The therapeutic use of A adenosine receptor (AR) antagonists for the treatment of neurodegenerative disorders, such as Parkinson and Alzheimer diseases, is a very promising approach. Moreover, the potential therapeutic role of A AR antagonists to avoid both immunoescaping of tumor cells and tumor development is well documented. Herein, we report on the synthesis and biological evaluation of a new set of piperazine- and piperidine- containing 7-amino-2-(furan-2-yl)thiazolo[5,4-]pyrimidine derivatives designed as human A AR antagonists/inverse agonists.

Phylogenomic proof of Recurrent Demipolyploidization and Evolutionary Stalling of the "Triploid Bridge" in (Poaceae).

Polyploidization is a frequent phenomenon in plants, which entails the increase from one generation to the next by multiples of the haploid number of chromosomes. While tetraploidization is arguably the most common and stable outcome of polyploidization, over evolutionary time triploids often constitute only a transient phase, or a "triploid bridge", between diploid and tetraploid levels. In this study, we reconstructed in a robust phylogenomic and statistical framework the evolutionary history of polyploidization in , a small genus from the Poaceae family with promising biomass, bioenergy and phytoremediation species.

[An update on pulmonary embolism-related mortality in Italy (2003-2015)].

Background: Data regarding pulmonary embolism (PE)-related mortality in Italy are scarce. We assessed PE-related mortality and its time trend in Italy by using the World Health Organization (WHO) Mortality Database.

Methods: The vital registration data of Italy from the WHO Mortality Database were analyzed for the period between 2003 and 2015, and compared with time trends in Southern Europe.

Perioperative Management of Elderly patients (PriME): recommendations from an Italian intersociety consensus.

Background: Surgical outcomes in geriatric patients may be complicated by factors such as multiple comorbidities, low functional performance, frailty, reduced homeostatic capacity, and cognitive impairment. An integrated multidisciplinary approach to management is, therefore, essential in this population, but at present, the use of such an approach is uncommon. The Perioperative Management of Elderly patients (PriME) project has been established to address this issue.

Treatment with ACE inhibitors or ARBs and risk of severe/lethal COVID-19: a meta-analysis.

Objective: It has been hypothesised that the use of ACE inhibitors and angiotensin receptor blockers (ARBs) might either increase or reduce the risk of severe or lethal COVID-19. The findings from the available observational studies varied, and summary estimates are urgently needed to elucidate whether these drugs should be suspended during the pandemic, or patients and physicians should be definitely reassured. This meta-analysis of adjusted observational data aimed to summarise the existing evidence on the association between these medications and severe/lethal COVID-19.

Vaccines in older age: moving from current practice to optimal coverage-a multidisciplinary consensus conference.

Vaccines are a key strategy to promote healthy aging, but vaccine coverage remains below target levels in at-risk adults and older individuals. We present here the results of a multidisciplinary consensus conference convened to perform a multidimensional assessment of vaccination in geriatric medicine, with a view to developing a well-defined strategy for the promotion of vaccines in older people. We discuss recommended vaccines in older individuals, and describe the wide regional heterogeneity between regions in the Italian context.

Paroxysmal eruptions tracked by variations of helium isotopes: inferences from Piton de la Fournaise (La Réunion island).

Helium (He) with its isotopes (He, He) is a key tracer enabling the Earth's mantle and dynamics to be characterized. Enrichment in primordial helium (He) has been detected in volcanic gases of numerous magmatic systems in different geodynamic settings. Despite past use to monitor volcano-tectonic unrest, temporal He/He variability in volcanic emissions is still poorly constrained.

Experimental and archaeological data for the identification of projectile impact marks on small-sized mammals.

The role of small game in prehistoric hunter-gatherer economy is a highly debated topic. Despite the general assumption that this practice was uneconomic, several studies have underlined the relevance of the circumstance of capture - in terms of hunting strategies and technology - in the evaluation of the actual role of small mammals in human foraging efficiency. Since very few studies have focused on the recognition of bone hunting lesions, in a previous work we explored the potential of 3D microscopy in distinguishing projectile impact marks from other taphonomic marks, developing a widely-applicable diagnostic framework based on experimental data and focused on Late Epigravettian projectiles.

Differential Diagnosis of Inflammatory Arthropathies by Musculoskeletal Ultrasonography: A Systematic Literature Review.

Differential diagnosis in early arthritis is challenging, especially early after symptom onset. Several studies applied musculoskeletal ultrasound in this setting, however, its role in helping diagnosis has yet to be clearly defined. The purpose of this work is to systematically assess the diagnostic applications of ultrasonography in early arthritis in order to summarize the available evidence and highlight possible gaps in knowledge.

Correlation between Mechanical Properties and Processing Conditions in Rubber-Toughened Wood Polymer Composites.

The use of wood fibers is a deeply investigated topic in current scientific research and one of their most common applications is as filler for thermoplastic polymers. The resulting material is a biocomposite, known as a Wood Polymer Composite (WPC). For increasing the sustainability and reducing the cost, it is convenient to increase the wood fiber content as much as possible, so that the polymeric fraction within the composite is thereby reduced.

Design, synthesis, in vitro and in vivo biological evaluation of 2-amino-3-aroylbenzo[b]furan derivatives as highly potent tubulin polymerization inhibitors.

