygen free radicals are involved in a vast amount of diseases such as cataract, atherosclerosis, rheumatism, arthritis, Parkinson's disease, reperfusion injuries and many others. The induction of defence systems against certain stresses (heat shock, inflammation) is also mediated by activated oxygen species. Oxygen-activation and -desactivation has to be regulated and well attenuated in aerobic cells and tissues.

Excessive release of reactive oxygen metabolites (ROM) from lung inflammatory cells has been claimed to be of major pathogenetic significance in diffuse fibrosing alveolitis. In the present study, the content of oxidized methionine residues [Met(O)] as a percentage of total methionine (Met) in BAL-derived proteins was used to assess the biologic effect of ROM. In addition, procollagen-III-peptide was measured in BAL fluid as a marker of fibroblast activation.

In this investigation we analysed the maintenance methylation activity of the mammalian cell DNA methyltransferase by microinjection of hemimethylated HSV-tk DNA into thymidine kinase-negative rat 2 cells. We found that the hemimethylated DNA was efficiently converted into symmetrical methylated molecules before DNA replication. Furthermore, integration of the trans-DNA into the host genome is an early event after gene transfer.

Metabolic myocardial preservation by means of preischemic insulin administration (glucose-potassium-insulin, GPI; acute parenteral alimentation, APA) with the aim of a preischemic myocardial glycogen enrichment was performed in 20 consecutive CABG patients (12 in the APA group, 8 in the control group). Before and after 30 min of an infusion (APA or 0.9% NaCl solution), blood levels of potassium, glucose, NEFA (non-esterified fatty acids) and lactate were determined from arterial (a), central venous (cv) and coronary sinus (cs) blood.

A functional analysis of the Arthrobacter oxidans 6-hydroxy-D-nicotine oxidase (6-HDNO) gene promoter (-35 region TTGACA and -10 region TATCAAT) and the UUG translation start codon was performed using site-directed mutagenesis. Deletion of the C residue from the -10 promoter region or mutations introduced upstream of the -10 region resulted in an increased 6-HDNO expression in Escherichia coli cells in vivo and in both E. coli and A.

The uncommon case of a 19 year old man is reported, whose corpse has been found lying in the middle of a road nearby an overpass. The investigators suspected a capital crime because of some strange findings--such as crossed legs, a stub of a cigarette between the fingers of the right hand and suspect head-injuries. The further forensic and criminalistic investigations proved, that the young man tumbled down the bridge head over heals and died of brain-injuries.

A laboratory study was conducted to examine the role of explorative decision making for interpersonal conflict resolution. Participants were 64 male students, who had to negotiate for an outcome distribution against bogus enactment of two confederates. Questionnaire and observational data, which were taken before, during and after the negotiation revealed strong support for hypothesized effects of coping with situational antagonism.

It is quite common that the different procedures for isolation of nuclei make use of the purifying effect of sedimentation through concentrated sucrose. For this reason we investigated by light and electron microscopy the changes of chromatin structure in dependence on the amount and sort of ions present in the solution used for isolation and resuspension of nuclei. A decreasing concentration of monovalent cations (Na+, K+) leads to a swelling of the whole nuclei in consequence of the decondensation of the chromatin fibres.

We present 2 cases of subcutaneous fat necrosis associated with pancreatic disease. In 1 of these cases the correct diagnosis led to the detection of a clinically asymptomatic chronic pancreatitis. The pathogenesis of this disease, not yet fully explained, and the differential diagnosis of other forms of panniculitis are reviewed.

The main clinical and bioelectrical features of the benign partial epilepsies and related conditions are described. Based on highly selected groups, the definition of these suggested syndromes disregards the considerable overlap between borderline and intermediate cases. The lipopeptide Pam3Cys-Ser, a synthetic analogue of the N-terminal part of bacterial lipoprotein, constitutes a potent activator for B lymphocytes, monocytes/macrophages and several lymphoid cell lines. We applied the novel method of electron energy loss spectroscopy (EELS) to determine, after stimulation, the distribution of the activator within the cell compartments of the lipopeptide sensitive cell line BCL1. Our results show that the lipopeptide, 20 min after the addition to the cell culture, was found at different locations within the cell: A major amount of the mitogen was found in the plasma membrane.

The anticonvulsant properties of ketamine and 2-APV were compared on 3 types of convulsant activity in hippocampal area CA1: the 'picrotoxin-epilepsy,' the 'low magnesium epilepsy' and the 'low calcium epilepsy.' In particular the spontaneous activity, the synaptically evoked responses and the changes in [Ca2+]0 were examined, since in many cases of epilepsy, Ca2+ uptake into cells is enhanced. In normal medium, ketamine and 2-APV have nearly no effect on stimulus evoked decreases in [Ca2+]0, although they clearly depress NMDA-induced ionic changes.

A retrospective study covering the period from 1959 through 1983 has been conducted by the authors to analyse resective approaches to lung cancer. Regulation of IgE antibody response is T-cell dependent. It has been suggested that IL-4 (BSF-1) induces the expression of Fc epsilon RII on B-lymphocytes. Soluble molecules of the FC epsilon RII with various molecular weights represent IgE-binding factors which according to the current opinion either enhance or suppress the IgE-antibody response.

Out of 921 endoscopic papillotomies (EPT) 110 (12%) were performed in patients with benign stenosis of the papilla of Vater. Among these patients were 96 after cholecystectomy and 14 with gallbladder in situ. The diagnostic aspects of the stenosis of papilla and the complications of EPT were investigated on the basis of these patients.

5'-Nucleotidase was isolated from the electric organ of the electric ray Torpedo marmorata after solubilization in Triton X-100 and deoxycholate by affinity chromatography on concanavalin A-Sepharose and AMP-Sepharose. The purified enzyme has a Km for AMP of 38 microM, with a maximal velocity of 31 units/mg of protein. Of the purine and pyrimidine mononucleotides, AMP is hydrolysed most effectively.

[Obstetrical urogenital fistulas then and now].

An analysis was made of the aetiology and localisation of 83 obstetric urogenital fistulae, in 15-year intervals, which had been treated at the Gynecological Hospital of Charité over the past 45 years, with the view to finding out, if obstetrics continues to play a causative role in fistulation. With overall figures having stayed nearly constant (31, 25 and 27 fistulae), an increase has been primarily recordable from causes relating to caesarean section and from fistula localisations in the vesico-uterine region. Obstetric causes accounted for 10 per cent and gynaecological causes for 90 per cent of 808 urogenital fistulae throughout the entire period under review.
