Institute of Pharmacology and Toxicology, University of Würzburg, Versbacher Str. 9, Würzburg 97078, Germany; Leibniz-Institut für Analytische Wissenschaften - ISAS - e.V., Bunsen-Kirchhoff-Str. 11, Dortmund 44139, Germany; Comprehensive Heart Failure Center, University Hospital of Würzburg, Am Schwarzenberg 15, Würzburg 97078, Germany. Electronic address:

Article Synopsis
  • Impaired calcium (Ca) handling in heart cells is a key feature of heart failure (HF), leading to issues like weakened heart contractions and irregular heartbeats.
  • The study used transgenic mice with a mutation affecting a calcium regulator (phospholamban) to understand how defects in calcium cycling contribute to HF, noting that these mice experience severe and fast-progressing heart failure.
  • Early treatment aimed at correcting calcium cycling using Raf kinase inhibitor protein (RKIP) was found to delay heart cell damage and improve overall health of the mice, indicating that addressing Ca dynamics early on could be crucial for preventing further complications in heart failure.

Predicting benefit from PARP inhibitors using deep learning on H&E-stained ovarian cancer slides.

Eur J Cancer

December 2024

Division of Digital Prevention, Diagnostics and Therapy Guidance, German Cancer Research Center (DKFZ), Heidelberg, Germany. Electronic address:

Article Synopsis
  • Ovarian cancer patients with Homologous Recombination Deficiency (HRD) may benefit from PARP inhibitor therapy after platinum chemotherapy, and predicting this benefit through whole slide images (WSIs) could provide a quicker and less costly alternative to molecular tests.
  • A Deep Learning (DL) model was trained on H&E stained WSIs using a specific HRD ground truth, and it was tested on a separate cohort to see how well it predicted HRD status and the benefit of olaparib treatment.
  • Although the model showed potential, with a significant improvement in progression-free survival (PFS) for HRD positive patients treated with PARP inhibitors, its overall prediction accuracy was lower than desired, indicating that further

Department of Psychiatry, University of Cambridge, Cambridge, UK; Department of Systems Neuroscience, University Medical Center Hamburg-Eppendorf, Hamburg, Germany; Department of Addictive Behaviour and Addiction Medicine, Central Institute of Mental Health, University of Heidelberg, Mannheim, Germany. Electronic address:

Background: A preference for sooner-smaller over later-larger rewards, known as delay discounting, is a candidate transdiagnostic marker of waiting impulsivity and a research domain criterion. While abnormal discounting rates have been associated with many psychiatric diagnoses and abnormal brain structure, the underlying neuropsychological processes remain largely unknown. Here, we deconstruct delay discounting into choice and rate processes by testing different computational models and investigate their associations with white matter tracts.