A new class of inhibitors of tubulin polymerization based on the 2-amino-3-(3',4',5'-trimethoxybenzoyl)benzo[b]furan molecular scaffold was synthesized and evaluated for in vivo and in vitro biological activity. These derivatives were synthesized with different electron-releasing or electron-withdrawing substituents at one of the C-4 through C-7 positions. Methoxy substitution and location on the benzene part of the benzo[b]furan ring played an important role in affecting antiproliferative activity, with the greatest activity occurring with the methoxy group at the C-6 position, the least with the substituent at C-4.

Molecular Basis of CLC Antiporter Inhibition by Fluoride.

CLC channels and transporters conduct or transport various kinds of anions, with the exception of fluoride, which acts as an effective inhibitor. Here, we performed sub-nanosecond DFT-based QM/MM simulations of the anion/proton exchanger ClC-ec1 and observed that fluoride binds incoming protons within the selectivity filter, with excess protons shared with the gating glutamate E148. Depending on E148 conformation, the competition for the proton can involve either a direct F/E148 interaction or the modulation of water molecules bridging the two anions.

High levels of LDL cholesterol (LDL-C) represent a causal factor for cardiovascular diseases on an atherosclerotic basis, with a direct correlation between these and mortality or cardiovascular events, such that the reduction of both is associated proportionally and linearly with the reduction of LDL-C.Statins and ezetimibe are used for LDL-C lowering but may not be sufficient to achieve the targets defined by the ESC/EAS guidelines, which recommend use of PCSK9 inhibitors for further LDL-C reduction in patients not at goal.This project submitted 86 clinical scenarios to a group of experts, cardiologists, internists and lipidologists, collecting their opinion on the appropriateness of different behaviors and decisions.

Methods- Retrospective analysis of patients with primary spontaneous ICH admitted at 3 academic hospitals in Italy. The study population was divided into a development and a replication cohort.

Early Levallois core technology between Marine Isotope Stage 12 and 9 in Western Europe.

Early Levallois core technology is usually dated in Europe to the end of Marine Isotope Stage (MIS) 9 and particularly from the beginning of MIS 8 to MIS 6. This technology is considered as one of the markers of the transition from lower to Middle Paleolithic or from Mode 2 to Mode 3. Recent discoveries show that some lithic innovations actually appeared earlier in western Europe, from MIS 12 to MIS 9, contemporaneous with changes in subsistence strategies and the first appearance of early Neanderthal anatomical features.

A Pan-European high resolution storm surge hindcast.

This contribution presents the high-resolution Pan-European storm surge (SSL) dataset, ANYEU-SSL, produced with the SCHISM circulation model. The dataset covers 40 years (1979-2018) of SSL data along the European coastline with 3-hour temporal resolution and has been extensively validated for the period spanning from 1979 to 2016, considering the whole time series, as well as for the extreme SSL values. Validation against tidal gauge data shows an average RMSE of 0.

Identifying fetal yawns based on temporal dynamics of mouth openings: A preterm neonate model using support vector machines (SVMs).

Fetal yawning is of interest because of its clinical, developmental and theoretical implications. However, the methodological challenges of identifying yawns from ultrasonographic scans have not been systematically addressed. We report two studies that examined the temporal dynamics of yawning in preterm neonates comparable in developmental level to fetuses observed in ultrasound studies (about 31 weeks PMA).

Risk factors of damage in early diagnosed systemic lupus erythematosus: results of the Italian multicentre Early Lupus Project inception cohort.

Objective: To investigate risk factors for damage development in a prospective inception cohort of early diagnosed SLE patients.

Methods: The Early Lupus Project recruited an inception cohort of patients within 12 months of SLE classification (1997 ACR criteria). At enrolment and every 6 months thereafter, the SLICC/ACR Damage Index was recorded.

Imaging Early Postoperative Complications of Cardiothoracic Surgery.

Imaging plays a central role in the evaluation of patients following cardiothoracic surgery, both for monitoring in the early postoperative period and for assessing for suspected complications. Patients with postsurgical complications can develop a range of signs and symptoms, from hypotension and tachycardia, as the result of severe bleeding, to fever and leukocytosis because of infection. The radiologist is an important member of the care team in the postoperative period, helping identify and manage complications of cardiothoracic surgery.

Wetting and recovery of nano-patterned surfaces beyond the classical picture.

Hydrophobic (nano)textured surfaces, also known as superhydrophobic surfaces, have a wide range of technological applications, including in the self-cleaning, anti-moisture, anti-icing, anti-fogging and friction/drag reduction fields, and many more. The accidental complete wetting of surface textures, which destroys superhydrophobicity, and the opposite process of recovery are two crucial processes that can prevent or enable the technological applications mentioned before. Understanding these processes is key to designing surfaces with tailored wetting and recovery properties.

Amino-3,5-Dicyanopyridines Targeting the Adenosine Receptors Ranging from Pan Ligands to Combined A/A Partial Agonists.

The amino-3,5-dicyanopyridine derivatives belong to an intriguing series of adenosine receptor (AR) ligands that has been developed by both academic researchers and industry. Indeed, the studies carried out to date underline the versatility of the dicyanopyridine scaffold to obtain AR ligands with not only a wide range of affinities but also with diverse degrees of efficacies at the different ARs. These observations prompted us to investigate on the structure-activity relationships (SARs) of this series leading to important previously reported results.
