Acute myeloid leukemia (AML) is a complex cancer, yet advances in recent years from integrated genomics methods have helped improve diagnosis, treatment, and means of patient stratification. A recent example of a powerful, multimodal method is DOGMA-seq, which can measure chromatin accessibility, gene expression, and cell-surface protein levels from the same individual cell simultaneously. Syphilis, a chronic infection caused by Treponema pallidum, is experiencing a global resurgence, posing significant public health challenges. This study examined the escalating trends of syphilis in the United States, China, and some other countries highlighting the impact of the COVID-19 pandemic, changes in sexual behavior, coinfection with the other infectious diseases such as AIDs, and the role of public health funding. The analysis revealed a stark increase in syphilis cases, particularly among high-risk groups such as men who have sex with men (MSM).

Background: Low rates of adolescent and young adult (YA; aged 15-39 y) clinical trial enrollment (CTE), particularly among underserved groups, have resulted in a lack of standardized cancer treatments and follow-up guidelines for this group that may limit improvement in cancer treatments and survival outcomes for YAs.
Objective: To understand and address unique barriers to CTE, we conducted focus groups to learn about informational, financial, and psychosocial needs of YAs surrounding CTE and identify strategies to address these barriers.
Methods: We conducted 5 focus groups in 2023 among a diverse sample of YA patients from across the United States.

Background: As percutaneous therapeutic options expand, the optimal management of severe aortic stenosis (AS) and concomitant coronary artery disease (CAD) is being questioned between coronary artery bypass grafting and surgical aortic valve replacement (CABG+SAVR) versus percutaneous coronary intervention and transcatheter aortic valve replacement (PCI+TAVR). We sought to compare perioperative and longitudinal risk-adjusted outcomes between patients undergoing CABG+SAVR versus PCI+TAVR.
Methods: Using the United States Centers for Medicare and Medicaid Services inpatient claims database, we evaluated all patient age 65 and older with AS and CAD undergoing CABG+SAVR or PCI+TAVR (2018-2022).

Air-pollution monitoring is sparse across most of the United States, so geostatistical models are important for reconstructing concentrations of fine particulate air pollution (PM) for use in health studies. We present XGBoost-IDW Synthesis (XIS), a daily high-resolution PM machine-learning model covering the contiguous US from 2003 through 2023. XIS uses aerosol optical depth from satellites and a parsimonious set of additional predictors to make predictions at arbitrary points, capturing near-roadway gradients and allowing the estimation of address-level exposures.

Numerous unregulated organic compounds (UOCs) including pharmaceuticals, opioids, and personal care products (PCPs) end up in wastewater. UOC presence in biosolids (a wastewater treatment byproduct), which are applied to soil for different reasons raises environmental and health risk concerns. In this study, two multi-class extraction methods were developed and validated to target 111 UOCs from 8 different major families simultaneously in biosolids and biosolids-impacted soil.

Objective: We sought to develop consensus recommendations for measurement and analysis of data on contraceptive-induced menstrual changes (CIMCs) in contraceptive clinical trials. We built upon previous standardization efforts over the last 50 years and prioritized input from a variety of global experts and current regulatory authority guidance on patient-reported outcomes.
Study Design: We completed a formal consensus-building process with an interdisciplinary group of 57 experts from 30 organizations and 14 countries in five global regions who work across academia, nonprofit research organizations, the pharmaceutical industry, and funding agencies.

Epilepsy, a neurological disorder causing recurring seizures, is often studied in zebrafish by exposing animals to pentylenetetrazol (PTZ), which induces clonic- and tonic-like behaviors. While adult zebrafish seizure-like behaviors are well characterized, manual assessment remains challenging due to its time-consuming nature, potential for human error/bias, and the risk of overlooking subtle behaviors. Aiming to circumvent these issues, we developed a machine learning model for automating the analysis of subtle abnormal and seizure-like behaviors in PTZ-exposed adult zebrafish.

Novel kappa opioid receptor (KOR) agonists that preferentially activate G-protein signaling versus β-arrestin-2 recruitment are described. Starting from a literature-reported phenol-containing diphenethylamine KOR agonist, structure-activity relationship (SAR) studies revealed replacement of the phenol with various non-hydroxylated bicyclic heteroaromatics led to tertiary diarylethylamines which retained KOR agonist activity and improved metabolic stability in human liver microsomes. Further optimizations produced compound 39, a potent activator of G-protein signaling (GTPγS EC = 14 nM, 83 % E) that did not elicit a β-arrestin-2 recruitment functional response (E < 10 %).
