Central nervous system (CNS) resident memory CD8 T cells (T) that express IFN-γ contribute to neurodegenerative processes, including synapse loss, leading to memory impairment. Here, we show that CCR2 signaling in CD8 T that persist within the hippocampus after recovery from CNS infection with West Nile virus (WNV) significantly prevents the development of memory impairments. Using CCR2-deficient mice, we determined that CCR2 expression is not essential for CNS T cell recruitment or virologic control during acute WNV infection.

Measuring the biomechanical properties of cell-derived fibronectin fibrils.

Biomech Model Mechanobiol

December 2024

Department of Biomedical Engineering, Virginia Commonwealth University, 401 W. Main St., Richmond, VA, 23284, USA.

Embryonic development, wound healing, and organogenesis all require assembly of the extracellular matrix protein fibronectin (FN) into insoluble, viscoelastic fibrils. FN fibrils mediate cell migration, force generation, angiogenic sprouting, and collagen deposition. While the critical role of FN fibrils has long been appreciated, we still have an extremely poor understanding of their mechanical properties and how these mechanical properties facilitate cellular responses.

As the number of cerebral aneurysms treated with flow diverters continues to increase, it is important to understand what factors influence not only thrombus formation within the aneurysm cavity but also fibrin accumulation across the device and its associated disruption and blockage of the inflow stream. Both processes contribute to the eventual occlusion of the aneurysm or its continued patency and incomplete occlusion which may require future re-treatment. To investigate fibrin accumulation on flow diverters placed across the neck of cerebral aneurysms, a previously developed computational model that couples flow and fibrin dynamics is used in combination with experimental in vitro models of cerebral aneurysms treated with flow diverters.

December 2024

Department of Chemistry and Polymer Science, Stellenbosch University, Private Bag X1, Matieland 7602, South Africa.

Membrane proteins (MPs) are critical to cellular processes and serve as essential therapeutic targets. However, their isolation and characterization are often impeded by traditional detergent-based methods, which can compromise their native states, and retention of their native lipid environment. Amphiphilic polymers have emerged as effective alternatives, enabling the formation of nanoscale discs that preserve MPs' structural and functional integrity.

With reduced dimensionality and a high surface area-to-volume ratio, two-dimensional (2D) semiconductors exhibit intriguing electronic properties that are exceptionally sensitive to surrounding environments, including directly interfacing gate dielectrics. These influences are tightly correlated to their inherent behavior, making it critical to examine when extrinsic charge carriers are intentionally introduced to the channel for complementary functionality. This study explores the physical origin of the competitive transition between intrinsic and extrinsic charge carrier conduction in extrinsically -doped MoS, highlighting the central role of interactions of the channel with amorphous gate dielectrics.

Cyclic poly(2-methyl-2-oxazine) (-PMOZI) brush shells on Au nanoparticles (NPs) exhibit enhanced stealth properties toward serum and different cell lines compared to their linear PMOZI (-PMOZI) counterparts. While selectively recruiting immunoglobulins, -PMOZI shells reduce overall human serum (HS) protein binding and alter the processing of complement factor 3 (C3) compared to chemically identical linear shells. Polymer cyclization significantly decreases NP uptake by nonphagocytic cells and macrophages in both complement-deficient fetal bovine serum (FBS) and complement-expressing HS, indicating ineffective functional opsonization.

From The Center for Professionalism & Value in Health Care, American Board of Family Medicine, Washington, DC (RLP); Research & Policy, American Board of Family Medicine, Washington, DC; and Center for Professionalism & Value in Health Care Washington, DC (AWB).

Despite producing mountains of data in the daily course of care, the documentation labors of frontline clinicians currently return very little value to them or to the health system. The potential of these painstakingly collected data are enormous and clinical registries can extract the extraordinary capacity of these data and transform them into research-ready datasets while protecting the confidentiality of the patients and clinicians. Clinical registries represent transformative tools for primary care research, bringing together the dimensions of clinical practice, research, quality improvement, and policy impact from a large, nationally reflective, diverse sample of practices and patients.
