December 2023

Maladies du Sang, CHU d'Angers, Angers, France; Federation Hospitalo-Universitaire Grand-Ouest Acute Leukemia, FHU-GOAL; Universite d'Angers, Inserm UMR 1307, CNRS UMR 6075, Nantes Universite, CRCI2NA, F-49000 Angers.

Whereas the prognosis of adult patients with Philadelphia-negative acute lymphoblastic leukemia (ALL) has greatly improved since the advent of pediatric-inspired regimens, the impact of initial central nervous system (CNS) involvement has not been formerly re-evaluated. We report here the outcome of patients with initial CNS involvement included in the pediatric-inspired prospective randomized GRAALL-2005 study. Between 2006 and 2014, 784 adult patients (aged 18-59 years) with newly diagnosed Philadelphia-negative ALL were included, of whom 55 (7%) had CNS involvement.

May 2023

Instituto de Medicina Molecular João Lobo Antunes, Faculdade de Medicina, Universidade de Lisboa, Lisbon, Portugal.

Article Synopsis
  • Cerebral venous sinus thrombosis due to vaccine-induced immune thrombotic thrombocytopenia (CVST-VITT) can lead to severe bleeding and high mortality, particularly among patients requiring decompressive surgery post-COVID vaccination.
  • A study reviewed 128 CVST-VITT patients, finding that 27% underwent surgery, with a 65% in-hospital mortality rate in this group compared to 29% in non-surgical patients, highlighting factors like preoperative coma and pupillary reflex absence as key mortality predictors.
  • Despite the high mortality, around 80% of the surgical survivors were functionally independent at a 6-month follow-up, indicating some potential for recovery even after critical interventions.

Obesity is a complex disease highly related to diet and lifestyle and is associated with low amount of thermogenic adipocytes. Therapeutics that regulate brown adipocyte recruitment and activity represent interesting strategies to fight overweight and associated comorbidities. Recent studies suggest a role for several fatty acids and their metabolites, called lipokines, in the control of thermogenesis.

Platelet P2Y receptor exhibits constitutive G protein signaling and β-arrestin 2 recruitment.

BMC Biol

February 2023

INSERM, UMR 1297, Institut des Maladies Métaboliques et Cardiovasculaires, Université de Toulouse, F-31432, Toulouse, France.

Background: Purinergic P2Y and P2Y receptors (P2Y-R and P2Y-R) are G protein-coupled receptors (GPCR) activated by adenosine diphosphate (ADP) to mediate platelet activation, thereby playing a pivotal role in hemostasis and thrombosis. While P2Y-R is the major target of antiplatelet drugs, no P2Y-R antagonist has yet been developed for clinical use. However, accumulating data suggest that P2Y-R inhibition would ensure efficient platelet inhibition with minimal effects on bleeding.
